The three-component reaction between amine, carbonyl compound and thioglycolic acid is now considered as a major strategy for synthesis of 1,3-thiazolidin-4-ones, which consists of the following steps: (i) condensation of aldehyde and amine which results the formation of an imine; (ii) the reaction between thioglycolic acid and the imine which is followed by an intramolecular cyclization reaction, which leads to the formation of the final product. In this way, if no suitable catalyst is employed, the completion of the reaction will not be achieved. Hence, it is of great importance to select an appropriate catalyst so that these compounds can be successfully synthesized. Estrogen-related receptor α (ERRα), which is overexpressed in a variety of cancers has been considered as an effective target for anticancer therapy. ERRα inverse agonists have been proven to effectively inhibit the migration and invasion of cancer cells. As few crystalline complexes have been reported, molecular dynamics (MD) simulations were carried out in this study to deepen the understanding of the interaction mechanism between inverse agonists and ERRα. The binding free energy was analyzed by the MM-GBSA method. The results show that the total binding free energy was positively correlated with the biological activity of an inverse agonist. The interaction of the inverse agonist with the hydrophobic interlayer composed of Phe328 and Phe495 had an important impact on the biological activity of inverse agonists, which was confirmed by the decomposition of energy on residues. As Glu331 flipped and formed a hydrogen bond with Arg372 in the MD simulation process, the formation of hydrogen bond interaction with Glu331 was not a necessary condition for the compound to act as an inverse agonist. In the present work, we performed immunochemical studies of LPS, especially the O-specific polysaccharide (O-PS) of Aeromonas veronii bv. sobria strain K133, which was isolated from the kidney of carp (Cyprinus carpio L.) during an outbreak of motile aeromonad infection/motile aeromonad septicemia (MAI/MAS) on a Polish fish farm. The structural characterization of the O-PS, which was obtained by mild acid degradation of the LPS, was performed with chemical methods, MALDI-TOF mass spectrometry, and 1H and 13C NMR spectroscopy. It was revealed that the O-PS has a unique composition of a linear tetrasaccharide repeating unit and contains a rarely occurring sugar 2,4-diamino-2,4,6-trideoxy-D-glucose (bacillosamine), which may determine the specificity of the serogroup. Western blotting and ELISA confirmed that A. veronii bv. sobria strain K133 belongs to the new serogroup PGO1, which is one of the most commonly represented immunotypes among carp and trout isolates of Aeromonas sp. in Polish aquacultures. Considering the increase in the MAI/MAS incidences and their impact on freshwater species, also with economic importance, and in the absence of an effective immunoprophylaxis, studies of the Aeromonas O-antigens are relevant in the light of epidemiological data and monitoring emergent pathogens representing unknown antigenic variants and serotypes. 相似文献

Introduction of C4 photosynthetic traits into C3 crops is an important strategy for improving photosynthetic capacity and productivity. Here, we report the research results of a variant line of sorghum–rice (SR) plant with big panicle and high spikelet density by introducing sorghum genome DNA into rice by spike-stalk injection. The whole-genome resequencing showed that a few sorghum genes could be integrated into the rice genome. Gene expression was confirmed for two C4 photosynthetic enzymes containing pyruvate, orthophosphate dikinase and phosphoenolpyruvate carboxykinase. Exogenous sorghum DNA integration induced a series of key traits associated with the C4 pathway called “proto-Kranz” anatomy, including leaf thickness, bundle sheath number and size, and chloroplast size in bundle sheath cells. Significantly, transgenic plants exhibited enhanced photosynthetic capacity resulting from both photosynthetic CO2-concentrating effect and improved energy balance, which led to an increase in carbohydrate levels and productivity. Furthermore, such rice plant exhibited delayed leaf senescence. In summary, this study provides a proof for the feasibility of inducing the transition from C3 leaf anatomy to proto-Kranz by spike-stalk injection to achieve efficient photosynthesis and increase productivity. 相似文献

