Despite the established efficacy of statin therapy, the risk of cardiovascular events remains high in many patients. We examined high-density lipoprotein (HDL) subclass distribution profiles among statin-treated coronary heart disease (CHD) patients undergoing percutaneous coronary intervention (PCI). Plasma HDL subclasses were measured in 85 patients with established CHD and quantified by two-dimensional gel electrophoresis and immunoblotting. In CHD patients with statin treatment, the mean value of total cholesterol (TC) reached the desirable level and the triacylglycerol level (TAG) was borderline high. Moreover, low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C), apolipoproteinA-I, and apolipoproteinB-100 levels in these patients resembled those in normolipidemic healthy subjects. The HDL subclass did not show a normal distribution and was characterized by the lower large-sized HDL2b contents and higher contents of small-sized preβ1-HDL in CHD patients, compared to those in normolipidemic control subjects. Multiple stepwise regression analysis revealed that the severity of coronary stenosis, determined by the Gensini Score, was significantly and independently predicted by HDL2b and HDL3b. Statin therapy was effective in modifying plasma lipids levels, but not adequate as a monotherapy to normalize the HDL subclass distribution phenotype of patients with CHD undergoing PCI. The aim of this study was to develop an improved general method for detecting and quantifying mixtures of hydroxy acids and other products of glycerol oxidation in aqueous media, to prevent the confusions that can occur due to similarities and interactions between these compounds depending on media conditions. Standard potential products of glycerol oxidation—glycerol, glyceraldehyde, dihydroxyacetone, glyceric acid, lactic acid, glycolic acid, glyoxylic acid, oxalic acid, tartronic acid, and mesoxalic acid—were analyzed by high-performance liquid chromatography (HPLC) and quantitative 13C nuclear magnetic resonance (NMR), in mixtures of known composition. The results obtained were concordant with the known compositions tested. HPLC was more accurate than quantitative 13C NMR for simple mixtures, but 13C NMR was required for complex mixtures containing dihydroxyacetone and glycerol, oxalic acid and mesoxalic acid, or glyoxylic acid and tartronic acid, pairs of compounds not well separated or detected by HPLC. As proof-of-concept, an unknown mixture generated by glycerol oxidation was analyzed by HPLC and quantitative 13C NMR. Nonlinear acoustic coefficients have a close relationship with structural cracks. A nonlinear ultrasonic modulation method for micro-crack quantitative evaluation is developed. The influence of phase threshold on the crack evaluation is discussed. Ultrasonic modulation using a unilateral incentive model is applied to the quantitative evaluation of micro-cracks for different specimens. The experimental results indicate that the method can be beneficially applied for micro-cracks detection. A proper phase threshold can improve the reliability of the method based on nonlinear ultrasonic modulation. The presented modulation factor can be used to quantitatively evaluate the structural cracks, and it is suitable for small-crack quantitative evaluation. 相似文献

目的建立检测重组人血小板生成素(rhTPO)含量的改良双抗体夹心ELISA方法。方法用棋盘滴定法确定包被单抗、一抗和酶标二抗的最佳工作浓度;绘制标准曲线,确定线性范围、检测限和定量限;检测该方法的准确性(回收率)、精密性和特异性,并与Lowry法检测结果进行比较。结果包被单抗的最佳包被浓度为1.0μg/ml,一抗和酶标二抗的最佳使用浓度分别为1∶5000和1∶10000;检测的线性范围为(125~4000)pg/ml,检测限为75pg/ml,定量限为189pg/ml;不同浓度rhTPO的回收率在(97.0±2.1)%~(105.0±8.5)%之间;6批样品的试验内变异系数在3.2%~8.1%之间,试验间变异系数在5.7%~10.4%之间,特异性良好;与Lowry法检测结果具有良好的相关性。结论已建立检测rhTPO含量的改良双抗体夹心ELISA法,准确性、精密性、特异性良好,可用于rhTPO研究及生产过程中的定量分析。 相似文献
