The aim of this study was to 1) detect QTL across the cattle genome that influence the incidence of clinical mastitis and somatic cell score (SCS) in Danish Holsteins, and 2) characterize these QTL for pleiotropy versus multiple linked quantitative trait loci (QTL) when chromosomal regions affecting clinical mastitis were also affecting other traits in the Danish udder health index or milk production traits. The chromosomes were scanned using a granddaughter design where markers were typed for 19 to 34 grandsire families and 1,373 to 2,042 sons. A total of 356 microsatellites covering all 29 autosomes were used in the scan. Among the across-family regression analyses, 16 showed chromosome-wide significance for the primary traits incidence of clinical mastitis in first (CM1), second (CM2), and third (CM3) lactations, and SCS. Regions of chromosomes 5, 6, 9, 11, 15, and 26 were found to affect CM and regions of chromosomes 5, 6, 8, 13, 22, 23, 24, and 25 affected SCS. Markers on chromosomes 6, 11, 15, and 26 can be used to perform marker-assisted selection on CM without a direct negative selection on milk yield, because no effects were detected on the milk traits. Comparing multi-trait models assuming either a pleiotropic QTL affecting 2 traits or 2 QTL each affecting 1 trait gave some evidence to distinguish between these models. For Bos taurus autosome 5, the most likely models were a pleiotropic QTL affecting CM2, CM3, and SCS, and a linked QTL affecting fat yield index. For Bos taurus autosome 9, the most likely model is a pleiotropic QTL affecting CM1 and CM2 at approximately 8 cM.  相似文献

The objective of this study was to test the hypothesis that lameness incidence in lactating dairy cows decreases milk production compared with controls and to determine the best method of the 5 combinations of 2 study designs (retrospective cohort and retrospective matched cohort) and 3 repeated measures statistical methods: ANOVA (with binary classification of lameness); ANOVA with a lameness index (LAMIX) as the variable of interest; and analysis of covariance (ANCOVA, controlling for mean milk production for the first 3 wk of lactation and with a binary classification of lameness). Data were from 1 dairy farm located near Ithaca, NY, and from cows that calved between January 1, 2004 and January 1, 2007. The LAMIX was defined as −1, −2, −3, −4, and −5 = 1, 2, 3, 4, and ≥5 mo before lameness was detected, respectively; 1, 2, 3, 4, and 5 = 1, 2, 3, 4, and 5 mo after diagnosis, respectively; and 0 = cows that were never lame. With the binary lameness classification analyzed by repeated measures ANOVA, there was no effect of lameness on milk yield. The model was biased because lame cows had higher milk yields before lameness compared with nonlame cows. When the LAMIX was used, milk production before lameness was greater than after lameness (3.1 ± 0.28 kg/d). Yet, point estimates generated for LAMIX were inaccurate because of the multicollinearity detected between LAMIX and week of lactation and because of the inability of adjusting the least squares means for the interaction of LAMIX and week of lactation. Therefore, the most appropriate models were the ANCOVA models (both for the matched and nonmatched retrospective-cohort designs). The estimated losses associated with lameness were 314 and 424 kg/cow per 305-d lactation, respectively, for the matched and nonmatched designs. Furthermore, high milk yield in the beginning of the lactation was a risk factor for lameness.  相似文献

The pattern of odd- and branched-chain fatty acids (OBCFA) in milk fat reflects rumen microbial activity and proportions of different rumen microbial groups. Therefore, these milk fatty acids (FA) are used to predict rumen proportions of volatile fatty acids, duodenal flow of microbial protein, and occurrence of rumen acidosis. However, current models do not correct for the potential effects of lactation stage on the level of OBCFA in milk fat. Hence, the objectives of this study were 1) to describe progressive changes related to lactation stage in concentrations of milk FA, with emphasis on the OBCFA, using the incomplete gamma function of Wood, and 2) to analyze whether lactation curves of milk FA on the one hand and milk production or milk fat content on the other hand coincide through evaluation of the correlation between the parameters of the Wood functions fitted to individual animal data. Data were collected from 2 trials in which milk FA during lactation were monitored. The first experiment was a stable trial with 2 groups of 10 cows receiving 2 dietary treatments from wk 1 to 40 of lactation. The second experiment was a grazing trial with 9 cows that were followed during the first 18 wk of lactation. Lactation curves of milk production, milk fat content, and individual milk FA were developed using the incomplete gamma function of Wood for each of the 3 dietary strategies separately. For almost all of the milk FA, lactation curve shapes were similar for all 3 dietary treatments. The OBCFA with chain lengths of 14 and 15 carbon atoms followed the lactation curves of the short- and medium-chain milk FA, which increased in early lactation. The OBCFA with chain length of 17 carbon atoms decreased during the early lactation period, following the pattern of milk long-chain fatty acids. The short- and medium-chain milk FA and OBCFA in the early lactation period seemed to be negatively correlated with the starting milk production and milk fat content, but correlations were modest. Information of milk FA lactation curves should be incorporated in predictive and classification models based on these milk FA, to improve their performance.  相似文献

