BACKGROUND: Nisin is a commercially available bacteriocin produced by Lactococcus lactis ATCC 11454 and used as a natural agent in the biopreservation of food. In the current investigation, milk whey, a byproduct from dairy industries was used as a fermentation substrate for the production of nisin. Lactococcus lactis ATCC 11454 was developed in a rotary shaker (30 °C/36 h/100 rpm) using two different media with milk whey (i) without filtration, pH 6.8, adjusted with NaOH 2 mol L?1 and without pH adjustment, both autoclaved at 121 °C for 30 min, and (ii) filtrated (1.20 µm and 0.22 µm membrane filter). These cultures were transferred five times using 5 mL aliquots of broth culture for every new volume of the respective media. RESULTS: The results showed that culture media composed of milk whey without filtration supplied L. lactis its adaptation needs better than filtrated milk whey. Nisin titers, in milk whey without filtration (pH adjusted), was 11120.13 mg L?1 in the second transfer, and up to 1628‐fold higher than the filtrated milk whey, 6.83 mg.L?1 obtained in the firstt transfer. CONCLUSIONS: Biological processing of milk byproducts (milk whey) can be considered a profitable alternative, generating high‐value bioproducts and contributing to decreasing river disposals by dairy industries. Saccharomyces cerevisiae present in common Baker’s yeast was used in a microbial fuel cell in which glucose was the carbon source. Methylene blue was used as the electronophore in the anode compartment, while potassium ferricyanide and methylene blue were tested as electron acceptors in the cathode compartment. Microbes in a mediator-free environment were used as the control. The experiment was performed in both open and closed circuit configurations under different loads ranging from 100 kΩ to 400Ω. The eukaryotic S. cerevisiae-based fuel cell showed improved performance when methylene blue and ferricyanide were used as electron mediators, rendering a maximum power generation of 146.71±7.7 mW/m3. The fuel cell generated a maximum open circuit voltage of 383.6±1.5 mV and recorded a maximum efficiency of 28±1.8 % under 100 kΩ of external load. 相似文献

In the work presented herein, the potential use of La4Sr8Ti12−xFexO38−δ (LSTF) materials as electrodes for a new concept of solid oxide fuel cells, symmetrical fuel cells (SFCs), is considered. Such fuel cells use simultaneously the same material as anode and cathode, which notably simplifies the assembly and further maintenance of the cells. Therefore, we search for materials showing high conductivity in a wide range of oxygen partial pressures in addition to certain degree of catalytic activity for the oxidation of the fuel and reduction of the oxidant, respectively. The preliminary electrochemical experiments performed reveal that the overall conductivity increases notably upon Fe substitution, being the main contribution electronic n-type. The fuel cell tests indicate that LSTF composites with YSZ and CeO2 perform reasonably well under H2 conditions, although the performance in methane is rather modest and require further optimisation. 相似文献

A new optimization model is discussed for the design of cellular manufacturing systems. It is based on an integer programming formulation that updates some other models by eliminating redundant machine assignment and cost coefficients dependent on cell configuration. To reduce computational burdens, a simplified integer programming model and a decomposition algorithm are proposed. There are two aspects to cell formation in flexible manufacturing systems, cell sizing or deciding on the optimum number of machines to be allocated to each cell, and then allocation of specific machines to each cell. Although the latter problem has been investigated extensively there is a paucity of published work on the former. A series of experiments have been conducted using a recombinant baculovirus/insect cell expression system (Bm5/Bm5.NPV.CAT) to establish the optimum temperature for both cell growth and virus infection. Bm5 cell growth was found to be limited at temperatures below 22°C and ceased completely at temperatures above 34°C. In the range between 24 and 28°C, final cell densities always reached 96% of the highest achievable viable cell density. The shortest population doubling time was obtained at 28°C. Overall, a consistent increase in metabolism with increasing temperatures was observed. During the infection/viral replication phase, an increase in the temperature from 25 to 31°C resulted in a faster decrease in viable cell density and an earlier production of chloramphenicol acetyltransferase (CAT). Furthermore, protein yield at temperatures above 28°C was significantly reduced. Overall, the best temperature for the infection phase for the Bm5/Bm5.NPV expression system was found to be 25°C when the cells are cultured in serum free media. 相似文献

Various aspects on probabilistic modelling of cleavage fracture are discussed. The investigation involves consideration of
a unit cell with an explicitly modelled void. The results from this model are compared with results for the case when the
void content is accounted for in the sense of a Gurson-Tvergaard law. It is found that explicit modelling of the void can
give substantially different results for the fracture probability. The effect depends on the exponent in the assumed Weibull
distribution, the threshold stress, the constraint conditions and the hardening of the material.
