We report that some prokaryotic repressors including CamR andTetR belong to the same family. CamR and TetR bind to DNA usinga multihelical DNA binding domain (DBD) at the N-termini ofthe proteins, while the C-termini are important for regulatingthe DNA binding in a manner dependent on their co-factors (camphorfor CamR, tetracycline for TetR). In all, 11 important aminoacid positions have been identified in the CamR DBD by the systematicsubstitution of residues by Ala. Of the 11 positions, 10 areeither buried in the core, and thus important for creating thehydrophobic environment, or exposed on the surface, and thusimportant for binding to DNA. The eleventh residue, Gly, seemsto be important for a loop structure. The DNA binding mode ofthis type of DBD and a general mechanism of regulating theirDNA binding are discussed in reference to the crystal structureof TetR [Hinrichs et al., (1994) Science, 264, 418–420].  相似文献

Concerns regarding the possible environmental effects of organochlorine by‐products from bleaching of pulp with chlorine‐based compounds have led to the pulp and paper industry developing new bleaching sequences. Ozone, oxygen and hydrogen peroxide are the main reagents in these Totally Chlorine Free (TCF) bleaching processes.

In this study, eucalypt kraft pulps from a variety of Australian wood sources were subjected to bleaching sequences comprising oxygen, ozone and hydrogen peroxide/alkali extraction stages. The aqueous liquid effluents from each stage were analyzed by GC/MS for aldehydes, ketones, alcohols, carboxylic acids and other by‐products. Pentafluorobenzyl oxime derivatives of the aldehydes and ketones were analyzed by electron impact GC/MS. The major carbonyl compounds detected were formaldehyde, glyoxal, dimethylglyoxal and acetone. An homologous series of n‐aldehydes corresponding to cleavage of ω‐3, 6, 9 and 12 unsaturated fatty acids also was detected. Aromatic aldehydes were identified in the oxygen stage and high consistency ozone stages, but not in any medium consistency ozone or post‐ozone bleach stages. In all stages a series of saturated alkyl carboxylic acids from formic to octacosanoic acid was detected. Formic and acetic acids were present in the highest yield. Only trace quantities of unsaturated fatty acids were detected. Details of these and other compounds detected are discussed.  相似文献

目的在大肠杆菌中表达结核分枝杆菌RD1区11kD蛋白,并检测表达产物的效力。方法构建11kD蛋白编码基因原核表达载体,转化大肠杆菌BL21(DE3)IPTG诱导表达,离子层析柱分离纯化重组蛋白。以结核分枝杆菌致敏豚鼠进行皮肤变态反应(DTH)测定。结果结核分枝杆菌11kD蛋白在大肠杆菌中以可溶形式表达,表达蛋白占总菌体蛋白的33%以上,纯化后纯度达95%以上,并可诱导结核分枝杆菌致敏豚鼠产生迟发型超敏反应,5μg/ml11kD蛋白的效力与50IU/ml TB-PPD相当。结论重组11kD蛋白已在大肠杆菌中成功表达,并有望成为结核病皮试鉴别诊断用新试剂。  相似文献

Copolymers of N‐acryloylcarbazole (A) and vinyl acetate (V) were synthesized by bulk polymerization using benzoyl peroxide (BPO) as free‐radical initiator at 65°C in different in‐feed ratios. The composition of the copolymer was determined by 1H‐NMR spectrum. The comonomer reactivity ratios, determined by Kelen–Tudos (KT) and nonlinear error‐in‐variables (EVM) methods, were rA= 16.75 ± 1.38, rV = 0.015 ± 0.002, and rA = 16.36, rV = 0.015, respectively. Complete spectral assignments of the 1H and 13C{1H} NMR spectra of the copolymers were done by the help of distortionless enhancement by polarization transfer (DEPT) and two‐dimensional NMR techniques such as heteronuclear single quantum coherence (HSQC) and total correlation spectroscopy (TOCSY). The methine and methylene carbon resonances were found to be compositional as well as configurational sensitive. The signals obtained were broad pertaining to the restricted rotation of bulky carbazole group. The thermal stability and glass‐transition temperatures (Tg) of the copolymers were found to be dependant on polymer composition and characteristic of rotational rigidity of the polymer chain. Variation in the values of Tg with the copolymer composition was found to be in good agreement with theoretical values obtained from Johnston and Barton equations. © 2006 Wiley Periodicals, Inc. In previous studies, asphaltenes and resins have been treated as two distinct fractions of a crude oil. The asphaltenes were assumed to be the only self‐associating fraction. However, there is evidence that resins also participate in this self‐association. In this study, molar masses of mixtures of asphaltenes and resins were measured with vapour pressure osmometry. Precipitation from the same mixtures dispersed in solutions of toluene and pentane were also measured. The data were modelled with previously developed self‐association and precipitation models. Model results with asphaltenes and resins characterized as a single distribution and as individual components are compared. The data and the modelling suggest that asphaltenes and resins are better characterized as a single distribution of self‐associating components.  相似文献

