The nitric oxide–guanylyl cyclase-1–cyclic guanylate monophosphate (NO–GC-1–cGMP) pathway is integral to the control of vascular tone and morphology. Mice lacking the alpha catalytic domain of guanylate cyclase (GC1−/−) develop retinal ganglion cell (RGC) degeneration with age, with only modest fluctuations in intraocular pressure (IOP). Increasing the bioavailability of cGMP in GC1−/− mice prevents neurodegeneration independently of IOP, suggesting alternative mechanisms of retinal neurodegeneration. In continuation to these studies, we explored the hypothesis that dysfunctional cGMP signaling leads to changes in the neurovascular unit that may contribute to RGC degeneration. We assessed retinal vasculature and astrocyte morphology in young and aged GC1−/− and wild type mice. GC1−/− mice exhibit increased peripheral retinal vessel dilation and shorter retinal vessel branching with increasing age compared to Wt mice. Astrocyte cell morphology is aberrant, and glial fibrillary acidic protein (GFAP) density is increased in young and aged GC1−/− mice, with areas of dense astrocyte matting around blood vessels. Our results suggest that proper cGMP signaling is essential to retinal vessel morphology with increasing age. Vascular changed are preceded by alterations in astrocyte morphology which may together contribute to retinal neurodegeneration and loss of visual acuity observed in GC1−/− mice.  相似文献

Vascular permeability is a selective mechanism that maintains the exchange between vessels, tissues, and organs. The regulation was mostly studied during the nineteenth century by physiologists who defined physical laws and equations, taking blood, tissue interstitial, and oncotic pressure into account. During the last decades, a better knowledge of vascular cell functions and blood-vessel interactions opens a new area of vascular biology. Endothelial cell receptors vascular cell adhesion molecule (VCAM), intercellular cell adhesion molecule (ICAM), vascular endothelial growth factor receptor (VEGFR-2), receptor for advanced glycation end products (RAGE), and mediators were identified and their role in homeostasis and pathological situations was described. The molecular differences of endothelial cell junctions (tight, gap, and adherens junctions) and their role in vascular permeability were characterized in different organs. The main mediators of vasomotricity and permeability, such as prostaglandins, nitric oxide (NO), prostacyclin, vascular growth factor (VEGF), and cytokines, have been demonstrated to possess major functions in steady state and pathological situations. Leukocytes were shown to adhere to endothelium and migrate during inflammatory situations and infectious diseases. Increased vascular permeability is linked to endothelium integrity. Glycocalyx, when intact, may limit cancer cell metastasis. Biological modifications of blood and tissue constituents occurring in diabetes mellitus were responsible for increased permeability and, consequently, ocular and renal complications. Vascular pressure and fluidity are major determinants of pulmonary and cerebral edema. Beside the treatment of the infectious disease, of the blood circulation dysfunction and inflammatory condition, drugs (cyclooxygenase inhibitors) and specific antibodies anti-cytokine (anti-VEGF) have been demonstrated to reduce the severity and the mortality in diseases that exhibited enhanced vascular permeability.  相似文献

The transition metal sulfides have gained extensive interest in energy storage devices owing to their unique features. However, the research-based on cobalt, copper and manganese sulfide composites is limited while they are considered as promising contenders for supercapacitor electrodes. The simplest and facile one-step electrodeposition technique was adopted for the direct growth of CuMnS and CoMnS on a Ni-substrate. The electrochemical properties of CuMnS and CoMnS electrodes were investigated and maximum specific capacitances of 1691 and 2290 F/g, respectively, were obtained at 10 A/g current density. Further, these electrodes are investigated with activated carbon (AC) electrode to fabricate asymmetric supercapacitor devices where CoMnS//AC exhibited superior energy density values than CuMnS//AC device. However, both the devices show a relatively uniform capacitance retention rate (~94%) after 2500 charging-discharging cycles. Furthermore, the role of capacitive- and diffusive-controlled contributions in the charge storage phenomenon of supercapacitor devices are explicitly scrutinized by employing Dunn's model. Co-electrodeposition of transition metal sulfides has great potential as electrode material for highly effective supercapacitor devices.  相似文献

To promote the densification and therefore the mechanical properties of boride-based ceramics, MgO was added as sintering aid into Os0.9Re0.1B2 powders for densification by using spark plasma sintering (SPS). The Os0.9Re0.1B2 powders were synthesized by mechanochemical method from powder mixture of Os, Re and amorphous B. The role of MgO on densification, phase composition, microstructure and mechanical properties (hardness, fracture toughness and wear behavior) were studied by using X-ray diffraction (XRD), scanning electron microscope (SEM) with energy-dispersive spectroscopy (EDS), micro indentation and ball-on-disk tribometer. The results show that, with the introduction of MgO as sintering aid, the relative density of the Os0.9Re0.1B2 ceramic samples increased. When the MgO content reached 9 wt%, the as-sintered sample is almost fully dense. No obvious regularity was found from the samples with the addition of different content of MgO. Vickers hardness values of the samples with 0, 3 wt% and 9 wt% MgO are found to be very close with each other within the experimental error (~30 GPa), while the sample with the addition of 6 wt% MgO exhibits the highest hardness of ~35 GPa. The fracture toughness of the samples is decreased slightly with the addition of MgO. The friction coefficient and wear rate of the sample with the addition of 6 wt% MgO was also found to be the lowest among all samples, which indicate best wear resistance. As a whole, with the addition content of 6 wt% MgO, the Os0.9Re0.1B2 ceramic sample performs relatively excellent mechanical properties among four groups of samples.  相似文献

In this study, boron carbide-metallic boride (B4C-MeBx, Me = Ti, Zr, Nb, Ta, or W) multiphase ceramics were fabricated via in situ pressureless sintering at 2250 °C for 1 h. The effects of transition metal carbides, namely, TiC, ZrC, NbC, TaC, and WC, on the phase composition, microstructure, and mechanical properties of the ceramics were investigated. The results showed that MeC could facilitate the sintering densification of B4C by distributing second-phase particles uniformly throughout the B4C. Additionally, the main phases observed were B4C and (Me, W)Bx (Me = Ti, Zr, Nb, or Ta) due to the doping of a small amount of WC during the ball milling process. As a result, the mechanical properties of B4C-MeBx showed significant improvements when compared with those of single-phase B4C ceramics. B4C–NbB2 ceramics were found to exhibit the best mechanical properties, with an elastic modulus of 393.0 GPa, a hardness of 28.7 GPa, a flexural strength of 368.0 MPa, and a fracture toughness of 6.94 MPa m1/2.  相似文献

Two-dimensional transistors are promising candidates for the next generation of nanoscale devices. Like the other alternatives, they also encounter problems such as instability under standard condition (STP), low channel mobility, small band gaps, and difficulty to integrate metal contacts. The latter poses a great challenge since metal/semiconductor interface significantly affects the transistor‘s performance. Some of these obstacles can be solved by using two-dimensional transition metal di-chalcogenides (TMDC) materials. In this study, we performed charge transport calculation based on density functional theory (DFT) followed by wave dynamics to evaluate the performance of six two-dimensional TMDC metal/semiconductor/metal systems. Each semiconductor monolayer was laterally connected, at both ends to metal contacts consisting of VS2 or FeS2 monolayers. We found that charge transport was more efficient in systems containing a CrS2 semiconductor monolayer compared to systems with MoS2 or WS2 as the semiconductor monolayer. The electronic characterization of the monolayer TMDC materials by DFT estimates well the trend in charge transport efficiency calculated using wave packet dynamics.  相似文献
