To gain a better understanding of the ultra-high molecular weight polyethylene (UHMWPE) wear mechanism in the physiological environment, the effects of protein and lipid constituents of synovial fluid on the specific wear rate of UHMWPE were examined experimentally. The multidirectional sliding pin-on-plate wear tester was employed to simulate the simplified sliding condition of hip joint prostheses. Bovine serum γ-globulin and synthetic l--DPPC were used as model protein and lipid constituents of synovia, respectively. Results of the wear test indicated that the UHMWPE wear rate primarily depended on the protein concentration of the test lubricant. Lipids acted as a boundary lubricant and reduced polyethylene wear in the low protein lubricants. However, the polyethylene wear rate increased with increasing lipid concentrations if the protein concentration was within the physiological level. Increased interactions between protein and lipid molecules and lipid diffusion to polyethylene surface might be responsible for the increased wear. 相似文献

针对下肢假肢穿戴者骑行相位识别的问题,提出基于灰狼算法优化的支持向量机(GWO-SVM)分类模型. 建立下肢多源信息系统,采集膝关节、踝关节的加速度信号以及膝关节角度信号. 应用奇异值分解,对采集到的信号进行降噪处理. 在对信号进行降噪处理之后,为了避免单一信号不确定的影响,从数据冗余角度,选取各信号的特征点,开展归一化处理,组成多维特征向量,作为SVM分类模型的输入. 为了能够进一步提高分类精度,加强全局优化能力,利用GWO算法对核参数进行优化. 通过与PSO-SVM分类模型、GA-SVM分类模型对比表明,基于GWO优化的SVM分类模型对骑行相位的识别率为94%,高于其他方法优化的SVM分类模型. 相似文献

The discovery of novel intronic variants in the ABCA4 locus has contributed significantly to solving the missing heritability in Stargardt disease (STGD1). The increasing number of variants affecting pre-mRNA splicing makes ABCA4 a suitable candidate for antisense oligonucleotide (AON)-based splicing modulation therapies. In this study, AON-based splicing modulation was assessed for 15 recently described intronic variants (three near-exon and 12 deep-intronic variants). In total, 26 AONs were designed and tested in vitro using a midigene-based splice system. Overall, partial or complete splicing correction was observed for two variants causing exon elongation and all variants causing pseudoexon inclusion. Together, our results confirm the high potential of AONs for the development of future RNA therapies to correct splicing defects causing STGD1. 相似文献

Disruption of retinal pigment epithelial (RPE barrier integrity is a hallmark feature of various retinal blinding diseases, including diabetic macular edema and age-related macular degeneration, but the underlying causes and pathophysiology are not completely well-defined. One of the most conserved phenomena in biology is the progressive decline in mitochondrial function with aging leading to cytopathic hypoxia, where cells are unable to use oxygen for energy production. Therefore, this study aimed to thoroughly investigate the role of cytopathic hypoxia in compromising the barrier functionality of RPE cells. We used Electric Cell-Substrate Impedance Sensing (ECIS) system to monitor precisely in real time the barrier integrity of RPE cell line (ARPE-19) after treatment with various concentrations of cytopathic hypoxia-inducing agent, Cobalt(II) chloride (CoCl2). We further investigated how the resistance across ARPE-19 cells changes across three separate parameters: Rb (the electrical resistance between ARPE-19 cells), α (the resistance between the ARPE-19 and its substrate), and Cm (the capacitance of the ARPE-19 cell membrane). The viability of the ARPE-19 cells and mitochondrial bioenergetics were quantified with 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay and seahorse technology, respectively. ECIS measurement showed that CoCl2 reduced the total impedance of ARPE-19 cells in a dose dependent manner across all tested frequencies. Specifically, the ECIS program’s modelling demonstrated that CoCl2 affected Rb as it begins to drastically decrease earlier than α or Cm, although ARPE-19 cells’ viability was not compromised. Using seahorse technology, all three concentrations of CoCl2 significantly impaired basal, maximal, and ATP-linked respirations of ARPE-19 cells but did not affect proton leak and non-mitochondrial bioenergetic. Concordantly, the expression of a major paracellular tight junction protein (ZO-1) was reduced significantly with CoCl2-treatment in a dose-dependent manner. Our data demonstrate that the ARPE-19 cells have distinct dielectric properties in response to cytopathic hypoxia in which disruption of barrier integrity between ARPE-19 cells precedes any changes in cells’ viability, cell-substrate contacts, and cell membrane permeability. 针对视网膜图像采集过程中由于疾病引起的图像光照反射过强问题,提出了一种修正的形态学与Otsu相结合的无监督视网膜血管分割算法。首先运用形态学中的高低帽变换增强血管与背景的对比度;然后提出了一种修正方法,消除部分由视网膜疾病引起的光照问题;最后使用Otsu阈值方法分割血管。算法在DRIVE和STARE视网膜图像数据库中进行了测试,实验结果表明,DRIVE数据库中的分割精度为0.9382,STARE数据库中的分割精度为0.9460,算法的执行时间为1.6s。算法能够精确地分割出视网膜血管,与传统的无监督视网膜血管分割算法相比,算法的分割精度高、抗干扰能力强。 相似文献

采用国产3Y-TZP亚微米粉体,通过双向模压结合等静压成形有效克服了条形试样的烧结弯曲,探讨了24h排胶工艺对预烧试样力学性能的影响,通过对预烧试样热膨胀曲线的测定、不同烧结温度试样力学性能和密度的比较及XRD定量分析确定了适宜的烧结工艺。结果表明,牙科支架用陶瓷,将预烧试样升至1550℃保温0.5h,排胶工艺对预烧试样的抗弯强度和断裂韧性并无显著影响,对于即可获得足够的抗弯强度,提高其断裂韧性。 相似文献

为了实现牙颌模型的三维数据可视化,通过计算机逼真的显示牙列形态,并进行交互测量及变形等操作。在口腔修复体计算机辅助设计中,以OpenGL为渲染工具,结合自主设计的牙颌模型三维激光扫描仪获取的点云数据,采用基于B样条曲面的三维重建技术,以Visual C++为开发工具实现数字化牙颌模型的三维可视化。以Visual C++和OpenGL为工具,减少了程序设计难度,提高了执行效率,图形显示更加逼真流畅。本系统的设计满足了口腔医生的需求,给患者带来了便利,并促进了口腔修复体的开发制作。 相似文献
