This study describes the first binding assay for glycine transporter 2 (GlyT2) following the concept of MS Binding Assays. The selective GlyT2 inhibitor Org25543 was employed as a reporter ligand and it was quantified with a highly sensitive and rapid LC-ESI-MS/MS method. Binding of Org25543 at GlyT2 was characterized in kinetic and saturation experiments with an off-rate of 7.07×10−3 s−1, an on-rate of 1.01×106 M−1 s−1, and an equilibrium dissociation constant of 7.45 nM. Furthermore, the inhibitory constants of 19 GlyT ligands were determined in competition experiments. The validity of the GlyT2 affinities determined with the binding assay was examined by a comparison with published inhibitory potencies from various functional assays. With the capability for affinity determination towards GlyT2 the developed MS Binding Assays provide the first tool for affinity profiling of potential ligands and it represents a valuable new alternative to functional assays addressing GlyT2.  相似文献

The investigation of radiation-absorption,chemical reaction,Hall and ion-slip impacts on unsteady MHD free convective laminar flow of an incompressible viscous,electrically conducting and heat generation/absorbing fluid enclosed with a semi-infinite porous plate within a rotating frame has been premeditated.The plate is assumed to be moving with a constant velocity in the direction of fluid movement.A uniform transverse magnetic field is applied at right angles to the porous surface,which is absorbing the fluid with a suction velocity changing with time.The non-dimensional governing equations for present inves-tigation are solved analytically making use of two term harmonic and non-harmonic functions.The graphical results of velocity,temperature and concentration distributions on the analytical solutions are displayed and discussed with reference to pertinent parameters.It is found that the velocity profiles decreased with an increasing in Hartmann number,rotation parameter,the Schmidt number,heat source parameter,while it increased due to an increase in permeability parameter,radiation-absorption param-eter,Hall and ion slip parameters.However,the temperature profile is an increasing function of radiation-absorption parameter,whereas an increase in chemical reaction parameter,the Schmidt num-ber Sc or frequency of oscillations decrease the temperature profile on cooling.Also,it is found that the concentration profile is decreased with an escalating in the Schmidt number or the chemical reaction parameter.  相似文献

In the present work, we performed immunochemical studies of LPS, especially the O-specific polysaccharide (O-PS) of Aeromonas veronii bv. sobria strain K133, which was isolated from the kidney of carp (Cyprinus carpio L.) during an outbreak of motile aeromonad infection/motile aeromonad septicemia (MAI/MAS) on a Polish fish farm. The structural characterization of the O-PS, which was obtained by mild acid degradation of the LPS, was performed with chemical methods, MALDI-TOF mass spectrometry, and 1H and 13C NMR spectroscopy. It was revealed that the O-PS has a unique composition of a linear tetrasaccharide repeating unit and contains a rarely occurring sugar 2,4-diamino-2,4,6-trideoxy-D-glucose (bacillosamine), which may determine the specificity of the serogroup. Western blotting and ELISA confirmed that A. veronii bv. sobria strain K133 belongs to the new serogroup PGO1, which is one of the most commonly represented immunotypes among carp and trout isolates of Aeromonas sp. in Polish aquacultures. Considering the increase in the MAI/MAS incidences and their impact on freshwater species, also with economic importance, and in the absence of an effective immunoprophylaxis, studies of the Aeromonas O-antigens are relevant in the light of epidemiological data and monitoring emergent pathogens representing unknown antigenic variants and serotypes.  相似文献

Microglial activity in the aging neuroimmune system is a central player in aging-related dysfunction. Aging alters microglial function via shifts in protein signaling cascades. These shifts can propagate neurodegenerative pathology. Therapeutics require a multifaceted approach to understand and address the stochastic nature of this process. Polyphenols offer one such means of rectifying age-related decline. Our group used mass spectrometry (MS) analysis to explicate the complex nature of these aging microglial pathways. In our first experiment, we compared primary microglia isolated from young and aged rats and identified 197 significantly differentially expressed proteins between these groups. Then, we performed bioinformatic analysis to explore differences in canonical signaling cascades related to microglial homeostasis and function with age. In a second experiment, we investigated changes to these pathways in aged animals after 30-day dietary supplementation with NT-020, which is a blend of polyphenols. We identified 144 differentially expressed proteins between the NT-020 group and the control diet group via MS analysis. Bioinformatic analysis predicted an NT-020 driven reversal in the upregulation of age-related canonical pathways that control inflammation, cellular metabolism, and proteostasis. Our results highlight salient aspects of microglial aging at the level of protein interactions and demonstrate a potential role of polyphenols as therapeutics for age-associated dysfunction.  相似文献

铬矿中砷的测定方法对完善铬矿检测方法和质量评价手段有重要意义。在0.5 g样品中加入2 g过氧化钠,于650 ℃熔融20 min,熔融物经盐酸酸化溶解,以10%盐酸为载流,15 g/L硼氢化钾溶液为还原剂,采用氢化物发生-原子荧光光谱法(HG-AFS)进行测定,建立了铬矿中砷测定的新方法。实验表明,砷质量浓度在0.025~100 μg/L范围内与荧光强度呈线性关系,相关系数为1.000 0,方法检出限为0.05 μg/g,定量限为0.17 μg/g。对产地为南非、土耳其、巴基斯坦等19个国家共2 649个铬矿样品的成分进行统计,发现铬矿主要成分为铬、铁、镁、铝和硅;干扰试验表明,这些样品中的共存元素均不干扰测定。按照实验方法对3个不同砷含量的铬矿样品进行精密度和加标回收试验,结果的相对标准偏差(RSD,n=11)为1.4%~6.0%,回收率为96%~109%。将实验方法应用于与铬矿基体近似的铁矿标准物质中砷的测定,测定值与认定值基本一致。  相似文献

