Molecular technologies have been developed for the transformation of barley. These technologies complement current methods of barley breeding. In addition, they offer the potential of altering specific components in barley that affect malting quality and of introducing foreign genes, controlling desirable traits, into barley. Application of genetic engineering to improving malt quality factors such as cell wall degradation, protein modification, starch hydrolysis and flavour stability, is discussed. Limitations to the use of this technology for improving malt-related functional properties of barley components such as cell walls and starch granules are also evaluated. The use of activated carbon beds for the removal of natural humic and fulvic substances found in water supplies, has recently received considerable attention in water treatment operation (Lee et al., 1980; Le Cloirec et al., 1983). Moreover, the use of carbon adsorption for the reduction of haloform precursors (Anderson et al., 1981) and trihalomethanes produced by chlorination process, has contributed to a comprehensive investigation of adsorption characteristics of natural organic compounds (McCreary and Snoeyink, 1981). Many recent works showed the influence of adsorption system characteristics, such as pH, salt type, salt concentration and ionic heterogeneity in multicomponent adsorption systems, on the removal efficiency of humic and fulvic substances by activated carbon (McCreary and Snoeyink, 1980; Randtke and Jepsen, 1982; Weber et al., 1983). The purpose of this study is to examine the effect of a main component of domestic detergents, sodium triphosphate (STP), on the adsorptive capacities of powdered activated carbon (PAC) for commercially supplied humic acids, at different pH values in distilled water. Also, the effect of STP concentration and pH on the adsorption affinity of the PAC for humic acids, is discussed in relation with electrokinetic properties of carbon particles (zeta potential measurements).A first batch equilibrium study (Figs 1 and 2), showed an effective enhancement of adsorption capacity for humic acids as a function of STP concentration, in a non buffered media (pH of distilled water, close to 5.0). For example, visible absorption analysis of humic acids indicates an increase of 93% (500 mg l?1 PAC) and 133% (1000 mg l?1 PAC) in the carbon adsorption efficiency for a STP concentration from 0.2 to 1.0mM. A second batch equilibrium study (Figs 3 and 4) led to adsorption isotherms for humic acids in distilled water, as a function of STP concentration and initial pH value of the non buffered multicomponent system. Freundlich isotherms showed an increase in the adsorption capacity of the PAC for humic acids, with a decrease in pH and an increase in STP concentration. However, the adsorption capacity for humic acids is quite reduced at high pH values in presence of STP, in comparison with results obtained with distilled water.Electrokinetic measurements on PAC suspensions (Fig. 5) indicates that both humic acids and STP induce a negative variation of the zeta potential of carbon particles. In such a binary system, the zeta potential is a linear function of the pH; the negative surface charge of the carbon increasing with an elevation of pH (Fig. 6). Therefore, it appears that some adsorption of triphosphate polyanion from solution could occur, contributing then to the apparent negative surface charge of PAC particles.It has been previously showed that the type of anion in sodium salts, had little effect on the enhancement of adsorptive capacities of activated carbon for humic substances (Lafrance and Mazet, 1985), due to Na+ ions. However, adsorption of TP anions on the carbon surface may produce a source of repulsive charges, unfavourable to the co-adsorption of humic acids as the pH of the binary system reach more basic conditions. The influence of possible electrostatic interactions between adsorbates at the carbon surface, on the adsorption efficiency for humic acids, could then be studied by zeta potential measurements of PAC particles during the adsorption process. 相似文献

Abstract : An alkaloid-poor line of Washington lupin (Lupinus polyphyllus Lindl var SF/TA) was developed in an experiment started in 1982. The nutritive quality (alkaloid content, protein and amino acids, fat and fatty acids. macro- and micronutrients, fibre, sugars) yields, and seed quality of this line were studied. The results show that the total alkaloid content was low and varied in different seeds from 226 μg g?1 to 366 μg g?1 of dry matter. The main alkaloid was lupanine, but 16 other alkaloids (including sparteine and gramine) were also present. The var SF/TA cannot yet be used for human nutrition without processing although it would be a valuable protein crop. Yellowfin tuna enhance their hunting success in the vast pelagic environment by using their sense of smell to detect intact (uninjured) prey that are beyond visual range. However, the olfactory cues that tuna use would normally face huge and rapid dilution in the open ocean. We demonstrate that these prey odors are complexed within biologically derived lipid structures that probably delay the dilution of the amino acids to subthreshold concentrations and provide persistent arousal and search cues for the tuna. This may be the first demonstration of an extracorporeal biological function for liposomes. Tuna may also form chemical search images to maximize feeding efficiency. We demonstrate that the amino acid profiles of various prey species are consistent over time and between schools, which makes the formation of search images feasible. 相似文献

The objective of this study was to investigate the effects of supplementing with L-tryptophan (L-Trp) on milk protein synthesis using an immortalized bovine mammary epithelial (MAC-T) cell line. Cells were treated with 0, 0.3, 0.6, 0.9, 1.2, and 1.5 mM of supplemental L-Trp, and the most efficient time for protein synthesis was determined by measuring cell, medium, and total protein at 0, 24, 48, 72, and 96 h. Time and dose tests showed that the 48 h incubation time and a 0.9 mM dose of L-Trp were the optimal values. The mechanism of milk protein synthesis was elucidated through proteomic analysis to identify the metabolic pathway involved. When L-Trp was supplemented, extracellular protein (medium protein) reached its peak at 48 h, whereas intracellular cell protein reached its peak at 96 h with all L-Trp doses. β-casein mRNA gene expression and genes related to milk protein synthesis, such as mammalian target of rapamycin (mTOR) and ribosomal protein 6 (RPS6) genes, were also stimulated (p < 0.05). Overall, there were 51 upregulated and 59 downregulated proteins, many of which are involved in protein synthesis. The results of protein pathway analysis showed that L-Trp stimulated glycolysis, the pentose phosphate pathway, and ATP synthesis, which are pathways involved in energy metabolism. Together, these results demonstrate that L-Trp supplementation, particularly at 0.9 mM, is an effective stimulus in β-casein synthesis by stimulating genes, proteins, and pathways related to protein and energy metabolism. 相似文献

Resveratrol can affect the physiology or biochemistry of offspring in the maternal–fetal animal model. However, it exhibits low bioavailability in humans and animals. Fifteen-week SD pregnant female rats were orally administered bisphenol A (BPA) and/or resveratrol butyrate ester (RBE), and the male offspring rats (n = 4–8 per group) were evaluated. The results show that RBE treatment (BPA + R30) compared with the BPA group can reduce the damage caused by BPA (p < 0.05). RBE enhanced the expression of selected genes and induced extramedullary hematopoiesis and mononuclear cell infiltration. RBE increased the abundance of S24-7 and Adlercreutzia in the intestines of the male offspring rats, as well as the concentrations of short-chain fatty acids (SCFAs) in the feces. RBE also increased the antioxidant capacity of the liver by inducing Nrf2, promoting the expression of HO-1, SOD, and CAT. It also increased the concentration of intestinal SCFAs, enhancing the barrier formed by intestinal cells, thereby preventing BPA-induced metabolic disruption in the male offspring rats, and reduced liver inflammation. This study identified a potential mechanism underlying the protective effects of RBE against the liver damage caused by BPA exposure during the peri-pregnancy period, and the influence of the gut microbiota on the gut–liver axis in the offspring. 相似文献
