This report described a method for simultaneous determination of cyanuric acid (CYA) and melamine (MEL) in dairy products. The samples were extracted by a mixture of acetonitrile/water and cleaned by graphite carbon/strong cation exchange (CARB/SCX) mixed-mode solid phase extraction column. The CYA and MEL were derivatized with bis(trimethylsily1)trifluoroacetamide (BSTFA) containing 1% Trimethylchlorosilane (TMCS), and quantified with the internal standards of 15N3-cyanuric acid and 13C3-melamine by GC–MS. The results indicated that CARB/SCX mixed solid phase extraction column could be used for sample pretreatment. The fortification recoveries were 80.8–101.5%, and the relative standard deviations (RSDs) were 3.6–7.9%. The detection limits of CYA and MEL were 0.025 mg kg−1 and 0.01 mg kg−1 respectively.  相似文献

Thirty strains of Listeria monocytogenes were screened for their pressure tolerance phenotype at 400 MPa for 2 min at 21 °C. The strains exhibited reductions ranging from 1.9 to 7.1log10 CFU/ml in tryptic soy broth with 6% yeast extract (TSBYE). The 3 most and the 3 least pressure-tolerant strains were further tested for their thermal resistance (based on their ability to survive at 55 °C), acid tolerance (based on their ability to survive in acidified TSBYE; pH 2.0) and for their nisin sensitivity. No correlation between pressure tolerance and heat, acid or nisin resistances was found. Nucleotide sequence analysis of the ctsR region in these 6 strains demonstrated that this gene codes for a CtsR protein with identical predicted amino acid sequences. The sequences of the 200-bp region located immediately upstream of the ctsR start codon of the different strains were virtually identical and it is therefore likely that differences in pressure tolerance are based on factors other than the stress gene regulator CtsR. The pressure sensitivity of a cocktail of the 2 most pressure-resistant strains and a cocktail of the 2 most-sensitive strains was investigated when the cocktails were inoculated into a real food system consisting of ground chicken meat. We demonstrated that the nature of the suspending substrate or the temperature did not change the expected pressure tolerance of the cocktails.  相似文献

The effect of different techniques for the isolation of Campylobacter from 198 samples of chicken giblets was studied. Four enrichment broths and two plating media were compared. A protocol of enrichment in Preston or Doyle and Roman's broth followed by plating on Preston agar is recommended for routine use. The duration of incubation of enrichment media was shown to have a significant effect on the number of Campylobacter isolates obtained, 48 h being statistically superior to either 24h or 72h for media incubated at 42°C. Of the 198 samples examined, 177 were found to contain Campylobacter by at least one of the procedures used.  相似文献

Previous studies demonstrated a hyperspectral imaging system has a potential for poultry fecal contaminant detection by measuring reflectance intensity. The simple image ratio at 565 and 517 nm images with optimal thresholding was able to detect fecal contaminants on broiler carcasses with high accuracy. However, differentiating false positives from real contaminants, especially cecal feces were challenging. Further image processing such as textural analysis in the spatial domain was able to reduce false positive errors. In this study, textural analysis of hyperspectral images was conducted to improve detection accuracy by reducing false positives. Specifically, textural analysis with co-occurrence matrix of hyperspectral images performed well to identify “true” contamination. In addition, co-occurrence matrix textural features including average, variance, entropy, contrast, correlation, moment of poultry hyperspectral images were investigated for selecting optimal features to represent contamination. Image pre-processing with co-occurrence textural analysis, specifically mean of co-occurrence textural feature from the matrix (0° angle and distance equals to one) followed by image ratio was able to improve fecal detection accuracy without additional optical filters that increase cost for system hardware of multispectral imaging system for on-line application. Trace amounts (0.004–0.55 ng) of beryllium (Be) in a dried bovine liver sample (20 mg) can be accurately determined by graphite-furnace atomic absorption spectrophotometry (GFAAS) after treating with microwave digestion (HNO3/H2O2) at 85 °C for 10 min and using acetylacetone as a chelating agent in the presence of an acetate buffer (pH 6.0). The method detection limit (MDL, 3σ) for Be was found to be 0.18 ng g−1 and the limit of quantification (LOQ, 10σ) was found to be 0.60 ng g−1; the calibration graph was linear up to 27 ng g−1. The Be contents measured in four liver and four muscle samples (BCR CRM-185R bovine liver, BCR CRM-384 pork muscle, and six samples collected in Kaohsiung, Taiwan, ROC) were between 2.3 and 4.7 ng g−1. Good spiked recoveries (96.0–103.0%) were obtained for these eight samples with a relative standard deviation (RSD, n = 3) ?3.0%. The method could be applied to measurements of Be in livers and muscles of poultry and livestock.  相似文献

