Black-bone silky fowl (Gallus gallus domesticus Brisson) (BSF) muscle was hydrolyzed by papain, and the hydrolysate was separated by preparative high performance liquid chromatography (HPLC). The amino acid composition of the BSF hydrolysate (BSFH) and its fractions was determined by HPLC precolumn derivation with 2,4-dinitrofluorobenzene. The molecular weight (MW) distribution of the BSFH and its fractions was measured by a peptide column on an HPLC system. Antioxidant activities of the BSFH and its fractions were studied by testing the reducing power and four radical scavenging systems: superoxide anion (O2), hydroxyl (·OH), 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+) radicals. The results demonstrated that the BSFH had strong antioxidant capacity to scavenge O2, DPPH and ABTS+, and displayed strong reducing power, but revealed less powerful ability to scavenge ·OH. Fraction II of the BSFH exhibited the highest activity in scavenging O2 and DPPH, and reducing power, whereas fraction I displayed the strongest ·OH scavenging ability. Besides Glu, Asp and Gly, the rich amino acids of Ala and Leu played an important role in antioxidant activity. The small-size peptides with MW ranging from approximately 200-6000 Da probably contributed to higher antioxidant activity. Results from this study indicated that BSFH and its fractions could be used as food additives and diet nutrients.  相似文献

A new DNA extraction method suitable for a wide variety of complex food matrices has been devised and applied in combination with a new polymerase chain reaction (PCR) method for the sensitive detection of a specific DNA sequence univocally identifying the presence of potentially allergenic hazelnut (Corylus avellana). A 156 base pair amplicon corresponding to an internal region of the complementary DNA of the major hazelnut allergen (Cor a 1) was designed, and was found to be highly specific; the method was tested on both pure and complex food matrices and was found to be able to confirm the presence of hazelnut down to 5 pg of its DNA. The sequence of the amplicon was further confirmed by high-performance liquid chromatography (HPLC) analysis with a specific peptide nucleic acid (PNA) probe. A 15-mer PNA probe was expressly designed and synthesized to hybridize an internal sequence of the previously described amplicon. Given its reported sequence specificity and high hybridization efficiency, the PNA probe was used to develop an anion-exchange HPLC method allowing for a fast and reliable confirmation of the identity of the amplified products. The PCR–HPLC method was successfully tested on commercial samples, allowing for the detection of the presence of potentially hidden allergens even in products where the presence of hazelnut as an ingredient or possible contaminant was not reported.  相似文献

In order to accurately predict the varying of formwork pressure over time, it is necessary to consider various factors influencing the development of formwork pressure. A prediction model has been previously proposed, but that model has some limitations in that only intrinsic material characteristics are taken into account. Extrinsic effects such as wall friction, formwork flexibility, and external temperature are excluded in the model. This study focuses on the wall friction effect as one of the extrinsic factors. First, by incorporating the intrinsic model and friction stress acting on the interface, a method of calculating formwork pressure considering the wall friction effect is suggested. To find out how much friction stress is acting on the interface and how it varies over time, formwork pressure tests were performed with circular column formworks having three different diameters. In these columns, the vertical pressure at the bottom and the lateral pressures were measured. To calibrate parameters of the intrinsic model for the same material as that used in the formwork pressure tests, additional tests were conducted with a specially designed apparatus that can exclude effects of extrinsic factors. From tests and analysis results, it was found that wall friction greatly affects the variation of formwork pressure over time. The newly suggested calculation method can give a good prediction of real formwork pressure.  相似文献

