以放牧和舍饲两种饲养条件下12 月龄苏尼特羊股二头肌为实验材料,利用酶联免疫吸附测定法和实时荧光定量聚合酶链式反应法对肌内脂肪沉积动态平衡中脂肪酸β氧化一磷酸腺苷激活蛋白激酶(AMP-activated protein kinase,AMPK)-乙酰辅酶A羧化酶(acetyl-CoA carboxylase,ACC)-肉毒碱脂酰转移酶1(carnitine palmitoyltransferase,CPT1)通路所涉及的相关酶活力和质量浓度、相关基因mRNA表达量及肉品品质等指标进行测定,研究不同饲养方式对AMPK-ACC-CPT1信号通路相关指标、肌内脂肪含量及肉品品质产生的影响。结果表明:放牧饲养组羊股二头肌AMPK质量浓度、AMPK活力、CPT1质量浓度、AMPK mRNA表达量、亮度、黄度和剪切力显著大于舍饲饲养组(P<0.05);放牧饲养组羊股二头肌ACC活力、ACC mRNA表达量、肌内脂肪含量显著低于舍饲饲养组(P<0.05)。由此可见,放牧饲养可以一定程度上激活AMPK-ACC-CPT1通路,从而减少肌内脂肪沉积,增加羊肉亮度和黄度,降低嫩度,进而影响肉品品质。 相似文献

建立了一种以微晶酚酞作吸附剂分离富集痕量Co的新方法。探讨了α 亚硝基 β 萘酚(HL)及酚酞用量、盐的种类及用量、酸度等因素对Co2+富集率的影响,得到了分离富集钴的最佳条件,讨论了Co2+的富集机理。结果表明:控制pH值为5.0,在1.0 g NaCl存在下,当2.0 g/L α 亚硝基 β 萘酚溶液用量和5%酚酞溶液用量分别为0.60 mL和0.10 mL时,Co2+与α 亚硝基 β 萘酚(HL)反应生成的红棕色螯合物沉淀CoL3被定量吸附在微晶酚酞上,Co2+被定量富集,而Zn2+, Mn2+, Al3+,Cd2+在此条件下不被吸附,从而实现了Co2+与这些离子的定量分离。方法用于合成水样中微量Co2+的定量富集分离测定,富集率为99.8%~108.6%。 相似文献

研究了TZNA β钛合金冷变形过程中的组织和性能。采用辊模冷拉拔的方式分别进行了20%、40%和60%冷变形,合金表现出良好的冷加工性能。当冷变形在20%时,出现了孪晶组织,使合金强度有明显提升。当冷变形量超过20%时,滑移成为了主要的变形方式。当冷变形达到60%时,晶粒完全破碎,晶粒尺寸在20-40nm之间,具有较高的强度和塑性。 相似文献

This paper presents the measurement results of gross β activities in fallout samples collected from the environment around Qinshan Nuclear Power Plant (NPP) during 1993-2002. The gross β activity of 191 samples collected at five monitoring spots were (0.11~3.64) Bq · m-2 · d-1, with an average of (0.91 ±0.49) Bq · m-2 · d-1. This value was lower than (1.02±0.35) Bq·m-2·d-1 , the average of the samples collected at the reference spot in Hangzhou.It indicated that no obvious rise in the gross β activity level of fallout was observed in ambience of Qinshan NPP. 相似文献

The focus of this study was the effect of spatial confinement on the development of nucleating agent-induced β phase polypropylene (PP) in the dynamically vulcanized thermoplastic elastomers (TPVs) based on dynamically vulcanized PP/ethylene-propylene-diene rubber (EPDM) blend. The melting behaviors, crystalline structures and the morphologies of the blends were studied by differential scanning calorimetry (DSC), wide-angle X-ray diffraction (WAXD) and scanning electron microscopy (SEM). The results indicate that the EPDM phase undergoes a series of changes from the dispersed phase to a continuous one, and again to the dispersed phase with increased content of curing agent, and the PP component always shows itself in a continuous phase. In this process, with the content of the nucleating agent unchanged, the content of β phase PP in the blends initially increases a little and then decreases with increasing PF (Phenolic resin) content. Serine‐ and metallo‐β‐lactamases present a threat to the clinical use of nearly all β‐lactam antibiotics, including penicillins, cephalosporins, and carbapenems. Efforts to develop metallo‐β‐lactamase (MBL) inhibitors require suitable screening platforms to allow the rapid determination of β‐lactamase activity and efficient inhibition. Unfortunately, the platforms currently available are not ideal for this purpose. Further progress in MBL inhibitor identification requires inexpensive and widely applicable assays. Herein the identification of an inexpensive and stable chromogenic substrate suitable for use in assays of clinically relevant MBLs is described. (6R,7R)‐3‐((4‐Nitrophenoxy)methyl)‐8‐oxo‐7‐(2‐phenylacetamido)‐5‐thia‐1‐azabicyclo[4.2.0]oct‐2‐ene‐2‐carboxylic acid 5,5‐dioxide (CLS405) was synthesised in a three‐step protocol. CLS405 was then characterised spectroscopically, and its stability and kinetic properties evaluated. With a Δλmax value of 100 nm between the parent and hydrolysis product, a higher analytical accuracy is possible with CLS405 than with commonly used chromogenic substrates. The use of CLS405 in assays was validated by MBL activity measurements and inhibitor screening that resulted in the identification of N‐hydroxythiazoles as new inhibitor scaffolds for MBLs. Further evaluation of the identified N‐hydroxythiazoles against a panel of clinically relevant MBLs showed that they possess inhibitory activities in the mid‐ to low‐micromolar range. The findings of this study provide both a useful tool compound for further inhibitor identification, and novel scaffolds for the design of improved MBL inhibitors with potential as antibiotics against resistant strains of bacteria. 相似文献

利用低本底α、β测量仪,通过一系列比较实验和数据获得影响水中总α、总β放射性活度的实验因素。按照现行国标中的标准曲线法进行实验并分析关系曲线的拟合结果;采用^(241)Am和^(40)K标准粉末源进行串道干扰实验;最后利用热释光测量仪分析影响样品源计数值的因素。结果表明:“质量厚度净计数率”关系曲线线性拟合结果良好,可应用在实验计算中;在测量放射性活度较高的水样时,α通道对β通道产生的串道干扰需进行修正;样品源制备完毕后,不宜立刻测量,应贮存在干燥低本底的环境中,贮存后应用红外灯干燥足够长的时间并完全冷却后进行测量。 相似文献