准确、快速地测定含铁尘泥中有害元素,对含铁尘泥节能降废项目研究及综合利用有着重要的指导意义。实验针对含铁尘泥试样因含碳量高,导致常规酸溶解方法如果不增加除碳步骤则很难完全分解试样的问题,采用高温熔融法将试样制成熔融液后倾倒于稀酸中溶解的方法,解决了碳含量高难溶解的问题。采用四硼酸锂-碳酸锂混合熔剂在高温下将试样熔融,熔融液高温倾倒于稀酸中制成澄清溶液,由于骤冷,试样炸裂成微小玻璃珠,无迸溅,操作简单,使得后续溶解过程快速完成,从而建立了电感耦合等离子体原子发射光谱(ICP-AES)测定P、Pb、K、Na、Zn共5种有害元素的方法。按照实验方法测定含铁尘泥中P、Pb、K、Na、Zn,测定值的相对标准偏差(RSD,n=9)在1.4%~5.7%之间,加标回收率在96%~110%之间。研究结果表明方法能够满足含铁尘泥样品中5种元素的定量分析要求。  相似文献

常规粉末压片制样是一种简单、高效的绿色环保制样技术,但是应用于某些沉积物样品制备存在样片表面粗糙和粉末容易脱落的问题。实验采用高于常规的压力进行样品制备,建立了X射线荧光光谱法(XRF)分析海洋沉积物样品中包括硫、氧化钠、氧化镁、三氧化二铝、二氧化硅、五氧化二磷、氧化钾、氧化钙、二氧化钛、氧化锰、三氧化二铁、钴、镍、铜、锌、钒、铬、镓、铌、锆、钇、锶、铷、铅、钡、镧、钕和铪在内的28种主、微量组分的方法。探讨了压力为300 kN和1 600 kN时的制样效果,并尝试引入BP神经网络模型利用其非线性拟合能力校正主量组分的基体效应。结果表明,采用1 600 kN压力制备的样片,表面致密、光滑、不龟裂和不掉粉,制样重复性和测试精密度也有较大提高。以55个有证标准物质中17种组分的数据集为训练样本,建立了海洋沉积物样品中主、微量组分的遗传算法-BP神经网络预测模型。按照实验方法对各组分含量相对较低的实际样品连续测试12次,计算得方法的检出限在 0.63~634 μg/g之间;精密度试验结果表明,各组分测定值的相对标准偏差(RSD, n=7)为0.16%~25.1%。方法用于海洋沉积物实际样品分析,其分析结果与国标法的测定结果吻合,能够满足海洋沉积物样品中多种组分准确分析的要求。  相似文献

锑在现代工业体系中占有重要地位,快速准确测定锑矿中锑含量对锑矿的采选具有重要指导意义。云南某矿山锑矿以萤石为主要伴生矿,现有的盐酸、硝酸、氢氟酸和高氯酸混酸体系无法彻底分解样品,而采用碱熔处理流程长且引入大量基体,均无法满足准确快速测定的要求。选择硫酸-氢氟酸混酸体系分解样品,再用酒石酸-盐酸混合溶液加热浸取,锑完全溶解于溶液中,选择Sb 206.833{463}nm为分析谱线,使用电感耦合等离子体原子发射光谱法(ICP-AES)测定锑。结果表明,锑的质量浓度在5.0~80 μg/mL范围内与其发射强度呈良好的线性关系,校准曲线线性相关系数r=0.999 8;方法的检出限为0.012 μg/mL。按照实验方法对锑矿石成分分析标准物质和锑矿选矿流程样品中锑进行测定,结果的相对标准偏差(RSD,n=5)为0.71%~5.7%;标准物质的认定值和测定值相一致;锑矿选矿流程样品中锑的加标回收率为97%~102%,满足国家地质矿产行业标准DZ/T 0130—2006的要求。  相似文献

An analytical method using high resolution inductively coupled plasma mass spectrometry (HR-ICP-MS) for rapid simultaneous determination of 24 elements (Be, Mg, Al, Ti, V, Cr, Mn, Fe, Ni, Cu, Zn, Ga, Ge, As, Se, Mo, Ag, Cd, Sn, Sb, Ba, Pt, Au, and Pb) in high purity cobalt was described. Sample digestions were performed in closed microwave vessels using HNO3 and HCl. The matrix effects because of the presence of excess HCl and Co were evaluated. The usefulness of high mass resolution for overcoming some spectral interference was demonstrated. The optimum conditions for the deter- mination were tested and discussed. The standard addition method was employed for quantitative analysis. The detection limits were 0.016-1.50 μg?g?1, the recovery ratios were 92.2%-111.2%, and the RSD was less than 3.6%. The method was accurate, quick, and convenient. It was applied to the determination of trace impurities in high purity cobalt with satisfactory results.  相似文献
