Di (2-ethylhexyl) phthalate (DEHP) is widely used as a plasticizer, and human exposure to DEHP is widespread and frequent. However, information about the combined effect of DEHP and ultraviolet (UV)-B on the skin are still limited. We investigated the cytotoxic effects of DEHP and UV-B on HaCaT keratinocytes and evaluated the related underlying mechanisms involving endoplasmic reticulum (ER) stress signals and the disruption of junction complexes as an effective target for skin inflammation. Our results revealed that co-treatment with DEHP and UV-B irradiation alleviated the cell cytotoxicity and markedly decreased X-box binding protein 1 (XBP1), endoplasmic reticulum oxidoreductase 1 alpha (Ero1α), and C/EBP homologous protein (CHOP) whereas a single dose of 40 mJ/cm2 UV-B generated mild ER stress to slightly less or similar levels as that seen with DEHP. DEHP was also shown to inhibit tight junctions (TJs) after UV-B irradiation, increased apoptosis by altering apoptotic gene Bax and stress kinases, JNK, and p38 MAPK. Furthermore, exposure of HaCaT cells to DEHP and UV-B irradiation resulted in the marked suppression of the nuclear factor kappa B (NF-κB)/p65 signaling pathway. Taken together, our data suggest that nontoxic DEHP and UV-B irradiation regulated ER stress and epidermal TJ disruption with the induction of apoptosis activation and the secretion of proinflammatory cytokines such as interleukin 1 beta (IL-1β) and IL-6 in human keratinocytes. Further investigation is needed to confirm the mechanisms implicated in its toxicity and determine the effects of exposure to DEHP and UV-B irradiation on markers involved in this study. 相似文献

Industrially important di‐carboxylic acids are synthesized from mono‐carboxylic unsaturated and unsaturated fatty acids. In this study, the aim is to perform the simultaneous catalytic oxidative C=C cleavage of oleic acid (OA) to azelaic acid and pelargonic acid, and oxidation of the terminal methyl group in pelargonic acid to azelaic acid using cobalt‐ and manganese‐acetate as catalyst, hydrogen bromide as co‐catalyst and air in acetic acid at elevated pressure (2.8–5.8 barg) and temperature (353–383 K). Oxygen solubility is determined under varying pressure, temperature and OA loading. The effect of OA loading, pressure and temperature on OA conversion and azelaic acid selectivity is studied by varying one variable at a time; however, the presence of the synergistic effect of the catalyst and co‐catalyst is investigated by central composite design assisted response surface methodology. Oxidation of terminal methyl group in saturated fatty acid is also confirmed by the oxidation of stearic acid to octadecanedioic acid using identical oxidation conditions of OA. Oxidation products of fatty acids are quantified by gas chromatographic analysis. The innovation of the work is thus the ability of the catalytic system to perform a total oxidation of a terminal methyl group of the hydrocarbon chain. OA oxidation kinetics relating to catalyst and co‐catalyst concentration along with oxygen solubility at elevated temperature and pressure is established. Melatonin (N-acetyl-5-methoxytryptamine, MLT), the main hormone produced by the pineal gland, not only regulates circadian rhythm, but also has antioxidant, anti-ageing and immunomodulatory properties. MLT plays an important role in blood composition, medullary dynamics, platelet genesis, vessel endothelia, and in platelet aggregation, leukocyte formula regulation and hemoglobin synthesis. Its significant atoxic, apoptotic, oncostatic, angiogenetic, differentiating and antiproliferative properties against all solid and liquid tumors have also been documented. Thanks, in fact, to its considerable functional versatility, MLT can exert both direct and indirect anticancer effects in factorial synergy with other differentiating, antiproliferative, immunomodulating and trophic molecules that form part of the anticancer treatment formulated by Luigi Di Bella (Di Bella Method, DBM: somatostatin, retinoids, ascorbic acid, vitamin D3, prolactin inhibitors, chondroitin-sulfate). The interaction between MLT and the DBM molecules counters the multiple processes that characterize the neoplastic phenotype (induction, promotion, progression and/or dissemination, tumoral mutation). A novel thermo- and pH-sensitive hydrogels based on N-isopropylacrylamide and containing acrylic acid (mono carboxylic), oxalic acid (oxalic) (dicarboxylic), and citric acid (citric) (tricarboxylic) have been prepared using mutual radiation technique. Fourier transform infrared spectroscopy was used to identify the structure of the prepared hydrogels. The surface morphology of the hydrogels was observed using scanning electron microscopy. Swelling percent found to be decreased with the increase in the temperature. It was proved that the prepared hydrogels were extremely sensitive to both temperature and pH. The possibility of using such hydrogels as a drug carrier was estimated using acetylsalicylic acid (aspirin) as a model drug. The drug release in pH 6.8 was more than that in pH 2. Owing to its considerable drug release, good swelling, it might be useful for its application as a drug delivery system in small intestine. 相似文献

To significantly reduce the bleeding of 4,4′,(5′)‐di(tert‐butylcyclohexano)‐18‐crown‐ 6 (DtBuCH18C6), an improved novel macroporous silica‐based polymeric composite (DtBuCH18C6+TBP)/SiO2‐P was synthesized. It was performed by impregnating and immobilizing DtBuCH18C6 into the pores of the SiO2‐P particles via the molecular modification of DtBuCH18C6 with a tri‐n‐butyl phosphate (TBP) through hydrogen bonding. The adsorption of a few typical simulated fission and non‐fission products Pd(II), La(III), Na(I), K(I), Sr(II), Ba(II), Ru(III), Cs(I), Mo(VI), and Y(III) onto (DtBuCH18C6+TBP)/SiO2‐P was investigated at 323 K. It was done by examining the effect of contact time and the HNO3 concentration in a range of 0.1–5.0 M. Sr(II), one of the main heat emitting nuclides, showed optimum adsorption onto (DtBuCH18C6+TBP)/SiO2‐P in 2.0 HNO3, while others showed very weak or almost no adsorption except a portion of Ba(II). The leaching of TBP and DtBuCH18C6 from (DtBuCH18C6+TBP)/SiO2‐P was evaluated. The average content of DtBuCH18C6, 298.7 ppm, leached from (DtBuCH18C6+TBP)/SiO2‐P in 2.0 M HNO3 at 323 K was obviously lower than that of 797.3 ppm leached from DtBuCH18C6/SiO2‐P at 298 K. The significant reduction of DtBuCH18C6 leaching from its macroporous silica‐based polymeric adsorbent was achieved. It is useful for the recycle operation of the silica‐based DtBuCH18C6 impregnated polymeric composite in chromatographic partitioning of Sr(II) from high level liquid waste (HLLW). 相似文献