The concentrations of 15 "priority pollutant" semivolatile and particulate polycyclic aromatic hydrocarbons (PAHs) were determined in three sets of samples supplied by the National Institute for Standards and Technology (NIST) as part of an interlaboratory analytical exercise. The purpose of the exercise, organized by NIST and the U.S. Environmental Protection Agency, was to determine the comparability of measurements for various organic analytes among the participating laboratories, and to establish consensus values for SRM 1649a and interim materials. The commercially available SRM 1649a Atmospheric Urban Dust and two subsamples of this popular reference material were analyzed: an extract designated as Air Particulate Extract, and a resieved portion labeled Air Particulate I. The method used in our laboratory for the exercise consists of the extraction of the PAHs from the solid samples by ultrasonication, followed by separation and quantification using high-performance liquid chromatographyfluorescence detection. The accuracy and precision of the results obtained by our analytical protocol and by 14 other participating laboratories were evaluated using the International Union of Pure and Applied Chemistry guidelines of z-scores ( = 25% of the exercise assigned value) and p -scores. Using these guidelines the accuracy of our method provided results that are satisfactory for all 15 target PAHs (|z| h 2) determined in the Air Particulate Extract and, except for fluorene, in the Air Particulate I sample. Finally, application of the methodology is demonstrated for the quantification of PAHs present at the pg m m 3 range in PM 2.5 samples collected from 163 m 3 of air in the Los Angeles basin.  相似文献

In the present study, we show that high-resolution 29Si and 27Al magic angle spinning (MAS) nuclear magnetic resonance (NMR) can be a powerful tool for analyzing actual concrete mixes. The influence of the amount of silica fume and of the type of cement in high-performance concrete (HPC), as well as the influence of the type of silica fume and of the granular packing in ultrahigh-performance concrete (UHPC) were investigated. Significant effects on the amount and shape of C-S-H, on the incorporation of aluminum in the C-S-H structure, and on the distribution of aluminum-containing hydrates were observed. Nuclear magnetic relaxation of protons was also performed and it showed the fractal feature of the pore size distribution in UHPC and the higher amount of larger pores in HPC. The microstructure of the surface of these same formulations leached by mineral water for up to 1 year exhibits slight modifications.  相似文献

Primary and secondary containment structures are the major components of the nuclear power plant (NPP). The performance requirements of the concrete of containment structures are mainly radiological protection, structural integrity and durability, etc. For this purpose, high-performance heavy density concrete with special attributes can be used. The aggregate of concrete plays an essential role in modifying concrete properties and the physico-mechanical properties of the concrete affect significantly on its shielding properties. After extensive trials and errors, 15 concrete mixes were prepared by using the coarse aggregates of barite, magnetite, goethite and serpentine along with addition of 10% silica fume (SF), 20% fly ash (FA) and 30% ground granulated blast-furnace slag (GGBFS) to the total content of OPC for each mix. To achieve the high-performance concrete (HPC- grade M60), All concrete mixes had a constant water/cement ratio of 0.35, cement content of 450 kg/m3 and sand-to-total aggregate ratio of 40%. Concrete density has been measured in the case of fresh and hardened. The hardened concrete mixes were tested for compressive strength at 7, 28 and 90 days. In some concrete mixes, compressive strength was also tested up to 90 days upon replacing sand with the fine portions of magnetite, barite and goethite. The attenuation measurements were performed by using gamma spectrometer of NaI (Tl) scintillation detector. The utilized radiation sources comprised 137Cs and 60Co radioactive elements with photon energies of 0.662 MeV for 137Cs and two energy levels of 1.173 and 1.333 MeV for 60Co. Some shielding factors were measured such as half-value layer (HVL), tenth-value layer (TVL) and linear attenuation coefficients (μ). Experimental results revealed that, the concrete mixes containing magnetite coarse aggregate along with 10% SF reaches the highest compressive strength values exceeding over the M60 requirement by 14% after 28 days of curing. Whereas, the compressive strength of concrete containing barite aggregate was very close to M60 and exceeds upon continuing for 90 days. The results indicated also that, the compressive strength of the high-performance heavy density concrete incorporating magnetite as fine aggregate was significantly higher than that containing sand by 23%. Also, concrete made with magnetite fine aggregate improved the physico-mechanical properties than the corresponding concrete containing barite and goethite. Therefore, high-performance concrete incorporating magnetite as fine aggregate enhances the shielding efficiency against γ-rays.  相似文献

A high-performance oscillatory combustor for burning low-grade coal was developed and operated continuously for periods of up to 24 hours. The oscillatory combustor is of extremely simple mechanical design. It has an output capacity of 15–25 kW and its system and combustion efficiencies are 65% and close to 100% respectively. The developed oscillatory combustor is environment-friendly and its pollutant emission is far below that of conventional combustors burning low-grade coal.  相似文献
