表达HIV-1 Gag-Pol抗原的重组痘病毒的构建及免疫原性

目的构建稳定高效表达HIV-1Gag-Pol蛋白的重组痘病毒载体疫苗,并检测其体液免疫效果。方法将修饰型HIV-1Gag-Pol基因插入到痘病毒表达载体pSC11m2启动子P7·5下游,经与野生型痘病毒同源重组后,进行蓝白斑筛选。用筛选得到的重组病毒rM-GP转染HEK293细胞,经SDS-PAGE和Westernblot检测表达蛋白。用该重组病毒免疫BALB/c小鼠,并检测体液免疫应答。结果已筛选到稳定高效表达HIV-1Gag-Pol蛋白的重组痘病毒载体rM-GP,经Westernblot,在免疫小鼠血清中检测到抗HIV-1p24蛋白的特异性抗体。结论重组痘病毒载体rM-GP能稳定高效表达HIV-1Gag-Pol蛋白,并能引起有效的体液免疫应答。     

Citrate-stabilized CdSe/CdS quantum dots as fluorescence probe for protein determination

A rapid, ultrasensitive and convenient fluorescence measurement technology based on the enhancement of the fluorescence intensity resulting from the interaction of functionalized CdSe/CdS quantum dots (QDs) with bovine serum albumin (BSA) was proposed. The citrate-stabilized CdSe/CdS (QDs) were synthesized by using Se powder and Na₂S as precursors instead of any pyrophoric organometallic precursors. The modified CdSe/CdS QDs are brighter and more stable against photobleaching in comparison with organic fluorophores. At pH 7.0, the fluorescence signal of CdSe/CdS is enhanced by increasing the concentration of BSA in the range of 0.1–10 μg/mL, and the low detection limit is 0.06 μg/mL. A linear relationship between the enhanced fluorescence peak intensity (ΔF) and BSA concentration (c) is established using equation ΔF=50.7c+16.4 (R=0.996 36). Results of determination for BSA in three synthetic samples are identical with the true values, and the recovery (98.9%–102.4%) and relative standard deviation (RSD, 1.8%–2.5%) are satisfactory.     

公共部门人力资源管理特点及其分析

荧光法研究3种黄酮化合物与牛血清白蛋白的结合作用

运用荧光光谱法研究了生理酸度条件下（pH=7.4）3种黄酮化合物白杨素、芹菜素和桑色素与牛血清白蛋白（BSA）的结合作用。结果表明,3种黄酮化合物均能与BSA形成基态复合物导致BSA内源荧光猝灭,猝灭机理为静态猝灭。结合常数K（芹菜素）〉K（白杨素）〉K（桑色素）,结合位点数近似等于1,白杨素与BSA之间以疏水作用为主,而芹菜素、桑色素与BSA之间的作用力主要是氢键和范德华力。黄酮分子结构中羟基位置及个数影响其与BSA间的结合作用,C4＇-OH对结合有促进作用,而C3-OH的取代则导致作用力减弱。由Frster非辐射能量转移理论,计算出3种黄酮化合物在蛋白质中的结合位置与212位色氨酸残基间的距离分别为2.44 nm（白杨素）、3.34 nm（芹菜素）和3.31 nm（桑色素）。

人白细胞介素-24基因的克隆、表达和纯化

目的对人白细胞介素-24(hIL-24)进行克隆、表达和纯化。方法分离人外周血单个核细胞,ConA刺激培养,提取细胞总RNA,应用RT-PCR技术从外周血单个核细胞中扩增hIL-24成熟肽编码区,定向克隆至融合表达载体pGEX-4T-1,重组载体pGEX-4T-1/hIL-24经DNA测序鉴定后,转化E.coliBL21(DE3),IPTG诱导表达,阴离子交换层析纯化融合蛋白,SDS-PAGE和Westernblot鉴定。结果所得hIL-24基因经序列分析,与GenBank公布一致。所构建融合表达载体pGEX-4T-1/hIL-24测序正确,转化BL21(DE3)后,37℃诱导表达相对分子质量约为44000的融合蛋白,主要以包涵体形式存在,占菌体总蛋白的30.63%。经纯化后,纯度达85%以上。Westernblot检测能与兔抗人IL-24的多克隆抗血清发生结合反应。结论已成功构建了hIL-24的融合表达载体pGEX-4T-1/hIL-24,并在大肠杆菌中高效表达,纯化后获得了高纯度的融合蛋白,为hIL-24的功能及活性研究奠定基础。     

基于信息物理融合的骨折复位机器人系统构建

针对骨折治疗中传统复位方法（手法复位、牵引复位、手术复位）存在的二次损伤、肌能失调、患肢僵硬、破坏血运及术后矫正动态性能差等问题,提出了一种人-机-物融合的信息物理系统（CPHS）用于指导机器人的复位动作。首先,从CPHS的数字孪生、信息感知、系统集成、手术流程、模拟复位等方面论证了并联机器人信息物理系统的构成,将机器人的高定位精度及可重复性与微创方法有效结合,从而指导医生完成仿真规划及术中监控等一系列操作;其次,根据临床骨折复位过程,在机器人操作下对5组不同骨折姿态的模拟骨折病例进行了复位实验;最后,计算出每组实验复位后的移位残余及角度误差,并与传统复位方法的相应数据进行对比。实验结果表明,CPHS骨折复位机器人相较于传统复位方法,在骨折复位过程和患者术后康复上均有着明显的优势。     

Cognitive work analysis to comprehend operations and organizations in the mining industry

Complex industrial systems, including mining, have a prominent challenge in understanding the interrelationship among the cognitive processes, working environment and available equipment. The concept of cognitive work analysis(CWA) transcends the traditional analytic methods of evaluating human tasks solely based on perceptual and physical traits, and rather implements the notions of behavioral and cognitive awareness indispensable for the intricacy of modern technology. In the last few decades, academic and industrial settings employ this type of analysis to set a suitable standard for a system's safety feasibility, and as a result reduce human-based errors. This research paper analyzes current CWA methods and proposes a five-level quantification model portraying the overall cognitive quality of a mining operation.     

Scanning electron microscopy of nerve fibers in human fetal cochlea

The cochleas of four human fetuses ranging 22–25 weeks gestation were studied by scanning electron microscopy (SEM) for the purpose of obtaining a better understanding of the nerve fiber arrangement in the human ear. After critical point drying, the specimens were dissected and the floor of the tunnel of Corti and the outer wall of Nuel's space were exposed for observation. Upper cochlear turns, especially the apical turn, seemed to be still immature. Observed nerve fibers were classified into three types:

• 1 Spiral fibers: Fibers traveling basalward and following the shape of the cochlea were found in both the tunnel of Corti and Nuel's space and believed to be the afferent nerves responsible for innervating the outer hair cells
• 2 Radial fibers: radiating outward from the osseous spiral lamina—one such radial fiber transversing high in the tunnel space (supposedly the efferent nerve servicing the outer hair cells), and another sort of radial fiber (found crossing the tunnel floor), the nature of which was uncertain.
• 3 Irregular fibers: Consisting of thin, randomly running fibers within the cochlea. The destination of these fibers was not determined, but possibly they represent transitory nerve branchings of afferent or more probably efferent nerves, which would later regress during maturation.