Different land uses directly affect the characteristics of a river basin and influence the aquatic biota and ecosystem processes. This study aimed to analyse the community structure and composition of aquatic invertebrates and the role of these organisms in the process of leaf litter breakdown in streams with different land uses. The study was conducted from September to December 2013 in five streams in the Neotropical region. At each stream, we placed 18 bags containing litter for colonization by aquatic invertebrates, and the bags were collected at different exposure times (5, 30, 45, 60, 75 and 90 days). We registered spatial differences in the aquatic invertebrate community structure and composition. There were no significant differences in the activity of invertebrates in the leaf litter breakdown process among streams with different land uses. However, the variability in mass decay rate was lower for the reference stream. This result may have been influenced by habitat quality, availability of organic matter and the structure and composition of benthic community present in the reference stream, which differs significantly among locations with different types of land use. The results of this study shows that human activities, particularly agriculture and urbanization, modify the structure and composition of the benthic community and acts on ecosystem processes, especially in the variability of the processing of allochthonous material invertebrates. However, we reject the hypothesis that land use negatively influences the decomposition of litter, measured by weight loss. Copyright © 2016 John Wiley & Sons, Ltd.  相似文献

Atmospheric CO2 concentrations have increased dramatically over the last century and continuing increases are expected to have significant, though currently unpredictable, effects on ecosystems. One important process that may be affected by elevated CO2 is leaf litter decomposition. We investigated the interactions among atmospheric CO2, herbivory, and litter quality within a scrub oak community at the Kennedy Space Center, Florida. Leaf litter chemistry in 16 plots of open-top chambers was followed for 3 years; eight were exposed to ambient levels of CO2, and eight were exposed to elevated levels of CO2 (ambient + 350 ppmV). We focused on three dominant oak species, Quercus geminata, Quercus myrtifolia, and Quercus chapmanii. Condensed tannin concentrations in oak leaf litter were higher under elevated CO2. Litter chemistry differed among all plant species except for condensed tannins. Phenolic concentrations were lower, whereas lignin concentrations and lignin/nitrogen ratios were higher in herbivore-damaged litter independent of CO2 concentration. However, changes in litter chemistry from year to year were far larger than effects of CO2 or insect damage, suggesting that these may have only minor effects on litter decomposition.  相似文献

Stable isotope and elemental analysis, together with statistical processing of the resultant data has been used to determine the geographical origin of poultry and hence provide a means to verify poultry labels originating from major producing countries/regions. Multivariate statistical analysis has demonstrated that 18 variables, including carbon, hydrogen and nitrogen stable isotope ratios and elemental concentrations of magnesium, thallium, rubidium and molybdenum, are important parameters in poultry origin determination. Using cross-validated discriminant analysis 88.3% of poultry geographical origins were correctly classified (n = 339). Individual correct-classification rates were as follows; China, 100% (n = 36); Brazil, 94.1% (n = 101); Europe 92% (n = 87); Chile 82.6% (n = 46); Thailand, 70.3% (n = 46) and Argentina 50% (n = 10). The main identification errors were associated with miss-classification of Argentinean samples with those originating from Chile and Thailand. Carbon stable isotope ratios of chicken meat indicate the quantity of maize in the diet and this leads to useful discrimination between a large proportion of European poultry and poultry reared in locations such as South America, Thailand and China where maize feeding predominates. The use of poultry carbon isotope values as a simple ‘screening’ parameter to differentiate European poultry meat from other major importers is not as reliable as for the differentiation of European and South American beef. However carbon isotope ratios will be useful in most instances to corroborate suspicion of mislabelling of non corn-fed European poultry. The stable isotopes of hydrogen and oxygen in chicken meat change in a similar way to surface waters around the globe. Our findings support the hypothesis that the global isotopic variation of stable isotopes in drinking water and feed are transferred into animal tissue and can be used to help establish an animal's geographic origin. This is a significant finding and mirrors our observations for beef skeletal muscle δ2H ‰ and δ18O ‰ values. These systematic variations can be exploited to give a ‘low-resolution’ indication of an animal's geographic origin (e.g. Northern Europe versus the tropics).  相似文献

