The incidence of hepatocellular carcinoma (HCC) is significantly elevated in a Hispanic community in Bexar County, Texas. Chronic exposure to dietary aflatoxins (AFs) is a major risk factor for HCC; increased risk has been linked to polycyclic aromatic hydrocarbon (PAH) co-exposure and hepatitis virus infection. The aims of this study were to assess AF and PAH exposures, investigate dietary factors that may contribute to increased AF exposure, and determine the prevalence of hepatitis virus infection in Bexar Co. Blood and urine samples were collected from 184 volunteers for biomarker analyses and hepatitis screening. Serum AFB1-lysine adduct, urinary AFM1 and 1-hydroxypyrene (1-OHP) levels were measured using high-performance liquid chromatography. The average AFB1-lysine adduct level detected in 20.6% of serums was 3.84 ± 3.11 pg/mg albumin (range 1.01-16.57 pg/mg). AFM1 was detected in 11.7% of urines, averaging 223.85 ± 250.56 pg/mg creatinine (range 1.89-935.49 pg/mg). AFM1 detection was associated with increased consumption of corn tortillas (p = 0.009), nuts (p = 0.033) and rice (p = 0.037). A significant difference was observed between mean 1-OHP values of non-smokers (0.07 ± 0.13) and smokers (0.80 ± 0.68) μmol/mol creatinine (p < 0.01). A high hepatitis C virus positivity rate (7.1%) was observed. Findings suggest that the incidence and level of AF and PAH exposure were less than those observed in a high-risk population; however, participants consuming higher amounts of foods prone to AF contamination may be more vulnerable to exposure and interactions with other environmental/biological factors (i.e., HCV).  相似文献

A first driving while impaired by alcohol (DWI) conviction is a key opportunity to identify offenders who are at high risk for recidivism. Detection of alcohol use disorder (AUD) is a major target of current DWI assessments. However, offenders frequently underreport their alcohol consumption, and use of biomarkers has been proposed as a more objective indicator. Among the best established are aspartate aminotranferase (AST), alanine aminotransferase (ALT), gamma-glutamyl transferase (GGT), mean corpuscular red blood cell volume (MCV), carbohydrate-deficient transferrin (CDT), and thiamine. To our knowledge, no research has directly verified whether AUD biomarkers predict DWI recidivism status. Using a cross-sectional design, this study tested three hypotheses related to the utility of biomarkers in DWI assessment.

Hypotheses

(1) DWI recidivists possess biomarkers indicative of greater prevalence of AUD compared to first-time offenders; (2) multiple biomarkers better differentiate first-time offenders from recidivists compared to individual biomarkers; and (3) biomarkers add significantly to the prediction of recidivism over and above psychosocial questionnaires.

Methods

First-time offenders (n = 49) and recidivists (n = 95) participated in the study. In addition to self-reported information on sociodemographic and driving characteristics, data from several AUD questionnaires were gathered: Michigan Alcoholism Screening Test, Alcohol Use Disorders Identification Test, Composite International Diagnostic Interview, and Timeline Follow-Back. Blood samples were collected to measure AST, ALT, GGT, MCV, CDT, and thiamine.

Results

AUD biomarkers, taken individually or in combination, did not indicate that recidivists had more frequent AUD compared to first-time offenders. Also, they failed to significantly differentiate first-time offenders from recidivists or predict recidivism status. Finally, the superiority of biomarkers over psychosocial AUD questionnaires was not supported in the laboratory setting.

Conclusion

The present findings suggest that biomarkers of chronic patterns of heavy drinking may not be adequate to capture the multiple processes that appear to promote recidivism (e.g., binge drinking, other risky behavioural and personality features). Despite their objectivity, caution is warranted in the interpretation of a positive score on these biomarkers in DWI assessment. Longitudinal research is needed to more comprehensively explore the relationship between positive biomarkers in first-time offenders and their risk of becoming recidivists.  相似文献

