低聚果糖对C57BL/6小鼠肥胖预防及其肠道菌群的调节作用   总被引:1,自引:0,他引:1  
为探究低聚果糖(fructooligosaccharides,FOS)对C57BL/6小鼠肥胖预防及其肠道菌群的调节作用,将50只雄性5周龄C57BL/6小鼠随机分为5组,正常组食用正常饲料,其他组通过食用高脂饲料构建肥胖模型,正常组、模型组灌胃给予生理盐水,实验组灌胃给予高、低剂量的FOS,阳性对照组灌胃给予奥利司他,实验进行6周。采用高通量16S rRNA基因扩增测序、酶联免疫吸附测试等方法,分析FOS对C57BL/6小鼠的肥胖预防及肠道菌群的调节作用。结果表明,高、低剂量FOS都显著降低了肥胖模型小鼠的Lee’s指数、附睾脂肪指数(P<0.05),并显著升高了肥胖模型小鼠血清高密度脂蛋白胆固醇(high density lipoprotein cholesterol,HDL-C)浓度(P<0.05);高剂量FOS显著降低了肥胖模型小鼠的体质量增量及血清低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDL-C)、肿瘤坏死因子α(tumor necrosis factor α,TNF-α)、脂多糖(lipopolysaccha...  相似文献

Inflammatory bowel diseases (IBD) are commonly considered as Crohn’s disease and ulcerative colitis, but the possibility that the alterations in gut microbiota and oxidative stress may affect the course of experimental colitis in obese physically exercising mice treated with the intestinal alkaline phosphatase (IAP) has been little elucidated. Mice fed a high-fat-diet (HFD) or normal diet (ND) for 14 weeks were randomly assigned to exercise on spinning wheels (SW) for 7 weeks and treated with IAP followed by intrarectal administration of TNBS. The disease activity index (DAI), grip muscle strength test, oxidative stress biomarkers (MDA, SOD, GSH), DNA damage (8-OHdG), the plasma levels of cytokines IL-2, IL-6, IL-10, IL-12p70, IL-17a, TNF-α, MCP-1 and leptin were assessed, and the stool composition of the intestinal microbiota was determined by next generation sequencing (NGS). The TNBS-induced colitis was worsened in obese sedentary mice as manifested by severe colonic damage, an increase in DAI, oxidative stress biomarkers, DNA damage and decreased muscle strength. The longer running distance and weight loss was observed in mice given IAP or subjected to IAP + SW compared to sedentary ones. Less heterogeneous microbial composition was noticed in sedentary obese colitis mice and this effect disappeared in IAP + SW mice. Absence of Alistipes, lower proportion of Turicibacter, Proteobacteria and Faecalibacterium, an increase in Firmicutes and Clostridium, a decrease in oxidative stress biomarkers, 8-OHdG content and proinflammatory cytokines were observed in IAP + SW mice. IAP supplementation in combination with moderate physical activity attenuates the severity of murine colitis complicated by obesity through a mechanism involving the downregulation of the intestinal cytokine/chemokine network and oxidative stress, the modulation of the gut microbiota and an improvement of muscle strength.  相似文献

Recent reports on cocoa are appealing in that a food commonly consumed for pure pleasure might also bring tangible benefits for human health. Cocoa consumption is correlated with reduced health risks of cardiovascular diseases, hypertension, atherosclerosis, and cancer, and the health-promoting effects of cocoa are mediated by cocoa-driven phytochemicals. Cocoa is rich in procyanidins, theobromine, (?)-epicatechin, catechins, and caffeine. Among the phytochemicals present in consumed cocoa, theobromine is most available in human plasma, followed by caffeine, (?)-epicatechin, catechin, and procyanidins. It has been reported that cocoa phytochemicals specifically modulate or interact with specific molecular targets linked to the pathogenesis of chronic human diseases, including cardiovascular diseases, cancer, neurodegenerative diseases, obesity, diabetes, and skin aging. This review summarizes comprehensive recent findings on the beneficial actions of cocoa-driven phytochemicals in molecular mechanisms of human health.  相似文献