OBJECTIVE: To describe the waste produced by and environmental implications of individual cigarette consumption (filter tips, packages, and cartons) and tobacco manufacturing. STUDY SELECTION: All available articles and reports published since 1970 related to cigarette consumption and production waste were reviewed. DATA SOURCES: Global cigarette consumption data were used to estimate cigarette butt and packaging waste quantities. Data from the Center for Marine Conservation's International Coastal Cleanup Project were used to describe some environmental impacts of tobacco-related trash. Data from the United States Environmental Protection Agency's (EPA's) Toxics Release Inventory and reported global cigarette consumption totals were used to estimate waste production from cigarette manufacturing. DATA EXTRACTION AND SYNTHESIS: In 1995, an estimated 5.535 trillion cigarettes (27,675 million cartons and 276,753 million packages) were sold by the tobacco industry globally. Some of the wastes from these products were properly deposited, but a large amount of tobacco consumption waste ends up in the environment. Some is recovered during environmental clean-up days. For the past eight years (1990-1997), cigarette butts have been the leading item found during the International Coastal Cleanup Project; they accounted for 19.1% of all items collected in 1997. The tobacco manufacturing process produces liquid, solid, and airborne waste. Among those wastes, some materials, including nicotine, are designated by the EPA as Toxics Release Inventory (TRI) chemicals. These are possible environmental health hazards. In 1995, the global tobacco industry produced an estimated 2262 million kilograms of manufacturing waste and 209 million kilograms of chemical waste. In addition, total nicotine waste produced in the manufacture of reduced nicotine cigarettes was estimated at 300 million kilograms. CONCLUSIONS: Laws against littering relative to cigarette butts could be better enforced. Additional taxes might be levied on cigarette products that would then be directed to environmental clean-up efforts. The tobacco industry should improve the biodegradability of filters, reduce packaging waste, and educate its customers. Worksites and public buildings should be encouraged or required to supply appropriate disposal mechanisms at all building entrances. Public awareness campaigns about the magnitude and prevention of cigarette consumption waste could be developed through partnerships among environmental groups, health organisations, and environmental protection agencies. Tobacco production waste should be a source of concern and regulation by governments throughout the world; it contains numerous chemicals which may be considered health hazards, not the least of which is nicotine produced in the manufacture of low-nicotine cigarettes.  相似文献

This paper presents a methodology for the estimation of the contribution of direct energy use to the greenhouse gases emissions of cattle, pig and poultry breeding in Cyprus. The energy consumption was estimated using the factors of 2034 MJ/cow, 2182 MJ/sow and 0.002797 MJ/bird. The greenhouse gases emissions for each animal species and energy source were estimated using emission factor of each greenhouse gas according to fuel type as proposed by the IPCC 2006 guidelines and for electricity according to national verified data from the Electricity Authority of Cyprus. Livestock breeding in Cyprus consumes electricity, diesel oil and LPG. The results obtained, show that the emissions from energy use in livestock breeding contribute 16% to the total agricultural energy emissions. Agricultural energy emissions contribute 0.7% to the total energy greenhouse gases (GHG) emissions. The three species of animal considered contribute 3% to their total livestock breeding emissions when compared with enteric fermentation and manure management, of which 2.6% is CO2. These results agree with the findings in available literature. The contribution of direct energy use in the greenhouse gases emissions of livestock breeding could be further examined with the influence of anaerobic digestion to the emissions.  相似文献