Wood dowels are commonly used in the construction furniture, but little information is available about the additive effects of dowels on the ultimate withdrawal strength of single or multidowel joints. This study was carried out to determine the tensile strength of 10‐mm‐diameter dowels produced from medium‐density fiberboard (MDF), plywood, scotch pine (Pinus sylvestris L.), and beech (Fagus orientalis lipsky), bonded parallel and vertical to the surface of MDF and particleboard (Pb) with poly(vinyl acetate) (PVAc) and Desmodur‐VTKA (D‐VTKA). Tensile strength was applied to the dowels according to the procedure in the ASTM‐D 1037 standard. The effects of dowel species, direction of tensile, composite material, and type of adhesive on tensile strength were determined. The results showed that the highest tensile strength was obtained in beech dowels bonded vertically with PVAc adhesive to the surface of MDF at 7.91 N/mm2. If the dowels used in furniture production are subjected to great tensile strength, beech dowels bonded with PVAc adhesive on MDF should be used. Two‐phase decanter olive pomace (TPOP) is the by‐product of a centrifugation system, used to produce olive oil, that separates olive oil and moist pomace. The water content in these olive pomaces is about 70%, while it was 45‐50% in the olive pomace stemming from three‐phase systems (oil, water, and pomace) and 30% in the old press system. The aim of this work is focused on quantification and changes undergone during olive pomace storage in ponds of esters of fatty acids with short‐chain linear alcohols, which can be considered as a refining loss. The results indicate that oil deterioration increases very rapidly, in particular when oil is extracted from the TPOP surface. Special attention should be paid to the storage of TPOP, establishing a maximum of 2 months in all cases.  相似文献

The hydrolytic condensation of 1,3‐dichloro‐1,3‐disila‐1,3‐diphenyl‐2‐oxaindane under neutral conditions produced α'ω‐dihydroxy‐1,3‐disila‐1,3‐diphenyl‐2‐oxaindane (polymerization degree ≈ 4). The homofunctional condensation of α'ω‐dihydroxy‐1,3‐disila‐1,3‐diphenyl‐2‐oxaindane in a toluene solution and in the presence of activated carbon was performed, and dihydroxy‐containing oligomers with various degrees of condensation were obtained. Through the heterofunctional condensation of dihydroxy‐containing oligomers with α'ω‐dichlorodimethylsiloxanes in the presence of amines, corresponding block copolymers were obtained. Gel permeation chromatography, differential scanning calorimetry, thermomechanical analysis, thermogravimetry, and wide‐angle roentgenography investigations were carried out. Differential scanning calorimetry and roentgenography studies of the block copolymers showed that their properties were determined by the ratio of the lengths of the flexible and linear poly(dimethylsiloxane) and rigid poly(1,3‐disila‐1,3‐diphenyl‐2‐oxaindane) fragments in the macromolecular chain. At definite values of the lengths of the flexible and rigid fragments, a microheterogeneous structure was observed in the synthesized block copolymers. © 2002 Wiley Periodicals, Inc. A series of novel copolymer superabsorbents based on monomer acrylamide (AM), potassium methacrylate (KMA), and 2‐hydroxyethyl methacrylate (HEMA) were prepared by copolymerization using ammonium persulfate (APS) as an initiator and N,N‐methylenebisacrylamide (MBA) as a crosslinking agent. The synthetic variables (the monomer concentration, crosslinker concentration, and initiator concentration) were also studied. The experimental results of superabsorbent polymers (SAPs) show a better absorbency in both water and NaCl solutions. The copolymers were characterized by IR spectroscopy. The water retention in the soil was enhanced using the above superabsorbents. The use of SAPs for the growth of groundnut plants was also investigated. SAPs can be considered for water‐managing materials for agriculture and horticulture purposes in desert and drought‐prone areas. © 2002 Wiley Periodicals, Inc. Summary: Non‐Newtonian fluid behavior has significant influence on quantities in chemical engineering like power input, mixing time, heat transfer etc. In the laminar flow region, the concept of effective viscosity by Metzner and Otto is well established. In the transition region between laminar and turbulent flow, the existing concepts use three and even more empirical parameters to determine the specific power input. Here, a unified and general but simple approach is introduced to calculate the power input for shear thinning fluids over the whole flow region using just one empirical parameter. The Metzner‐Otto relation is obtained as a limiting case for the laminar region. The empirical parameter of the new approach is related to the Metzner‐Otto constant. The concept is validated for eight different stirrer systems. Mixing time and maximum shear rate and heat transfer can also be calculated using this approach. The new concept presented should also be applicable for other apparatuses, e.g., static mixers.
