Visual evaluation experiments of color discrimination threshold and suprathreshold color‐difference comparison were carried out using CRT colors based on the psychophysical methods of interleaved staircase and constant stimuli, respectively. A large set of experimental data was generated ranged from threshold to large suprathreshold color difference at the five CIE color centers. The visual data were analyzed in detail for every observer at each visual scale to show the effect of color‐difference magnitude on the observer precision. The chromaticity ellipses from this study were compared with four previous published data, of CRT colors by Cui and Luo, and of surface colors by RIT‐DuPont, Cheung and Rigg, and Guan and Luo, to report the reproducibility of this kind of experiment using CRT colors and the variations between CRT and surface data, respectively. The present threshold data were also compared against the different suprathreshold data to show the effect of color‐difference scales. The visual results were further used to test the three advance color‐difference formulae, CMC, CIE94, and CIEDE2000, together with the basic CIELAB equation. In their original forms or with optimized KL values, the CIEDE2000 outperformed others, followed by CMC, and with the CIELAB and CIE94 the poorest for predicting the combined dataset of all color centers in the present study. © 2005 Wiley Periodicals, Inc. Fluorescent speckle microscopy (FSM) uses low levels of fluorescent proteins to create fluorescent speckles on cytoskeletal polymers in high‐resolution fluorescence images of living cells. The dynamics of speckles over time encode subunit turnover and motion of the cytoskeletal polymers. We sought to improve on current FSM technology by first expanding it to study the dynamics of a non‐polymeric macromolecular assembly, using focal adhesions as a test case, and second, to exploit for FSM the high contrast afforded by total internal reflection fluorescence microscopy (TIR‐FM). Here, we first demonstrate that low levels of expression of a green fluorescent protein (GFP) conjugate of the focal adhesion protein, vinculin, results in clusters of fluorescent vinculin speckles on the ventral cell surface, which by immunofluorescence labelling of total vinculin correspond to sparse labelling of dense focal adhesion structures. This demonstrates that the FSM principle can be applied to study focal adhesions. We then use both GFP‐vinculin expression and microinjected fluorescently labelled purified actin to compare quantitatively the speckle signal in FSM images of focal adhesions and the actin cytoskeleton in living cells by TIR‐FM and wide‐field epifluorescence microscopy. We use quantitative FSM image analysis software to define two new parameters for analysing FSM signal features that we can extract automatically: speckle modulation and speckle detectability. Our analysis shows that TIR‐FSM affords major improvements in these parameters compared with wide‐field epifluorescence FSM. Finally, we find that use of a crippled eukaryotic expression promoter for driving low‐level GFP‐fusion protein expression is a useful tool for FSM imaging. When used in time‐lapse mode, TIR‐FSM of actin and GFP‐conjugated focal adhesion proteins will allow quantification of molecular dynamics within interesting macromolecular assemblies at the ventral surface of living cells.  相似文献

In executing tasks involving intelligent information processing, the human brain performs better than the digital computer. The human brain derives its power from a large number [O(1011)] of neurons which are interconnected by a dense interconnection network [O(105) connections per neuron]. Artificial neural network (ANN) paradigms adopt the structure of the brain to try to emulate the intelligent information processing methods of the brain. ANN techniques are being employed to solve problems in areas such as pattern recognition, and robotic processing. Simulation of ANNs involves implementation of large number of neurons and a massive interconnection network. In this paper, we discuss various simulation models of ANNs and their implementation on distributed memory systems. Our investigations reveal that communication-efficient networks of distributed memory systems perform better than other topologies in implementing ANNs.  相似文献

In this work, optimal shaft shapes for flow in the annular space between a rotating shaft with axially-periodic radius and a fixed coaxial outer circular cylinder, are investigated. Axisymmetric steady flows in this geometry are determined by solving the full Navier-Stokes equations in the actual domain. A measure of the flow field, a weighted convex combination of the volume averaged square of the L2-norm of the velocity and vorticity vectors, is employed. It has been demonstrated that boundary shape can be used to influence the characteristics of the flow field, such as its velocity component distribution, kinetic energy, or even vorticity. This ability to influence flow fields through boundary shape may be employed to improve microfluidic mixing or, possibly, to minimize shear in biological applications.  相似文献

In this paper the results of thermally stimulated discharge current (TSDC) and dielectric constant for 40PbO-60Bi2O3 glass thermoelectrets are presented. Measurements of TSDC and dielectric constant, ǵe, have been carried out in the temperature range 30–300°C. The thermoelectrets were prepared at different polarizing fields. The various observed peaks in the thermograms are discussed on the basis of space charge polarization. The trap energy is evaluated from the Garlick-Gibson plot of initial rise method. Similarly other parameters such as relaxation time, charge release etc are evaluated.  相似文献