1,3-dinitrobenzene (DNB) and 1,3,5-trinitrobenzene (TNB) are used primarily in explosive compositions and munitions and have been detected as environmental contaminants of surface waters as well as ground waters near production waste disposal sites. Hemoglobin (Hb) adducts have recently been proposed as biological markers of exposure assessment for various environmental compounds, including nitroaromatics. In the present study, we have investigated the formation of DNB and TNB hemoglobin adducts in vivo and in vitro in the blood of shrew (Cryptotis parva). DNB and TNB hemoglobin adducts were detected by GC/MS after either basic (0.1N NaOH) or acid (2N HCl) hydrolysis followed by organic solvent extraction and derivatization of the corresponding amines. The levels of DNB-Hb adducts detected after basic hydrolysis (238.7 ± 50.2 pg/mg Hb) are higher than the corresponding levels detected after acid hydrolysis (52.5 ± 16.2 pg/mg Hb). For the TNB-Hb the levels after acid hydrolysis (132.2 ± 37.8 pg/mg Hb) are higher than the levels detected after basic hydrolysis (44.7 ± 15.3 pg/mg Hb.) These results demonstrate the effectiveness of the hemoglobin adduct model for monitoring exposure to nitroaromatics.  相似文献

An overview on the influence of metabolic and DNA repair polymorphisms on biological indicators of genotoxic risk in occupational, environmental, or lifestyle exposure is presented in this article. Indicators of genotoxic risk include biomarkers of internal dose (urinary concentration of the substance or its metabolites and urinary mutagenicity), biological effective dose (protein and DNA adducts), and indicators of early biological effects (chromosome aberrations, sister chromatid exchanges, micronuclei, COMET assay, HPRT mutants). Genetic polymorphisms include those involved in the activation and detoxification (i.e., P-450 cytochrome, acetyltransferase, glutathione transferase) of various classes of carcinogens and the newly discovered polymorphisms affecting DNA repair enzymes. Genetic polymorphisms are assessed for their importance in detecting the impact of genotype on biological indicators of genotoxic risk.  相似文献

The study of 22 oils from sidewall cores taken at different depths in the Lower Lagunillas Member, well LSJ-AB, Tia Juana Field, Maracaibo Lake is presented, with the purpose of predicting the intervals that present the best crude oil quality. Differences were detected in the biodegradation levels of the studied samples, which are correlated with the depth at which the sidewall core was taken. The API gravity was considered for the oils from each sidewall core and it was found that toward the top of the sequence, the oils have an API gravity of 10.6-11.2°, while toward the base part of the sequence, the well produces extra heavy oils with an API gravity that varies between 8.2 and 8.7°.  相似文献

The present study aimed to enrich the group of sentinel organisms of terrestrial pollution biomonitoring, by investigating the efficacy of the land snail Eobania vermiculata. For this reason, a package of biomarkers was performed on land snails E. vermiculata collected from polluted areas in the field or treated with heavy metals in the laboratory. The biomarkers used were neutral red lysosomal retention assay of the haemocytes, acetylcholinesterase activity in the digestive gland and the haemolymph, and metallothionein content of the digestive gland. Moreover, the morphometric changes in the lysosomal system and the morphometric alterations of the neutral lipids were also investigated. In addition, the content of cadmium, lead and copper was evaluated in the digestive gland of the snails. The results revealed appreciable alterations in the biomarker values both in field- and laboratory-conditions, accompanied by significant correlations among the biomarkers. Therefore, this exploratory study suggests the utility of E. vermiculata as a sentinel organism for biomonitoring the biologic impact of terrestrial pollution, and supports the package's efficacy of the selected biomarkers.  相似文献

The aim of the study was to evaluate the impact of environmental contaminants on oxidative stress biomarkers in hepatocytes of Indian estuarine water grey mullet, Mugil cephalus collected from unpolluted Kovalam and polluted Ennore estuaries. Initially, a comparison was made between the general water chemistry and environmental pollutants like heavy metals to identify the ecotype of the estuaries. Biomarker responses and bioaccumulation of metals were determined along with histological studies of fish hepatocytes to assess contamination impact. Water chemistry data with higher temperature, salinity, pH, biochemical oxygen demand, chemical oxygen demand and low dissolved oxygen and environmental pollutants with higher metal concentration (p < 0.05) were observed at Ennore than Kovalam indicating its polluted environment. A significant increase (p < 0.05) in lipid and protein oxidation markers, decrease (p < 0.05) in thiol status and antioxidant enzyme activities were observed in the Ennore fish hepatocytes compared to Kovalam counterpart. Bioaccumulation of heavy metals was significantly higher (p < 0.05) in fish from Ennore. Hepatocytes from Ennore also featured extensive lipid-type vacuolation, increased size and membrane disruption. All the findings highlighted the value of oxidative stress biomarkers and membrane disruption as the sensitive parameters of environmental pollutant contamination and their importance in biomonitoring of aquatic ecosystems. This is also the first such attempt reported at the cellular level from South India stressing the importance of biomarkers in biomonitoring programmes using fish hepatocytes as the model system.  相似文献
