The purpose of this study was to prepare sustained-release pellets of nifedipine (NSPs) based on MCC matrix. Wet-milling and extrusion-spheronization techniques were employed to prepare the microcrystals and pellets, respectively. The drug release mechanism and the influencing factors were investigated. After milled with HPMC (E5), the mean particle size of nifedipine in co-grinding mixture (CGM) was 5?μm, which is 15-fold smaller than that of raw material. DSC, X-ray powder diffraction and microscopic observation confirmed the microcrystals of drug were maintained in the CGM. With increased milling time and the content of HPMC, the dissolution rate was greatly enhanced compared with the raw material. The NSPs prepared by MCC and the CGM, which was obtained by cogrinding nifedipine with 5% HPMC solution for 210?min, exhibited sustained release pattern within 8?h. Nifedipine release from MCC-based NSPs followed the Korsmeyer model and closely related to the microstructure of pellet. High stability of NSPs was confirmed after 6 months of accelerated stability test. Using commercially available sustained product as reference, bioequivalence study in beagle dogs was executed and two formulations were bioequivalent. Bioavailability of a poorly soluble drug can be improved by preparing a drug nanosuspension and subsequently drying it into nanocomposite microparticles (NCMPs). Unfortunately, drug nanoparticles aggregate during milling and drying, causing incomplete recovery and slow dissolution. The aim of this study is to investigate the impact of various classes of dispersants on drug dissolution from drug NCMPs, with the ultimate goal of enhancing the bioavailability of poorly water-soluble drugs via high drug nanoparticle loaded, surfactant-free NCMPs. Precursor suspensions of griseofulvin (GF, model drug) nanoparticles in the presence of various dispersants were prepared via wet stirred media milling and spray dried to form the NCMPs. Hydroxypropyl cellulose (HPC, polymer) alone and with sodium dodecyl sulfate (SDS, surfactant) was used as a base-line stabilizer/dispersant during milling. Two swellable crosslinked polymers, croscarmellose sodium (CCS) and sodium starch glycolate (SSG), and a conventional soluble matrix former, Mannitol, were used in addition to HPC. Besides being used as-received, CCS was also wet co-milled with GF for two different durations to examine the impact of CCS particle size. Laser diffraction, scanning electron microscopy, powder X-ray diffraction (XRD), UV spectroscopy, NCMP redispersion and dissolution tests were used for characterization. The results show that incorporation of CCS/SSG, preferably wet-milled to a wide particle size distribution, into the spray-dried NCMPs resulted in fast release and dispersion of drug nanoparticle clusters. The swellable dispersants were superior to Mannitol in dissolution enhancement, and could achieve fast release comparable to SDS, demonstrating the feasibility of spray drying to prepare high drug-loaded, surfactant-free nanocomposites. 相似文献

This paper is concerned with the computer-based sliding mode control of flexible rotor—magnetic bearing systems (FR-MBS). The plant dynamics consisting of actuator dynamics and flexible rotor dynamics are described. The reduced-order model for controller design is given by eliminating higher-order modes of the mechanical and electrical magnetic interaction system. A discrete-time sliding mode controller with a new robust reduced-order variable structure system (VSS) observer is proposed and its robust performance is evaluated with several simulations based on a calculation model. This digital controller is implemented to replace a linear analogue PID compensator. Levitation tests using the proposed digital controller are performed and compared with those of the PID compensator. The unstable modes can be easily controlled with the good stability and the spillover phenomena due to ignored higher-order modes are not generated. It is indicated that the discrete-time sliding mode control has robustness to the model parameter variations and external disturbances. Using the discrete time sliding mode controller with the reduced-order VSS observer, the test rig of the magnetic bearing system can be successfully rotated in a speed range of 0–35 000 rpm, which includes the first critical shaft frequency of the flexible rotor. 相似文献

实验研究了不同超高压条件对湿粉条中腐败霉菌黑曲霉(Aspergillus 305.01和Aspergillus 305.02)杀菌效果以及对粉条蒸煮特性的影响。实验结果表明随着处理压力升高和时间延长,超高压对黑曲霉的杀菌效果增强。经400 MPa/10 min的超高压处理后,粉条的Aspergillus 305.01与Aspergillus 305.02均下降了6.01个对数值;与未处理菌悬液相比,Aspergillus 305.01和Aspergillus 305.02的核酸吸光值分别从0.03和0.04增加至0.56和0.48、蛋白质吸光值分别从0.04和0.08增加至0.70和0.44、电导率分别从0.04 mS/cm和0.06 mS/cm增加至0.12 mS/cm和0.20 mS/cm。在400 MPa处理下,提高超高压处理时间,电导率、核酸损失和蛋白质损失增多,表明超高压是通过改变细胞膜的部分功能,引起胞内核酸、蛋白质和电解质等物质流出,从而导致细胞死亡。此外,随着处理时间增加,粉条的煮沸损失、膨润度增大,而断条率变化不显著。综合超高压对湿粉条的杀菌效果和蒸煮特性的影响,400 MPa处理10 min是湿粉条杀菌的较优条件,为超高压改善湿粉条的杀菌效果和品质提供了理论依据。 相似文献
