Mirrors will be used in ITER in all optical diagnostic systems observing the plasma radiation in the ultraviolet, visible and infrared ranges. Diagnostic mirrors in ITER will suffer from electromagnetic radiation, energetic particles and neutron irradiation. Erosion due to impact of fast neutrals from plasma and deposition of plasma impurities may significantly degrade optical and polarization characteristics of mirrors influencing the overall performance of the respective diagnostics. Therefore, maintaining the best possible performance of mirrors is of the crucial importance for the ITER optical diagnostics. Mirrors in ITER divertor are expected to suffer from deposition of impurities. The dedicated experiment in a tokamak divertor was needed to address this issue. Investigations with molybdenum diagnostic mirrors were made in DIII-D divertor. Mirror samples were exposed at different temperatures in the private flux region to a series of ELMy H-mode discharges with partially detached divertor plasmas. An increase of temperature of mirrors during the exposure generally led to the mitigation of carbon deposition, primarily due to temperature-enhanced chemical erosion of carbon layers by D atoms. Finally, for the mirrors exposed at the temperature of ∼160 °C neither carbon deposition nor degradation of optical properties was detected.  相似文献

We recently reported the successful use of the loop-mediated isothermal amplification (LAMP) reaction for hepatitis B virus (HBV) DNA amplification and its optimal primer design method. In this study, we report the development of an integrated isothermal device for both amplification and detection of targeted HBV DNA. It has two major components, a disposable polymethyl methacrylate (PMMA) micro-reactor and a temperature-regulated optical detection unit (base apparatus) for real-time monitoring of the turbidity changes due to the precipitation of DNA amplification by-product, magnesium pyrophosphate. We have established a correlation curve (R2 = 0.99) between the concentration of pyrophosphate ions and the level of turbidity by using a simulated chemical reaction to evaluate the characteristics of our device. For the applications of rapid pathogens detection, we also have established a standard curve (R2 = 0.96) by using LAMP reaction with a standard template in our device. Moreover, we also have successfully used the device on seven clinical serum specimens where HBV DNA levels have been confirmed by real-time PCR. The result indicates that different amounts of HBV DNA can be successfully detected by using this device within 1 h.  相似文献

In examining the performance of screening scales, a distinction should be made between principal and additional diagnoses. The Psychiatric Diagnostic Screening Questionnaire (PDSQ) is a brief, psychometrically strong self-report scale designed to screen for the most common Diagnostic and Statistical Manual of Mental Disorders (4th ed.; DSM-IV) Axis I disorders encountered in outpatient mental health settings. In the present report, the authors compared the performance of the PDSQ in identifying principal and comorbid disorders. Seven hundred ninety-nine psychiatric outpatients completed the PDSQ and were interviewed with the Structured Clinical Interview for DSM-IV. The diagnostic parameter—time dependence, usually used for prediction of the residual service time, is given here its theoretical grounding. The relationships involved are derived on the basis of the machine's heat balance for two time approaches—short-term (starting time) and—long-term (service time). The diagnostic parameter considered is the temperature change rate during the running-in period of the machine. The reference dependence obtained from theoretical values of the diagnostic parameter is compared against its counterpart by the traditional empirical method. The residual service life calculated by these two approaches is also analyzed. It is shown that the theoretical consideration is valid and can be applied in reliable machine diagnostics.  相似文献

目的制备高效价、高纯度的布鲁菌多克隆抗体,作为检测布鲁菌的诊断试剂。方法采用弗氏佐剂乳化抗原家兔背部皮下多点免疫,制备抗体,纯化后用SDS-PAGE进行鉴定,ELISA法检测抗体水平,紫外分光光度仪检测蛋白质含量,AlphaScreenRabbitIgGDetectionKit检测IgG含量,并计算纯度。结果SDS-PAGE结果显示多抗片段大小与预期相符,B.melitensis16M和B.abortus544A抗体效价分别为1∶25600和1∶51200,纯化后抗体蛋白含量分别为11·32和14·03mg/ml,IgG含量分别为10·45和13·12mg/ml,且纯度分别达到92·3%和93·5%。结论已获得高效价、高纯度的多克隆抗体IgG,为建立布鲁菌免疫检测方法奠定了基础。  相似文献

Nuclear magnetic resonance (NMR) spectroscopy has emerged as a technology that can provide metabolite information within organ systems in vivo. In this study, we introduced a new method of employing a clustering algorithm to develop a diagnostic model that can differentially diagnose a single unknown subject in a disease with well-defined group boundaries. We used three tests to assess the suitability and the accuracy required for diagnostic purposes of the four clustering algorithms we investigated (K-means, Fuzzy, Hierarchical, and Medoid Partitioning). To accomplish this goal, we studied the striatal metabolomic profile of R6/2 Huntington disease (HD) transgenic mice and that of wild type (WT) mice using high field in vivo proton NMR spectroscopy (9.4 T). We tested all four clustering algorithms (1) with the original R6/2 HD mice and WT mice, (2) with unknown mice, whose status had been determined via genotyping, and (3) with the ability to separate the original R6/2 mice into the two age subgroups (8 and 12 weeks old). Only our diagnostic models that employed ROC-supervised Fuzzy, unsupervised Fuzzy, and ROC-supervised K-means Clustering passed all three stringent tests with 100% accuracy, indicating that they may be used for diagnostic purposes.  相似文献
