The staining of intracellular antigenic sites in postembedded samples is a challenging problem. Deterioration of antigenicity and limited antibody accessibility to the antigen are commonly encountered on account of processing steps. In this study preservation of the antigen was achieved by fixing the tissues with mild fixatives, performing partial dehydration, and embedding in a low crosslinked hydrophilic acrylic resin, LR-White. Permeabilization of cell membranes with Triton X-100 is well documented but can affect some antigen conformations. We tested the effect of Triton X-100 on the ED1 antigen present in the lysosomal membrane of the macrophage in cell culture. The ED1 antigen in the lysosome was resistant to extraction by Triton X-100. Interestingly pretreating the LR-White sections of macrophage pellets with Triton X-100 improved the staining intensity of ED1. The most intense and clear specific fluorescent staining was observed when sections were pretreated with 0.2% Triton X-100 for 2 min. Longer exposure of sections to 0.2% Triton or 2 min exposure to 2% Triton lead to reduced ED1 labeling. SEM observations indicated that the detergent extracted a component from the cells and not the resin and was determined to be lipid. This novel technique could be applied in many research areas where postembedding fluorescent immunolabeling with higher labeling intensity is desired.  相似文献

Alcoholic patients commonly experience cognitive decline. It is postulated that cognitive dysfunction is caused by an alcohol-induced region-selective brain damage, particularly to the prefrontal region, and grey and white matter may be affected differently. We used a proteomics-based approach to compare protein expression profiles of the dorsolateral prefrontal cortex (Brodmann area 9 (BA9)) from human alcoholic and healthy control brains. Changes in the relative expression of 110 protein 'spots' were identified in the BA9 grey matter, of which 54 were identified as 44 different proteins. In our recent article, 60 protein spots were differentially expressed in the BA9 white matter and 18 of these were identified (Alexander-Kaufman, K., James, G., Sheedy, D., Harper, C., Matsumoto, I., Mol. Psychiatry 2006, 11, 56-65). Additional BA9 white matter proteins are identified here and discussed in conjunction to our grey matter results. Thiamine-dependent enzymes transketolase and pyruvate dehydrogenase (E1β ubunit) were among the proteins identified. To our knowledge, this is the first time a disruption in thiamine-dependent enzymes has been demonstrated in the brains of 'neurologically uncomplicated' alcoholics. By identifying protein expression changes in prefrontal grey and white matter separately, hypotheses may draw upon more mechanistic explanations as to how alcoholism causes the structural alterations associated with alcohol-related brain damage and cognitive dysfunction.  相似文献

比较AWGN信道与瑞利信道的误码性能,研究瑞利信道下分集支数与信息传输可靠性之间的关系。推导线性分组码在瑞利信道下误码率上界的理论表达式,得到分集增益与其最小汉明距离的关系。基于蒙特卡洛法对误码性能进行仿真,并与理论误码率上界进行比较,结果表明线性分组码在瑞利信道下获得的分集增益等于其最小码距。  相似文献

This work deals with the experimental tracking of a dispersion of very fine silica particles in PDMS using a new, easily implemented methodology. The dispersion experiments are carried out in an 8-l capacity mixer equipped with a torquemeter. Using a Couette analogy, the torque-rotational speed data can be converted into process viscosity-effective shear rate data. Simultaneously, the Particle Size Distribution (PSD) evolution is followed through Laser Light Scattering measurements on diluted samples. The PSD evolution allowed us to interpret the dispersion process in terms of different mechanisms, such as breaking-up and erosion of aggregates, as well as, in some cases, re-agglomeration (or flocculation) of particles.  相似文献

