We describe an extremely simple method by which optically sectioned fluorescence images may be obtained with conventional microscopes using laser illumination. A one-dimensional grid pattern is introduced into the illumination system, together with a rotating ground glass diffuser. This causes an image of the grid pattern to be projected into the specimen. Images taken at three spatial positions of the grid are processed in a simple manner to provide optically sectioned images of fluorescent specimens.  相似文献

UVscope is a portable multi-pixels photon detector developed at IASF-Pa to support experimental activities in the high-energy astrophysics and cosmic rays field. The instrument, working in single photon counting mode, is designed to directly measure light flux in the wavelengths range 300-650 nm. Thanks to its features and operational flexibility, the instrument can be used in a wide field of applications where the knowledge of the environmental luminosity is required, as in the characterization of sites for ground-based Cherenkov and fluorescence telescopes, and for cross-calibration of their cameras.The present version of UVscope is based on a Multi Anode Photo Multiplier Tube and it is completed by a motorized mount and a filter wheel which allow to make low-light measurements, at programmed pointing directions and at different filter wavelengths. In this paper, the instrument is firstly presented in all its components; then the procedures adopted for its absolute and relative calibration are detailed. The performance of UVscope is evaluated and, finally, current and planned scientific applications are described.  相似文献

Fluorescence spectra of coal derived asphaltenes sourced from Barari coke plant situated at Dhanbad, India, have been studied as a function of concentration in benzene, toluene and carbon tetrachloride with an eye to study the self-aggregation of the solute. Aggregation has been found to be a gradual process and the concentration characterizing onset of aggregation process coincide for the solvents. The critical aggregation constants (CAC) appear to be ca 90-100 and 180-200 for all the solvents. A small red shift of fluorescence band is observed when traces of water (∼0.05%, v/v) are present in carbon tetrachloride solution.  相似文献

采用帕纳科Axios4.0X射线荧光光谱仪,建立了对碳酸盐中的CaO、MgO含量测定的X射线荧光光谱法。称取300目的样品,用低压聚乙烯镶边衬底,32MPa压力制备碳酸盐样品,用X射线荧光光谱法测定碳酸盐中的CaO、MgO含量;采用国家标准物质验证方法,其结果与标准值相符,CAO的精密度(RSD,n=9)小于0.3%,MgO的精密度(RSD,n=9)小于11%。该方法制样方便、快捷、经济,分析周期短,测量准确度、精密度较好,与容量法和熔片法结果一致,可以满足并适用于常规碳酸盐中的CaO、MgO含量的测定。  相似文献

Fluorescent quantum dots (QDs) are a new class of fluorescent label and have been extensively used in cell imaging. Streptavidin-conjugated QDs have a diameter of ca. 10–15 nm; therefore when used as probes to label cell-surface biomolecules, they can provide contrast enhancement under atomic force microscopy (AFM) and allow specific proteins to be distinguished from the background. In addition, the size and fluorescent properties potentially make them as probes in correlative fluorescence microscopy (FM) and AFM. In this study, we tested the feasibility of using QD-streptavidin conjugates as probes to label wheat germ agglutinin (WGA) receptors on the membrane of human red blood cells (RBCs) and simultaneously obtain fluorescence and AFM images. The results show that the distribution of QDs labeled on human RBCs was non-uniform and that the number of labeled QDs on different erythrocytes varied significantly, which perhaps indicates different ages of the erythrocytes. Thus, QDs may be employed as bifunctional cell-surface markers for both FM and AFM to quantitatively investigate the distribution and expression of membrane proteins or receptors on cell surface.  相似文献

一、引言 微量铀的测量方法已有大量报道,但绝大部分的方法,一般需经化学分离,因此不适用于远距离的在线分析。近年来,随着光纤传感器的出现及激光荧光技术的发展,远距离在线分析Purex流程中铀和钚的方法引起了人们的兴趣。J.R mer等报道了以染料激光器为光源的光纤分光光度法,用以测定Purex流程中U(Ⅳ),U(Ⅵ),Pu(Ⅲ),Pu(Ⅳ)及Pu(Ⅲ)/Pu(Ⅳ)的混合物。R.A.Malstrom等研究了用416nm激光为激发光源的 光纤荧光光度计,并讨论了用于Savannah河后处理厂铀的在线分析问题。 为探索一种比较经济、简单,又能实现铀的远距离在线分析的测量方法,我们研制了一种以小型脉冲氮分子激光器为激发光源的光纤激光荧光法。我们考虑到Purex流程的实际情况,还研究了硝酸体系中各种因素对铀的荧光强度测量的影响。  相似文献

The assembly of multifunctional nanocomposite materials is demonstrated by exploiting the molecular sieving property of SBA‐16 nanoporous silica and using it as a template material. The cages of the pore networks are used to host iron oxide magnetic nanoparticles, leaving a pore volume of 0.29 cm3 g?1 accessible for drug storage. This iron oxide–silica nanocomposite is then functionalized with amine groups. Finally the outside of the particle is decorated with antibodies. Since the size of many protein molecules, including that of antibodies, is too large to enter the pore system of SBA‐16, the amine groups inside the pores are preserved for drug binding. This is proven using a fluorescent protein, fluorescein‐isothiocyanate‐labeled bovine serum albumin (FITC‐BSA), with the unreacted amine groups inside the pores dyed with rhodamine B isothiocyanate (RITC). The resulting nanocomposite material offers a dual‐targeting drug delivery mechanism, i.e., magnetic and antibody‐targeting, while the functionalization approach is extendable to other applications, e.g., fluorescence–magnetic dual‐imaging diagnosis.  相似文献