The discovery of novel intronic variants in the ABCA4 locus has contributed significantly to solving the missing heritability in Stargardt disease (STGD1). The increasing number of variants affecting pre-mRNA splicing makes ABCA4 a suitable candidate for antisense oligonucleotide (AON)-based splicing modulation therapies. In this study, AON-based splicing modulation was assessed for 15 recently described intronic variants (three near-exon and 12 deep-intronic variants). In total, 26 AONs were designed and tested in vitro using a midigene-based splice system. Overall, partial or complete splicing correction was observed for two variants causing exon elongation and all variants causing pseudoexon inclusion. Together, our results confirm the high potential of AONs for the development of future RNA therapies to correct splicing defects causing STGD1. 相似文献

Since 2010, several treatment options have been available for men with metastatic castration-resistant prostate cancer (mCRPC), including immunotherapeutic agents, although the clinical benefit of these agents remains inconclusive in unselected mCRPC patients. In recent years, however, immunotherapy has re-emerged as a promising therapeutic option to stimulate antitumor immunity, particularly with the use of immune checkpoint inhibitors (ICIs), such as PD-1/PD-L1 and CTLA-4 inhibitors. There is increasing evidence that ICIs may be especially beneficial in specific subgroups of patients with high PD-L1 tumor expression, high tumor mutational burden, or tumors with high microsatellite instability/mismatch repair deficiency. If we are to improve the efficacy of ICIs, it is crucial to have a better understanding of the mechanisms of resistance to ICIs and to identify predictive biomarkers to determine which patients are most likely to benefit. This review focuses on the current status of ICIs for the treatment of mCRPC (either as monotherapy or in combination with other drugs), mechanisms of resistance, potential predictive biomarkers, and future challenges in the management of mCRPC. 相似文献

采用水热法制备GRQD-NiCo2O4复合物,利用XRD、SEM及TEM分析其微结构,并探讨其作为DMFC阳极催化剂使用时的电化学性能。微结构分析表明所得GRQD-NiCo2O4复合物皆为具NiCo2O4单一相的尖晶石结构,且GRQD质量浓度高于0.25 g/mL后表面形貌将转变GRQD与NiCo2O4相互结合的状态。电化学分析表明添加GRQD可有效增强NiCo2O4的导电性并提升其电化学稳定性,其中GRQD质量浓度为0.25 g/mL时所得样品经500次循环测试后电流密度约为77.5 A/g,与循环5次后相比其电流密度剩余量最大(约为69.7%),该样品作为DMFC阳极催化剂使用时性价比最佳。 相似文献

铁基载氧体是一种具有工业应用前景的载氧体,但存在氧利用率低、在高温下易烧结等问题。虽可通过制备双金属复合载氧体或添加惰性组分改进其性能,但均存在一定缺陷。若将活性组分和惰性材料融入到一个晶体结构制备尖晶石结构载氧体,则可实现利用双金属协同作用提高载氧体活性的同时,利用Al3+提高载氧体的稳定性。采用共沉淀法和溶胶凝胶法制备了具有尖晶石结构的NiFeAlO4载氧体,考察了制备方法、载氧体与煤质量比对NiFeAlO4载氧体化学链燃烧特性和循环稳定性的影响,并分析了载氧体对煤转化过程的作用。结果表明,溶胶凝胶法制备的NiFeAlO4载氧体具有更好的反应性,载氧体与煤质量比为20∶1时,碳转化率为86.7%,远高于煤单独热解时的碳转化率(34%),此时CO2体积分数为93.6%。对反应前后NiFeAlO4载氧体晶相结构和形貌进行分析,表明循环过程中经“还原-氧化”后生成的NiO和载氧体颗粒团聚是导致载氧体活性下降的主要原因。相较于载热作用,NiFeAlO4载氧体在煤化学链燃烧中主要起供氧作用,其不仅会促进挥发分向煤气的转化,且NiFeAlO4载氧体与焦炭之间也存在固-固反应,利于更多CO2的生成。 相似文献
