Diporphyrin tweezer for multichannel spectroscopic analysis of enantiomeric excess

Chiral 1,1’-binaphthyl-linked diporphyrin ‘tweezers’ (R)-1/(S)-1 and the corresponding zinc(II) complexes (R)-2/(S)-2 were prepared as chiral host molecules, and their utility for chiral analyses (especially enantiomeric excess (ee) determinations) were evaluated. Tris(1-n-dodecyl)porphyrins were used for the first time as the interacting units. Host capabilities of the diporphyrin tweezers were investigated by titrations with (R,R)- and (S,S)-cyclohexane-1,2-diamine (CHDA). The host molecules could be used as multichannel probes of ee by using UV-vis, circular dichroism (CD), fluorescence emission and ¹H nuclear magnetic resonance (¹H-NMR) methods. Chiral configurations could also be differentiated using CD or ¹H-NMR spectroscopy. All three optical techniques give good resolution of ee with reasonable sensitivity considering the low concentrations used (ca. 10⁻⁶ mol·L⁻¹). The ee determination of CHDA enantiomers using NMR spectroscopy is also possible because of the reasonably well separated resonances in the case of (R,R)- and (S,S)-CHDA. Non-metallated (R)-1/(S)-1 hosts could not be used to detect chiral information in a strongly acidic chiral guest. This work demonstrates the utility of 1,1’-binapthyl-linked chiral hosts for chiral analysis of ditopically interacting enantiomers.     

靶向CD93分子探针制备及其生物学实验研究

CD93分子作为一种新型新生血管生成激活剂,有可能成为肿瘤监测的重要分子靶点,因此制备125I标记抗CD93单抗(125I-anti-CD93 mAb),研究其在荷瘤鼠体内生物学分布及磷屏显像特点,探讨其对A549肺癌靶向性及CD93阳性肿瘤监测的可行性。本文制备125I-anti-CD93 mAb及125I-IgG并进行鉴定;建立A549肺癌荷瘤小鼠模型,经尾静脉注射125I标记抗CD93单抗,并设125I-IgG为对照组,注药后不同时间进行生物学分布及磷屏放射自显影;肿瘤组织行HE染色及CD93免疫组织化学染色;采用统计软件对定量数据(x+s)进行统计学处理,组间数据用t检验,P<0.05为差异有统计学意义。结果表明,125I-anti-CD93 mAb及125I-IgG标记率、放射性比活度、放化纯度分别为91.37%和90.24%,1096.44 MBq/mg和1082.88 MBq/mg,96.49%和94.82%,放置72 h后两标记物的稳定性仍>90%,两者间无统计学差异(生理盐水组P=0.93,t=0.09,血清组P=0.13,t=1.90)。荷瘤鼠生物学分布结果显示125I-anti-CD93 mAb主要经肝肾代谢,注射后48 h时肿瘤组织放射性摄取率为(6.42±0.71)%ID/g,T/NT(瘤/对侧肌肉)摄取比值为4.45±0.86,显著高于对照组125I-IgG组(2.45±0.33/1.71±0.24),P<0.05,t=3.07和P<0.01,t=5.10。动态全身磷屏自显影结果可见,24 h两组小鼠轮廓清楚,48 h时实验组肿瘤显像最清晰,肿瘤放射性摄取比(肿瘤/对侧肌肉)为3.34±0.18,明显高于对照组(1.62±0.19),P<0.01,t=6.52。免疫组化染色结果证实肿瘤高表达CD93。125I-anti-CD93 mAb制备简便,对A549肺癌组织靶向性好,有望成为CD93阳性肿瘤监测的新型分子探针。     

掺粉煤灰水泥胶砂的正交试验研究

采用正交试验方法,研究了粉煤灰掺量、细度和Na2SO4掺量对水泥胶砂试块抗折、抗压强度影响规律。结果表明:粉煤灰掺量对水泥胶砂试块的7d抗折、抗压强度影响最显著;粉煤灰掺量和粉煤灰细度对28d抗折、抗压强度影响都比较显著;而Na2SO4掺量对7、28d抗折、抗压强度影响不大。     

