蛙皮素受体亚型-3在肺疾病中的研究进展

蛙皮素受体亚型-3（BRS-3）属G蛋白偶联受体,至今未发现天然配体。BRS-3具有广泛的生物学作用,可调控神经内分泌功能、能量代谢及细胞生长增殖。在啮齿类动物及哺乳动物体内都有分布,大多数组织表达水平很低（包括正常肺组织）,但在某些肺部疾病及胎肺中表达增加。就BRS-3受体特点、激活后信号转导机制及其与肺肿瘤、肺发育、慢性炎症性肺病的联系作一综述。     

Kidney injury and cell therapy: Preclinical study

The aim of this study is to show histological and immunofluorescence analysis of renal parenchyma of agoutis affected by gentamicin‐induced renal disease after the infusion of bone marrow mononuclear cells (BMMC) stained with Hoechst®. Nine agouti's males were divided into three groups: Test group (TG): renal disease by gentamicin induced (n = 3), cell therapy group (CTG): renal disease by gentamicin induced and BMMC infusion (n = 3), and control group (CG): nonrenal disease and BMMC infusion (n = 3). TG and CTG were submitted to the protocol of renal disease induction using weekly application of gentamicin sulfate for 4 months. CG and CTG received a 1 × 108 BMMC stained with Hoechst and were euthanized for kidney examination 21 days after BMMC injection and samples were collected for histology and immunofluorescence analysis. Histological analysis demonstrated typical interstitial lesions in kidney similarly to human disease, as tubular necrosis, glomerular destruction, atrophy tubular, fibrotic areas, and collagen deposition. We conclude that histological analysis suggest a positive application of agouti's as a model for a gentamicin inducing of kidney disease, beyond the immunofluorescence analysis suggest a significant migration of BMMC to sites of renal injury in CTG. 阿科特素体外处理乳腺癌细胞的基因表达通路分析

近年来有研究表明黑升麻提取物阿科特素具有抗肿瘤活性。但其分子机制仍然不够清楚。研究用基因集富集分析的方法分析了20μg,／mL和40μg／mL两种不同质量浓度的阿科特素处理乳腺癌细胞MDA—MB--4536h或24h后的基因表达谱,获取处理后的乳腺癌细胞中的基因集富集情况．结果表明：处理6h的乳腺癌细胞中没有获得有统计学意义的基因集,而在处理24h的乳腺癌细胞中与细胞分裂、细胞周期等相关的生物学通路则表达下调,40Ixg／mL阿科特素处理还可以使得乳腺癌细胞中大分子降解相关通路表达上调．由于阿科特素可以促进细胞凋亡并抑制细胞周期,为其今后用于乳腺癌的预防和治疗的可能性提供了良好的理论依据．     

一个医学数据集上的决策树分类研究

人乳头瘤病毒(HPV)是导致宫颈癌的一大要素。利用Weka这个数据挖掘平台,对中国肿瘤防治数据库山西子宫颈癌医学数据集进行C4.5决策树分类研究,从决策树中提取了一些比较有意义的规则,发现一些容易导致感染HPV病毒的因素。     

Optimistic Personality and Psychosocial Well-Being During Treatment Predict Psychosocial Well-Being Among Long-Term Survivors of Breast Cancer.

In considering well-being among survivors of life-threatening illnesses such as breast cancer, 2 important questions are whether there is continuity between initial adjustment and longer term adjustment and what role personality plays in long-term adjustment. In this research, a sample of 163 early stage breast cancer patients whose psychosocial adjustment was first assessed during the year after surgery completed the same measures 5-13 years after surgery. Initial reports of well-being were relatively strong predictors of follow-up well-being on the same measures. Initial optimism and marital status also predicted follow-up adjustment, even controlling for earlier adjustment, which exerted a substantial unique effect in multivariate analyses. In contrast, initial medical variables played virtually no predictive role. There is substantial continuity of subjective well-being across many years among survivors of breast cancer, rooted partly in personality and social connection. 抗乳腺癌单克隆抗体6C6和人-鼠嵌合抗体6C6CHI的放射性标记及动物实验研究