Three types of silica particles modified with vinyl groups were obtained: (i) xerogel formed by hydrolytic polycondensation of the mixture of tetramethoxysilane (TMOS) and 1,1,1,7−tetramethoxy-3,5,7-trimethyl-3,5,7-trivinyltetrasiloxane, (ii) mesoporous silica obtained from the same precursors in the presence of the cetyltrimethylammonium bromide (CTAB), and (iii) commercial Fluka silica gel 60A with a vinyltriethoxysilane-treated surface. Vinyl groups on these silica materials were transformed into silyl chloride by hydrosilylation with HMe2SiCl. These groups were used to graft living polysiloxane that was synthesized by anionic ring-opening polymerization of 2,4,6-tri(3-chloropropyl)-2,4,6-trimethylcyclotrisiloxane and initiated by BuLi. Chloropropyl groups on the grafted polymer were used to quaternize N,N-dimethyl-n-octylamine. Silica particles with grafted polysiloxane having quaternary ammonium salt (QAS) groups pendant to polymer chains were obtained. Silica material with QAS groups directly attached to the surface were generated by the action of N,N-dimethyl-n-octylamine on particles obtained by the sol–gel process involving tetraethoxysilane (TEOS) with 3-chloropropyltriethoxysilane. The bacteriocidal properties of all these materials were tested in water suspension against five representative strains for Gram-positive and Gram-negative bacteria. Some of the silica–polysiloxane hybrid materials have good antibacterial properties against Gram-positive strains, but not as good as the non-tethered QAS-substituted polysiloxane in water solution. The QAS groups that are directly bonded to the silica material surface are inactive.  相似文献

In this paper, a representative unit volume finite element (RUV FE) model was employed to simulate thermoforming process of carbon fabric-polymeric foam sandwich structures. Thermoforming simulations, which capture crimp angles and amplitude changes of carbon fabric with respect to different operational pressure, were conducted with the help of RUV FE model. Micro-deformation of tow structures after thermoforming was rigorously reflected in the second stage FE model to determine the in-plane directional stiffness of skin parts. Tensile tests were carried out to investigate mechanical properties of skin parts in sandwich structures for the potential usage of automotives, electronic housings or the satellite structure design.  相似文献

Using cheap waterglass as silica source, silica aerogels were synthesized via a novel fast ambient drying by using an ethanol/trimethylchlorosilane (TMCS)/Heptane solution for modification of the wet gel. One-step solvent exchange and surface modification were simultaneously progressed by immersing the hydrogel in EtOH/TMCS/Heptane solution, in which TMCS reacting with pore water and Si–OH group on the surface of the gel, with ethanol and heptane helping to decrease the rate of TMCS reacting with pore water and extrude water from gel pores. The synthesized silica aerogel was a light and crack-free solid, with the density of 0.128–0.136 g/cm3 and 93.8–94.2% porosity. The microstructure, morphology and properties of the aerogels were studied by FTIR, SEM, TEM and BET measurement. The results indicate that silica aerogels exhibit a sponge structure with uniform nano-particle and pores size distribution. The specific surface areas of silica aerogels are 559–618 m2/g. And there is an obvious Si–CH3 group on the surface of the silica aerogel.  相似文献

Three-wavelength white organic light-emitting diodes (WOLEDs) were fabricated using two doped layers, which were obtained by separating the recombination zones into three emitter layers. A sky blue emission originated from the 4,4′-bis(2,2′-diphenylethen-1-yl)biphenyl (DPVBi) layer. A green emission originated from a tris(8-quinolinolato)aluminum (III) (Alq3) host doped with a green fluorescent 10-(2-benzothiazolyl)-1,1,7,7-tetramethyl-2,3,6,7-tetrahydro-1H,5H,11H-[1]benzopyrano [6,7,8-ij]-quinolizin-11-one (C545T) dye. An orange emission was obtained from the N,N′-bis(1-naphthyl)-N,N′-diphenyl-1,1′-biphenyl-4,4′-diamine (NPB) host doped with a red fluorescent dye, 4-(dicyanomethylene)-2-tert-butyl-6-(1,1,7,7-tetramethyljulolidyl-9-enyl)-4H-pyran (DCJTB). A white light resulted from the partial excitations of these three emitter layers by controlling the layer thickness and concentration of the fluorescent dyes in each emissive layer simultaneously. The electroluminescent spectrum of the device was not sensitive to the driving voltage of the device. The white light device showed a maximum luminance of approximately 53,000 cd/m2. The external quantum and power efficiency at a luminance of approximately 100 cd/m2 were 2.62% and 3.04 lm/W, respectively.  相似文献