A 2 × 2 factorial continuous experiment was conducted with 28 Norwegian Red dairy cows in early lactation to compare milk content of phytoestrogens when feeding ad libitum white clover (WCS) or red clover (RCS) grass silages prepared from the second and third cut without and with 10 kg/d supplementation of a standard concentrate. The cows were offered either RCS or WCS for 88 d (period 1) and thereafter a mixed red clover-white clover-grass silage for 48 d (period 2). Total dry matter intake and milk yield were not affected by forage type but increased with concentrate supplementation. Intake of isoflavones was several times greater in RCS than in WCS, whereas intake of lignans was greater in WCS. Concentrate supplementation reduced the intake of most phytoestrogens. Compared with WCS, RCS diets yielded milk with a greater content of flavonoids, whereas milk from WCS diets had greater contents of the mammalian lignans enterodiol and enterolactone. The content of the isoflavan equol was particularly high in RCS diets. There was no apparent carryover effect of clover type on milk phytoestrogen content because there was no difference in content between the silage treatments 3 wk after the cows were transferred to the same silage diet (period 2). Concentrate supplementation reduced the milk contents of the flavonoids equol, biochanin A, and daidzein and increased the content of mammalian lignans. The effects of silage type and concentrate supplementation on milk contents of the individual phytoestrogens were related to the intake of the compound or its precursor, except for the effect of concentrate on mammalian lignans, for which the intake of the known precursors was also reduced. Overall, this study shows that feeding cows with silage containing red clover increases the milk content of flavonoids at both low and high concentrate supplementation levels, and decreases the content of nonflavonoids such as mammalian lignans, when compared with silage containing white clover. The increased content of phytoestrogens in milk may be important when the health benefits of milk are studied.  相似文献

The effects of supplementation with grass silage and replacement of some corn in the concentrate with soybean meal (SBM) on milk production, and milk fatty acid (FA) profiles were evaluated in a replicated 4 × 4 Latin square study using 16 dairy cows grazing pasture composed of ryegrass, Kentucky bluegrass, and white clover. Each experimental period lasted for 3 wk. The 4 dietary treatments were PC, 20 h of access to grazing pasture, supplemented with 6 kg/d of corn-based concentrate mixture (96% corn; C); PCSB, 20 h of access to grazing pasture, supplemented with 6 kg/d of corn- and SBM-based concentrate mixture (78% corn and 18% SBM; CSB); SC, 7 h of access to grazing pasture during the day and 13 h of ad libitum access to grass silage at night, supplemented with 6 kg/d of C concentrate; and SCSB, 7 h of access to grazing pasture during the day and 13 h of ad libitum access to grass silage at night, supplemented with 6 kg/d of CSB concentrate. The concentrate mixtures were offered twice each day in the milking parlor and were consumed completely. Grass silage supplementation reduced dietary crude protein and concentration of total sugars, and dietary SBM inclusion increased dietary crude protein concentration and decreased dietary starch concentration. Milk yield and energy-corrected milk were increased by SBM supplementation of cows with access to grass silage. Milk protein concentration was lower in cows offered grass silage, regardless of whether SBM was fed. Dietary SBM inclusion tended to increase milk fat concentration. Plasma urea N was reduced by silage feeding and increased by SBM supplementation. Supplementation with grass silage overnight could represent a useful strategy for periods of lower pasture availability. Dietary inclusion of SBM in solely grazing cows had no effects on milk production and composition, exacerbated the inefficient capture of dietary N, and increased diet cost. Grass silage supplementation affected milk FA profiles, increasing both the FA derived from de novo synthesis and those derived from rumen microbial biomass, and decreasing the sum of C18 FA (mostly derived from diet or from mobilization of adipose tissue reserves). Milk fat concentrations of conjugated linoleic acid cis-9, trans-11, vaccenic acid (18:1 trans-11), and linolenic acid (18:3n-3) were unaffected by grass silage supplementation, suggesting that partial replacement of pasture by unwilted grass silage does not compromise the dietary quality of milk fat for humans.  相似文献

Antimicrobial peptides derived from food proteins constitute a new field in the combined use of antimicrobial agents in food. The best examples of milk-derived peptides are those constituted by bovine lactoferricin [lactoferrin f(17-41)] (LFcin-B) and bovine αs2-casein f(183-207). The aim of this work was to study if the antimicrobial activity of a natural compound employed in food preservation, nisin, could be enhanced by combination with the aforementioned milk-derived peptides. Furthermore, the possibility of a synergistic effect between these peptides and bovine lactoferrin (LF) against Escherichia coli and Staphylococcus epidermidis was also studied. Finally, the most active combinations were assayed against the foodborne pathogens Listeria monocytogenes and Salmonella choleraesuis. Results showed a synergistic effect when LFcin-B was combined with bovine LF against E. coli. In the same way, the combination of LFcin-B with bovine LF was synergistic against Staph. epidermidis. Bovine LF and nisin increased their antimicrobial activity when they were assayed together with bovine αs2-casein f(183-207). It is important to note the synergistic effect among LFcin-B and bovine LF, because both compounds might be simultaneously in the suckling gastrointestinal tract and could, therefore, have a protective effect on it. The other synergistic effect high-lighted is that between αs2-casein f(183-207) and nisin against L. monocytogenes because of the ability of L. monocytogenes to develop resistance to nisin.  相似文献