为了研究99Tcm标记的抗乳腺癌单抗6C6及嵌合抗体6C6CHI在动物体内的生物学分布及在荷瘤裸鼠肿瘤中的摄取情况,采用改进的Schwartz法进行99Tcm标记抗体。标记后的抗体由小鼠尾静脉注射,观察不同时间的小鼠血液清除、全身清除以及两种99Tcm标记的抗体在荷人乳腺癌MCF7细胞的裸鼠中肿瘤的摄取情况,并在γ相机下于注射后不同时间显像。结果表明,99Tcm标记两种抗体的标记率均大于90％,标记抗体的免疫活性大于80％。99Tcm-6C6的全身清除半衰期为4.1 h,在血液中的半衰期分别为Tα=0.55 h、Tβ=12.42 h;99Tcm-6C6CHI的全身清除半衰期为4.28h,在血液中半衰期分别为Tα=0.98h、Tβ=12.42h。在荷人乳腺癌裸鼠中的体内分布结果是:在注射99Tcm-6C6后23h肿瘤和血的％ID·g-1分别为7.42±0.85和5.67±1.44。除肾外,T/NT(肿瘤/非肿瘤)比值均大于1。在注射99Tcm-6C6CHI后23h肿瘤和血的％ID·g-1分别为4.23±0.64和6.97±0.23,除瘤/血、瘤/肾比值小于1外,其余组织的T/NT均大于1。荷瘤鼠γ显像结果显示:在注射标记抗体后24h,肿瘤显像清晰,除肾外其它脏器无异常浓聚。抗乳腺癌单抗6C6在肿瘤中有较好的特异性,而人源化抗体6C6-CHI的肿瘤摄取稍低于6C6抗体,且血中放射性比6C6抗体高,但肿瘤显像清晰,而其它脏器的T/NT比值大于1,     

栅列藻蛋白质提取液促进人子宫颈癌细胞的生长研究

本文采用ＭＴＴ［３（４，５ｄｉｍｅｔｈｙｌｔｈｉａｚｏｌ－２ｙｌ）－２，５ｄｉｐｈｅｎｙｌｔｅｔｒａｚｏｌｉｕｍｂｒｏｍｉｄｅ］快速微量比色技术发现斜生栅列藻（Ｓｃｅｎｅｄｅｓｍｕｓｏｂｌｉｑｕｕｓ）蛋白质提取液可促进动物细胞的生长．在０．５％胎牛血清（ＦＢＳ）的ＲＰＭＩ１６４０培养液中，加入９５μｇ／ｍｌ栅列藻提取液可使人子宫颈癌细胞（ＨｅＬａｃｅｌ）比对照组细胞增殖３倍左右．这种刺激动物细胞生长的活性物质分子量在３５００－８０００道尔顿（ｄａｌｔｏｎｓ）之间，经胰蛋白酶处理或高温处理后均失去活性     

Mismatches in Social Support and Psychosocial Adjustment to Breast Cancer.

Emotional support is known to provide psychosocial benefits for women with breast cancer, but women can experience a mismatch between support that is wanted and support that is received from their personal supporter. The role of wanted and unwanted support in psychosocial adjustment was examined in 79 women recovering from breast cancer. Four distinct patterns of desired support actions were found using cluster analysis. Patterns of wanted support were not related to better or worse psychosocial adjustment. However, a misalignment of support between the provider and the receiver significantly influenced psychosocial adjustment, and unwanted but received support (support commission) was uniquely associated with poor psychosocial adjustment. Clinical interventions using the support instrument could help match support providers' actions to receivers' preferences. (PsycINFO Database Record (c) 2010 APA, all rights reserved)     

Written Emotional Disclosure Buffers the Effects of Social Constraints on Distress Among Cancer Patients.

The aims of the present study were to examine whether written emotional disclosure would reduce distress among cancer patients and whether it would buffer the effects of high levels of social constraint (negative social responses to patients' expressions of emotion regarding their cancer) on distress. Cancer patients (N=104) were randomly assigned to write about their emotions regarding their cancer 20 min a day for 3 days or to write about a nonemotional topic. They completed questionnaires at baseline and 6 months postintervention. Results showed that written disclosure buffered the effects of social constraints on stress at the 6-month follow-up and that avoidance partly mediated these effects. Psychological Response to Test Results in an Ovarian Cancer Screening Program: A Prospective, Longitudinal Study.

To identify the psychological impact of receipt of an abnormal yet benign screening test result, the authors examined the response to a transvaginal ultrasound screening (TVS) test for ovarian cancer (OC) in asymptomatic women (N=540) undergoing an initial TVS screening test. Interviews were conducted prior to undergoing TVS screening and at 2 weeks and 4 months following this baseline. Women receiving an abnormal yet benign TVS test result (n=33) reported elevated OC-specific, but not general, distress at 2-week follow-up. Distress returned to baseline levels at 4-month follow-up. Consistent with the monitoring process and cognitive-social health information processing models, response to an abnormal TVS result was moderated by a monitoring coping style, low optimism, and a family history of OC.
