In ovarian cancer, therapy resistance mechanisms complicate cancer cell eradication. Targeting Musashi RNA-binding proteins (MSI) may increase therapeutic efficacy. Database analyses were performed to identify gene expression associations between MSI proteins and key therapy resistance and cancer stem cell (CSC) genes. Then, ovarian cancer cells were subjected to siRNA-based dual knockdown of MSI-1 and MSI-2. CSC and cell cycle gene expression was investigated using quantitative polymerase chain reaction (qPCR), western blots, and flow cytometry. Metabolic activity and chemoresistance were assessed by MTT assay. Clonogenic assays were used to quantify cell survival post-irradiation. Database analyses demonstrated positive associations between MSI proteins and putative CSC markers NOTCH, MYC, and ALDH4A1 and negative associations with NOTCH inhibitor NUMB. MSI-2 expression was negatively associated with the apoptosis regulator p21. MSI-1 and MSI-2 were positively correlated, informing subsequent dual knockdown experiments. After MSI silencing, CSC genes were downregulated, while cell cycle progression was reduced. Metabolic activity was decreased in some cancer cells. Both chemo- and radioresistance were reduced after dual knockdown, suggesting therapeutic potential. Dual knockdown of MSI proteins is a promising venue to impede tumor growth and sensitize ovarian cancer cells to irradiation and chemotherapy. 相似文献

为了揭示位于“V”形沟谷中的某座3级加筋土挡墙的变形规律,在现场实测数据的基础上,重点分析了高填方下3级加筋土高挡墙墙面板位移、墙后填土体沉降、高填方下路面沉降以及挡墙下涵洞底部沉降在施工期间和竣工后的变化情况。结果表明:①加筋土挡墙墙面板的水平位移、竖向位移以及墙后填土体的沉降主要发生在施工期间,且数值都比较大,竣工后1~2 a的时间内,其变形趋于稳定,但总的累积变形较大,说明加筋土挡墙能够适应较大变形的填方工程,这是其优势所在;②在“V”形沟谷中采用高填方下加筋土高挡墙的结构形式能满足路面沉降要求,高填方下的涵洞也是安全的;③由于挡墙筋带的特殊性,使得形成的加筋土挡墙具有锚定板挡墙和土钉墙的某些优点,既能约束墙内土体的变形,又能作为整体很好地同周围“V”形沟谷变形相协调,使得填方内应力重分布,路面沉降变形平缓过渡,未产生明显差异沉降,使用效果良好。这种挡墙结构在山区公路铁路建设中具有广阔的应用前景,值得推广。 相似文献

Using Time-Varying AR (TVAR) model and adaptive notch filter is a new method for the non-stationary jammer suppression in Direct Sequence Spread Spectrum (DSSS). The performance of TVAR model for Instantaneous Frequency (IF) estimation will be affected by some factors such as basis functions. Focusing on this problem, the optimal basis function of TVAR model for the IF estimation of the LFM signal is obtained in this paper. Besides the depth and width of notching, the phase properties of notch filter affect the Signal-to-Interference plus-Noise Ratio (SINR) of correlation output to the narrowband jammer suppression in DSSS, in response to the problem the closed solution of correlation output SINR improvement has been derived when a single frequency jammer passes through direct IIR notch filter, and its performance has been compared with those of five coefficient FIR filters. Later, a novel method for LFM jammer suppression based on Fourier basis TVAR model and direct IIR notch filter is proposed. The simulation results show the effectiveness of the proposed method. 相似文献

A novel method for estimating the instantaneous phasor of a fault current signal is proposed for high-speed distance protection
immune to a DC-offset. The method uses a modified notch filter in order to eliminate the fundamental frequency component from
the fault current signal. Since the output of the modified notch filter is the delayed DC-offset, delay compensation results
in the same waveform as the original DC-offset. Subtracting the obtained DC-offset from the fault current signal yields a
sinusoidal waveform, which becomes the real part of the instantaneous phasor. The imaginary part of the instantaneous phasor
is based on the first difference of the fault current signal. Since a DC-offset also appears in the first difference, the
DC-offset is removed from the first difference using the results of the delay compensation. The performance of the proposed
method was evaluated for a–phase to ground faults on a 345 kV 100 km overhead transmission line. The Electromagnetic Transient
Program was used to generate fault current signals for different fault locations and fault inception angles. The performance
evaluation showed that the proposed method can estimate the instantaneous phasor of a fault current signal with high speed
and high accuracy. The paper concludes by describing the hardware implementation of the proposed method on a prototype unit