Lactation artificially induced (ART) by steroid hormones and natural lactation (NAT) after lambing were compared in 2 dairy sheep breeds (Manchega and Lacaune) in 2 experiments conducted during winter and spring. In experiment 1, ART ewes (14 Manchega and 9 Lacaune) were induced into lactation in winter by the standard protocol, which consisted of s.c. injections of estradiol and progesterone administered in 2 portions daily from d 1 to 7. Hydrocortisone acetate was injected s.c. daily on d 18 to 20. Milking was initiated on d 21 and continued for 13 wk. A similar group of NAT ewes was selected for the contemporary comparison of NAT vs. ART lactation. All Lacaune ewes, but only 3 of the 14 Manchega ewes (21%), were successfully induced into lactation. Despite the successful induction of lactation in Lacaune ewes, milk yield was much lower than that obtained in NAT lactation (1.23 ± 0.14 vs. 2.51 ± 0.15 L/ d). Milk composition from wk 5 to 13 did not differ between groups, except for whey protein, which was greater in ART than in NAT ewes (1.47 vs. 1.25%). In experiment 2, 19 Manchega ewes were divided into 2 groups and induced into lactation in spring by using the standard induction protocol, similar to that used in experiment 1 (control, n = 9), or the standard protocol modified with bovine somatotropin (bST, 250 mg/ewe on d 11; n = 10). Manchega ewes had an improved response to the standard protocol of lactation induction in spring compared with winter. Milk yield in bST-treated Manchega ewes was 98% greater than that in control ewes (402 ± 85 vs. 203 ± 86 mL/d). The use of bST during mammogenesis did not affect milk composition. In conclusion, marked differences between Manchega and Lacaune dairy ewes were observed in their response to lactation induction when using the standard protocol during different photoperiod conditions. The Manchega ewes were unable to establish lactation in winter but were able to do so in spring. The response to lactation induction in dairy ewes seems to be related to their endogenous levels of prolactin and growth hormone, the use of which should be explored more deeply in future research.  相似文献

Whey protein concentrate (WPC) has many applications in the food industry. Previous research demonstrated that treatment of whey proteins with high hydrostatic pressure (HHP) can enhance solubility and foaming properties of whey proteins. The objective of this study was to use HHP to improve functional properties of fresh WPC, compared with functional properties of reconstituted commercial whey protein concentrate 35 (WPC 35) powder. Fluid whey was ultrafiltered to concentrate proteins and reconstituted to equivalent total solids (8.23%) as reconstituted commercial WPC 35 powder. Solutions of WPC were treated with 300 and 400 MPa (0- and 15-min holding time) and 600 MPa (0-min holding time) pressure. After HHP, the solubility of the WPC was determined at both pH 4.6 and 7.0 using UDY and BioRad protein assay methods. Overrun and foam stability were determined after protein dispersions were whipped for 15 min. The protein solubility was greater at pH 7.0 than at pH 4.6, but there were no significant differences at different HHP treatment conditions. The maintenance of protein solubility after HHP indicates that HHP-treated WPC might be appropriate for applications to food systems. Untreated WPC exhibited the smallest overrun percentage, whereas the largest percentage for overrun and foam stability was obtained for WPC treated at 300 MPa for 15 min. Additionally, HHP-WPC treated at 300 MPa for 15 min acquired larger overrun than commercial WPC 35. The HHP treatment of 300 MPa for 0 min did not improve foam stability of WPC. However, WPC treated at 300 or 400 MPa for 15 min and 600 MPa for 0 min exhibited significantly greater foam stability than commercial WPC 35. The HHP treatment was beneficial to enhance overrun and foam stability of WPC, showing promise for ice cream and whipping cream applications.  相似文献

The levels of trace elements are an important component of safety and quality of milk. While certain elements such as chromium are essential at low levels, an excess can result in deleterious effects on human health. International quality control standards for milk are published by the Codex Alimentarious Commission and levels of heavy metals in milk intended for human consumption are routinely monitored. This paper describes a new method for demonstrating the levels of V, Cr, Mn, Sr, Cd and Pb in raw cow’s milk, using an ICP-MS. Samples (n = 24) of raw cow’s milk were collected from dairy farms close to mines in Gauteng and North West Provinces of South Africa. In order to destroy organic matrix, each freeze dried milk sample was mineralised by using a microwave assisted digestion procedure. Concentrations of trace elements in digested milk samples were measured by ICP-MS. A whole milk powder reference material (NIST SRM 8435) was used to evaluate the accuracy of the proposed method. It was found that the levels of V, Cr, Mn, Sr, Cd and Pb obtained using the new method showed concordance with certified values.  相似文献
