Transmission electron microscopy (TEM) is an important analysis technique to visualize (bio)macromolecules and their assemblies, including collagen fibers. Many protocols for TEM sample preparation of collagen involve one or more washing steps to remove excess salts from the dispersion that could hamper analysis when dried on a TEM grid. Such protocols are not standardized and washing times as well as washing solvents vary from procedure to procedure, with each research group typically having their own protocol. Here, we investigate the influence of washing with water, ethanol, but also methanol and 2-propanol, for both mineralized and unmineralized collagen samples via a protocol based on centrifugation. Washing with water maintains the hydrated collagen structure and the characteristic banding pattern can be clearly observed. Conversely, washing with ethanol results in dehydration of the fibrils, often leading to aggregation of the fibers and a less obvious banding pattern, already within 1 min of ethanol exposure. As we show, this process is fully reversible. Similar observations were made for methanol and propanol. Based on these results, a standardized washing protocol for collagenous samples is proposed. 相似文献

Limiting current density at different temperatures, backpressures, and balance gases can be used to separate molecular diffusion resistance, Knudsen diffusion resistance and local transport resistance of membrane electrode assembly (MEA). However, the measurement of limiting current density has no unified protocol. The diverse choices in the literature, either in the control of current or voltage or in the atmosphere like relative humidity and O2 concentrations, make it difficult to compare the results and identify the true bottleneck hindering the mass transport. In this work, the current-voltage curves obtained by current scanning/stepping and voltage scanning/stepping methods under dilute O2 of different concentrations and a wide range of relative humidity were measured and analyzed systematically. It is found that the voltage stepping method is superior to the other three ways of control for the reliable determination of the limiting current density. Aided with simultaneous electrochemical impedance spectroscopy measurement, the limiting current density can be determined with pinpoint accuracy. When the limiting current density is just used to qualitatively evaluate different MEA, the voltage scanning method can be used instead for its high time efficiency. The selection of the atmosphere also plays an important role in suppressing the distortion from excessive water and reducing the spurious contribution from proton conduction resistance. It is found that O2 concentrations at 0.5 vol% and relative humidity at 90% can give the best estimation of O2 transport resistance in membrane electrode assembly. 相似文献

The SARS-CoV-2 pandemic has created a great demand for a better understanding of the spread of viruses in indoor environments. A novel measurement system consisting of one portable aerosol-emitting mannequin (emitter) and a number of portable aerosol-absorbing mannequins (recipients) was developed that can measure the spread of aerosols and droplets that potentially contain infectious viruses. The emission of the virus from a human is simulated by using tracer particles solved in water. The recipients inhale the aerosols and droplets and quantify the level of solved tracer particles in their artificial lungs simultaneously over time. The mobile system can be arranged in a large variety of spreading scenarios in indoor environments and allows for quantification of the infection probability due to airborne virus spreading. This study shows the accuracy of the new measurement system and its ability to compare aerosol reduction measures such as regular ventilation or the use of a room air purifier. 相似文献

This paper presents a fast distance relay for series compensated transmission lines based on the R–L differential-equation algorithm using the theory of equal transfer process of transmission lines. The measuring distances based on the proposed algorithm can fast approach the actual value of fault distance when a fault occurs in front of the series capacitor. When a fault occurs behind of the series capacitor, the fault loop, including the series capacitor, does not match the R–L transmission line model, so the measuring distances fluctuate severely. Based on this, the relative position of the fault with respect to the series capacitor can be judged effectively according to the fluctuation range of the measuring distances, and the accurate fault location can be obtained fast. A variety of PSCAD/EMTDC simulation tests show that the new relay has fast operating speed and high accuracy when applied to the long series compensated transmission lines. 相似文献

An equiatomic CoCrFeNiMn high-entropy alloy was synthesized by mechanical alloying (MA) and spark plasma sintering (SPS). During MA, a solid solution with refined microstructure of 10 nm which consists of a FCC phase and a BCC phase was formed. After SPS consolidation, only one FCC phase can be detected in the HEA bulks. The as-sintered bulks exhibit high compressive strength of 1987 MPa. 在通讯设备爆炸式增长的时代,移动边缘计算作为5G通讯技术的核心技术之一,对其进行合理的资源分配显得尤为重要。移动边缘计算的思想是把云计算中心下沉到基站部署(边缘云),使云计算中心更加靠近用户,以快速解决计算资源分配问题。但是,相对于大型的云计算中心,边缘云的计算资源有限,传统的虚拟机分配方式不足以灵活应对边缘云的计算资源分配问题。为解决此问题,提出一种根据用户综合需求变化的动态计算资源和频谱分配算法(DRFAA),采用"分治"策略,并将资源模拟成"流体"资源进行分配,以寻求较大的吞吐量和较低的传输时延。实验仿真结果显示,动态计算资源和频谱分配算法可以有效地降低用户与边缘云之间的传输时延,也可以提高边缘云的吞吐量。 相似文献

This paper reviews recent studies, that not only includes both experiments and modeling components, but celebrates a close coupling between these techniques, in order to provide insights into the plasticity and failure of polycrystalline metals. Examples are provided of studies across multiple-scales, including, but not limited to, density functional theory combined with atom probe tomography, molecular dynamics combined with in situ transmission electron miscopy, discrete dislocation dynamics combined with nanopillars experiments, crystal plasticity combined with digital image correlation, and crystal plasticity combined with in situ high energy X-ray diffraction. The close synergy between in situ experiments and modeling provides new opportunities for model calibration, verification, and validation, by providing direct means of comparison, thus removing aspects of epistemic uncertainty in the approach. Further, data fusion between in situ experimental and model-based data, along with data driven approaches, provides a paradigm shift for determining the emergent behavior of deformation and failure, which is the foundation that underpins the mechanical behavior of polycrystalline materials. 相似文献

Y2Hf2O7 possesses low thermal conductivity and high melting point, which make it promising for a new anti-ablation material. For evaluating the thermal stability and the potential applications of Y2Hf2O7 on anti-ablation protection of C/C composites, Y2Hf2O7 ceramic powder was synthesized by solution combustion method and Y2Hf2O7 coating was prepared on the surface of SiC coated C/C composites using SAPS. Results shown that the coating exhibits good ablation resistance under the heat flux of 2.4?MW/m2 with the linear and mass ablation rates are 0.16?μm?s?1 and ?0.028?mg?s?1, respectively, after ablation for 40?s. With the prolonging of the ablation time, the increasing thermal stress causes the increase of cracks. Moreover, the chemical erosion from SiO2 and the physical volatilization of low temperature molten products aggravate failure of the Y2Hf2O7 coating. 相似文献
