Whispering gallery mode (WGM) microresonators have been used as optical sensors in fundamental research and practical applications. The majority of WGM sensors are passive resonators that require complex systems, thereby limiting their practicality. Active resonators enable the remote excitation and collection of WGM‐modulated fluorescence spectra, without requiring complex systems, and can be used as alternatives to passive microresonators. This paper demonstrates an active microresonator, which is a microdisk laser in a hyperboloid‐drum (HD) shape. The HD microdisk lasers are a combination of a rhodamine B‐doped photoresist and a silica microdisk. These HD microdisk lasers can be utilized for the detection of label‐free biomolecules. The biomolecule concentration can be as low as 1 ag mL?1, whereas the theoretical detection limit of the biosensor for human IgG in phosphate buffer saline is 9 ag mL?1 (0.06 aM). Additionally, the biosensors are able to detect biomolecules in an artificial serum, with a theoretical detection limit of 9 ag mL?1 (0.06 aM). These results are approximately four orders of magnitude more sensitive than those for the typical active WGM biosensors. The proposed HD microdisk laser biosensors show enormous detection potential for biomarkers in protein secretions or body fluids.  相似文献

Newborn Holstein (n = 48) and Jersey (n = 30) calves were studied to compare absorption of immunoglobulin G (IgG) from maternal colostrum (n = 39) or colostrum replacement containing an Ig concentrate derived from bovine serum (n = 39). Calves were also fed milk replacer with (n = 38) or without (n = 40) animal plasma (20% of crude protein) to 29 d of age to determine effect of plasma protein on IgG status, health, and growth. Calves were fed maternal colostrum or colostrum replacement at 1.5 and 13.5 h of age and provided a total of 250 or 249 and 180 or 186 g of IgG for Holsteins and Jerseys fed maternal colostrum or colostrum replacement, respectively. Milk replacer (12.5% DM) was fed at 31% of metabolic birth weight (2 feedings/d). Plasma was sampled at 0 h, 24 h, and weekly to determine IgG by turbidimetric immunoassay. At blood collection, calves were weighed and measured to determine growth. Health scores, fecal scores, and grain intake were measured daily. Plasma IgG at 24 h did not differ between calves fed maternal colostrum (13.78 +/- 0.39 g/ L) and colostrum replacement (13.96 +/- 0.38 g/L). Average daily gain, withers height, hip height, body length, heart girth, health, and incidence of diarrhea were not different between treatment groups. Calves fed maternal colostrum used feed more efficiently than calves fed colostrum replacement. Plasma IgG and performance were not affected by the addition of animal plasma to milk replacer. The colostrum replacement used in this study provided adequate IgG for newborn calves. Animal plasma was an acceptable source of protein but did not enhance growth or immunity under the conditions of this study.  相似文献

介绍以放大的盐析法从猪血中提取IgG。通过正交试验,结合721分光光度计吸光率测定与电泳分析,确定最佳工艺条件为:pH7.0~7.3,温度40℃,盐析时间1h,搅拌转速40r/min。盐析沉淀采用PBS液错流管渗析脱NH4 及部分杂质,再作超滤设备选型、工艺流程设计、工艺条件正交试验,获浓缩液,冷冻干燥,测得IgG(纯度≥95%)收得率1.16%,为工业化生产推广探索工艺及操作参数。  相似文献

Changes in the physicochemical composition of Murciano‐Granadina goat colostrum were studied with postpartum time and season. Samples that were taken immediately after partum showed the best characteristics, with the highest concentrations of proteins, solids, urea and immunoglobulin G (IgG). Samples obtained in the spring showed the highest concentration of total IgG at all postpartum times assayed. Exhaustive statistical analysis showed that the physicochemical composition of the colostrum was greatly affected by the postpartum time and only slightly affected by the season. The high protein and total IgG concentrations make goat colostrum an interesting product for uses beyond the traditional ones.  相似文献

The aim of this work was to develop an assay capable of detecting adulteration of high premium milk with milk from cheaper sources. An indirect, competitive ELISA was developed for the rapid detection of cows' milk in the milk of goat, sheep, and buffalo. The assay uses a monoclonal antibody produced against bovine IgG. This antibody recognizes a species-specific epitope on the heavy chain of both bovine IgG1 and IgG2. A peroxidase-conjugated anti-mouse IgG antibody was used to detect bound monoclonal antibody and subsequent enzymatic conversion of substrate resulted in clear differences in absorbance when assaying different mixtures of milks adulterated with cows' milk. Once optimized, the ELISA was found to be highly specific. Detection limits of the assay are 1.0 microg/mL of bovine IgG, or 0.1% (vol/vol) adulteration with cows' milk. The assay was highly reproducible (CV < 10%) and performed equally well when used to detect bovine IgG in mixtures with the 3 types of milk tested. The ELISA performance makes it suitable for development as a kit, for use in the field as a high throughput screening ELISA.  相似文献

选择速滑专业运动员7名,观察一次赛前训练对速滑运动员IgG,IgM,IgA及WBC,LY的影响。于安静时和训练后即刻及恢复24 h,分别采静脉血5 mL,用全血细胞分析仪测定WBC,LY;IgG,IgM,IgA用特定蛋白分析仪采用散射比浊法完成。并于训练后即刻采指尖微量血,快速测试血乳酸。结果表明,与安静值比较运动员训练后血WBC、LY出现明显增高;安静时和训练后IgG个体差异比较大,有增高、下降、不变的不同表现。与安静时比较IgM、IgA的平均水平在训练后没有出现规律性变化。  相似文献

目的探讨利用高密度介质固相抗人球蛋白法检测孕妇ABO血型系统IgG抗体效价的可行性。方法应用不完全抗体检测试剂盒(高密度介质固相抗人球蛋白法)检测40例O型血孕妇ABO血型系统IgG抗体效价,并与传统抗人球蛋白试管法进行对比。结果以IgG抗A(B)效价≥1:64定为阳性,两种方法的阳性检出率一致,均为50%(20/40);两种方法检测的抗体效价的几何均数差异无统计学意义(P>0.05);两种方法高度相关,r=0.9079(tr=13.36,P<0.001)。结论高密度介质固相抗人球蛋白法与传统抗人球蛋白试管法比较,操作简单,耗时短,结果准确,可应用于ABO血型系统IgG抗体效价的检测。  相似文献