提高载重尼龙轮胎速度性能的研究

分析了载重汽车斜交尼龙轮胎在高等级公路上使用损坏的原因，阐述了提高速度性能的技术措施，从而使１１．００－２０１６ＰＲ轮胎全面达到攻关指标。耐久性最高达１５８．７２ｈ，高速性能通过１１０ｋｍ．ｈ＾－１２ｈ均超过耐久性１００ｈ，高速１００ｋｍ．ｈ＾－１２ｈ的攻关指标。     

一种用于折叠翼飞机的行星齿轮机构动力学分析

针对折叠翼飞机的折叠机构,在齿轮啮合机构基础上提出一种行星齿轮传动机构实现机翼折叠,利用ADAMS软件对两种机构进行了动力学仿真分析.通过对两种机构的对比分析,证明针对折叠翼飞机设计的行星齿轮机构较原齿轮啮合机构能够有效减小齿轮间的接触力,改善机构的运动平稳特性,降低对驱动电机转矩的要求,有利于提高机构的效率.经仿真分析,证明所设计的行星齿轮传动机构可作为折叠翼飞机的折叠传动机构方案.     

人CD271基因的克隆及原核表达

目的克隆人CD271基因,并在大肠杆菌中表达。方法采用RT-PCR技术从人REH细胞系中扩增CD271基因,将其克隆入pET22b表达载体,转化大肠杆菌BL21(DE3),IPTG诱导表达,并对表达产物进行纯化及Western blot鉴定。结果重组表达质粒pET22b/CD271经双酶切鉴定,可见1197bp的目的基因条带。表达产物经SDS-PAGE分析,在相对分子质量约75000处可见表达条带。IPTG浓度为0.5mmol/L时,目的蛋白表达量最高。纯化后蛋白纯度为82.5%。Western blot检测表明纯化后的蛋白具有良好的反应原性。结论已成功克隆了人CD271基因,并在大肠杆菌中获得表达。

CD146 Expression Correlates with Epithelial-Mesenchymal Transition Markers and a Poor Prognosis in Gastric Cancer

CD146 has been regarded as a novel potential therapeutic target for multiple cancers. The aim of the study was to investigate the expression of CD146 in gastric cancer and evaluate its clinical-pathological and prognostic significance. The expression of CD146 and three epithelial-mesenchymal transition (EMT)-related proteins (E-cadherin, β-catenin and vimentin) was examined in 144 gastric cancers by immunohistochemistry. Fifty-nine cases (41.0%) were defined as positive for CD146 expression. We found that CD146 expression correlated positively with lymph node involvement and a poor prognosis, and retained an independent prognostic factor for gastric cancer patients. Furthermore, positive expression of CD146 was strongly associated with loss of the epithelial marker E-cadherin and acquisition of the expression of the mesenchymal markers nuclear β-catenin and vimentin. These findings suggest that CD146 might promote EMT and progression in gastric cancer, and thus may be a potential therapeutic target for patients with gastric cancers.     

Butanol isomer separation using polyamide-imide/CD mixed matrix membranes via pervaporation

Mixed matrix membranes (MMMs) comprising polyamide-imide (PAI) and α-, β- or γ-cyclodextrin (CD) have been investigated experimentally and computationally for isomeric n-butanol/tert-butanol (n-BuOH/t-BuOH) separation via pervaporation. Consistent with molecular simulation, experimental results show that the CD inclusion ability and butanol discrimination ability are dependent on both CD cavity size and butanol molecular size. The PAI membrane incorporated with α-CD has the smallest cavity and has the highest discrimination ability for the n-BuOH/t-BuOH pair but with a low butanol flux. The mixed matrix membrane embedded with γ-CD has the lowest selectivity and the highest flux. The PAI/β-CD membrane has a comparable selectivity and flux, and exhibits preferential sorption and diffusion selectivity toward n-BuOH. A maximum separation factor of 1.53 with a corresponding flux of 4.4 g/m² h are obtained at an optimal β-CD loading of 15 wt%. Further increments in the CD content eventually lead to a decrease in separation performance because of CD agglomeration and severe phase separation. To better understand the influence of CD on the separation performance of mixed matrix membranes, SEM, FTIR and XRD have been employed for membrane characterizations. The effect of n-butanol/t-butanol ratio in the feed composition has also been studied. It is found that both flux and separation factor decrease with increasing n-butanol content in the feed. The decline is attributed to the change in total vapor pressure at the upstream and the mutual drag effect of isomeric butanol molecules.