Objective: Stress has been identified as a significant factor in health and in racial/ethnic health disparities. A potential mediator in these relationships is body weight. Design: Cross-sectional and longitudinal relationships between stress, race, and body weight were examined in an ethnically diverse sample of overweight and obese women with Type 2 diabetes (n = 217) enrolled in a behavioral weight loss program. Main Outcome Measures: Stress (Perceived Stress Scale) was assessed at baseline only and body weight (body mass index) was assessed at baseline and 6 months. Tea is one of the most heavily consumed beverages in the world. The relationship between tea drinking and human health is becoming a subject of intense study by scientists throughout the world. In this paper, we first provide a comprehensive analysis of the medical literature on tea published in China during the past 20 years, and then highlight some recent studies in China on the relationship between tea and several human diseases. During the period 1982–2002, 691 research papers related to tea and health have been published in 290 Chinese journals. These studies showed that tea and tea constituents have various biological activities and suggested that tea drinking might be beneficial to human health. Tea has potential in the prevention or adjuvant treatment of several diseases including cancer, cardiovascular diseases and obesity. The trend and future direction in medical research on tea in China are also briefly discussed.  相似文献

Historically, carbohydrates have been thought to play only a minor role in promoting weight gain and in predicting the risk of development of chronic disease. Most of the focus had been on reducing total dietary fat. During the last 20 years, fat intake decreased, while the number of individuals who were overweight or developed a chronic conditions have dramatically increased. Simultaneously, the calories coming from carbohydrate have also increased. Carbohydrates can be classified by their post-prandial glycemic effect, called the glycemic index or glycemic load. Carbohydrates with high glycemic indexes and high glycemic loads produce substantial increases in blood glucose and insulin levels after ingestion. Within a few hours after their consumption, blood sugar levels begin to decline rapidly due to an exaggerated increase in insulin secretion. A profound state of hunger is created. The continued intake of high-glycemic load meals is associated with an increased risk of chronic diseases such as obesity, cardiovascular disease, and diabetes. In this review, the terms glycemic index and glycemic load are defined, coupled with an overview of short- and long-term changes that occur from eating diets of different glycemic indexes and glycemic loads. Finally, practical strategies for how to design low-glycemic-load diets consisting primarily of low-glycemic carbohydrates are provided.  相似文献

Eating represents a choice among many alternative behaviors. The purpose of this review is to provide an overview of how food reinforcement and behavioral choice theory are related to eating and to show how this theoretical approach may help organize research on eating from molecular genetics through treatment and prevention of obesity. Special emphasis is placed on how food reinforcement and behavioral choice theory are relevant to understanding excess energy intake and obesity and how they provide a framework for examining factors that may influence eating and are outside of those that may regulate energy homeostasis. Methods to measure food reinforcement are reviewed, along with factors that influence the reinforcing value of eating. Contributions of neuroscience and genetics to the study of food reinforcement are illustrated by using the example of dopamine. Implications of food reinforcement for obesity and positive energy balance are explored, with suggestions for novel approaches to obesity treatment based on the synthesis of behavioral and pharmacological approaches to food reinforcement. Meta-analysis was used to integrate research on the relations between parental socialization behavior and child and adolescent physical activity (PA) levels. Four major databases were examined: PubMED, ERIC, Web of Science, and PsychLit (1960 -2005). Thirty studies met the following inclusion criteria: (a) child age (2-18 years) and (b) statistical information permitting calculation of an effect size between parent socialization behavior and child PA. Mean age of participants across studies ranged from 2.54 to 15.5 years. The unweighted mean and median effect sizes (as indexed by r) were .17 and .13, respectively, indicating that a moderate positive relation exists between parental support and modeling behavior and child and adolescent PA levels. The moderating effect of type of parental socialization behavior, population characteristics, and methodological factors were investigated. Theoretical and methodological implications concern the inclusion of mediated models of parental influence and the use of longitudinal investigations in determining causal direction. From an applied viewpoint, these results are useful for the design of future, more effective childhood obesity prevention programs by suggesting child-age-appropriate parental influences.
