The objective of this study was to control the survival or biofilm formation of Cronobacter spp. on stainless steel surfaces using Paenibacillus polymyxa. The antibacterial activity of a cell‐free culture supernatant (CFCS) of P. polymyxa against Cronobacter spp. was found to vary with P. polymyxa incubation time. Maximum activity occurred when P. polymyxa was incubated at 25 or 30 °C for 96 h. When the CFCS was introduced to Cronobacter spp. adhered to stainless steel strips at 25 °C for up to 72 h, the CFCS successfully inhibited Cronobacter biofilm formation. Additionally, stainless steel surfaces with a preformed P. polymyxa biofilm were exposed to Cronobacter spp. suspensions in PBS or 0.1% peptone water at 3, 5, or 7 log CFU/mL to facilitate its attachment. The Cronobacter population significantly decreased on this surface, regardless of inoculum level or carrier, when the P. polymyxa biofilm was present. However, the microbial population decreased within 6 h and remained unchanged thereafter when the surface was immersed in an inoculum suspended in 0.1% peptone water at 5 or 7 log CFU/mL. These results indicate that P. polymyxa is able to use a promising candidate competitive‐exclusion microorganism to control Cronobacter spp.  相似文献

Incorporating spent coffee grounds (SCGs), a by‐product from coffee brewing, in growth substrate of beneficial edible mushrooms is an approach that has to be further studied due to its potential positive outcomes: environmental impact mitigation, production costs reduction and beneficial impact on consumer health. Hence, cultivation of Pleurotus citrinopileatus and Pleurotus salmoneo‐stramineus was tested using SCG which enabled maximum production yield of P. citrinopileatus which was of 25.1% (w/w). Variable antidiabetic potential was observed between aqueous and enzymatic extracts (3.8%–29% inhibition) regardless species and substrates, whereas aqueous extract of P. citrinopileatus grown in substrate without SCG stood out presenting the highest antioxidant activity and inhibition activity of angiotensin I‐converting enzyme (IC50 = 123 μg mL?1). Ethanolic and aqueous extracts of both Pleurotus species grown in the presence or absence of SGC proved to be an interesting prebiotic source for growth of Bifidobacterium animalis Bo in comparison with fructooligosaccharides (FOS).  相似文献

Logs from the base of Douglas fir (Pseudotsuga menziesii) trees cut in October 1993 were randomly assigned to one of three treatment groups: (1) wet logs—cut from the fallen tree and left exposed to rain, (2) dry logs—cut from the fallen tree, placed on blocks, and protected from rain under a plastic tent, and (3) crown logs—left attached to the fallen tree with its branches intact and exposed to rain. The following May, ethanol concentrations were highest in the phloem and sapwood of wet logs (0.24 and 0.35 mol/g fresh wt, respectively). Ethanol concentrations in tissues from dry and crown logs were similar to each other (ranging from 0.002 to 0.03 mol/g fresh wt), but were significantly lower than in wet logs. It appears that rain absorbed by the outer bark of wet logs creates a barrier to gas exchange between living tissues and the atmosphere, which facilitates the development of hypoxic conditions necessary for ethanol synthesis and accumulation. Branches on crown logs exposed to rain help maintain low ethanol concentrations in the log tissues; we discuss several potential mechanisms to explain this response. By early September, the densities of Gnathothrichus spp. gallery entrance holes were high on wet logs (21.5/m2) and low on dry (2.5/m2) and crown logs (5.8/m2), indicating their preference for logs with higher ethanol concentrations. Protecting logs from rain will significantly reduce ethanol concentrations and the density of ambrosia beetle galleries. Leaving branches attached to logs will produce similar results, but its effectiveness may vary depending on the environmental conditions. Host selection by secondary scolytid beetles that use ethanol as a kairomone can be manipulated and possibly managed by controlling the production of ethanol in the host resource.  相似文献

Ticks and mites respond to a limited spectrum of stimuli in their search for hosts and mates. Airborne chemical signals include carbon dioxide, ammonia, organic acids, terpenoids, 2,6-dichlorophenol, and other phenolic compounds. These are detected primarily by sensilla in and adjacent to Haller's organ. Most ixodid species examined have one or more multiporose sensilla that detect such volatiles. These olfactoreceptors enable the ticks to respond to remote volatile chemicals from hosts and from the other ticks, e.g., sex pheromones. Other sensilla, probably mechanogustatory in function, also occur on the tarsi. Gustatory sensilla on the palps detect assembly pheromones that enable ticks and mites to respond to conspecific or heterospecific chemical stimuli in their environment. Responses to those stimuli in ticks result in clustering, i.e., arrestant behavior. Arrestant behavior also occurs in certain mites. Finally, cheliceral chemosensilla enable ticks to recognize specific phagostimulants in host blood, e.g., ATP and glutathione, that stimulate feeding. InDennacentor variabilis andD. andersoni, these same cheliceral chemosensilla recognize species-specific genital sex pheromones in the vulvae of conspecific mates, without which they do not copulate.  相似文献

The food selection, growth, and fecundity of insect herbivores are largely dictated by the chemical composition and nutritive values of plant foliage. We studied the host-use efficiency of larvae of the generalist moth, Operophtera brumata (Lepidoptera: Geometridae) on three chemically divergent but nutritively similar willows (Salix spp.). The 4th instars were able to use the salicylate-free leaves of S. phylicifolia efficiently. Growth was slightly reduced on S. pentandra, which contained a moderate level of acetylated salicylates. The high concentration of salicylates found in the leaves of S. myrsinifolia seemed to provide efficient protection against non-specialized O. brumata. We also studied assimilation of nutrients and degradation of salicylates and other secondary compounds in the digestive tract of O. brumata larvae. Neither the assimilation of nitrogen nor of carbon were affected by secondary chemicals of ingested food. Salicylates were shown to be degraded to salicin and catechol, while further degradation of salicin to saligenin was rather slow. In an artificial diet experiment, we showed that two degradation products of salicylates, catechol and saligenin markedly reduced the growth of the larvae. Neither salicin nor chlorogenic acid affected larval growth. We conclude that salicylates reduced the growth of the generalist winter moth mainly by feeding deterrence caused by 6-hydroxy-2-cyclohexenone and catechol. Compared to the deleterious effects of salicylates the effects of other secondary compounds were minor.  相似文献

BACKGROUND: A bioelectrochemical process for trichloroethene (TCE) dechlorination was developed. In this new process, a solid‐state electrode polarized to ?450 mV versus the standard hydrogen electrode (SHE), in combination with a redox mediator (i.e., methyl viologen, MV) is employed as an electron donor for the microbial reductive dechlorination of TCE. In this study we compared the performance of the process with the redox mediator immobilized at the surface of electrodes or dissolved in the bulk liquid, using both a culture highly enriched in Desulfitobacterium spp., capable of dechlorinating TCE to cis‐dichloroethene (cis‐DCE), and a culture highly enriched in Dehalococcoides spp. capable of dechlorinating cis‐DCE to ethene. RESULTS: Short‐term potentiostatic (?450 mV versus SHE) experiments showed that TCE or cis‐DCE was dechlorinated both in the presence of soluble (500 µmol L?1) and immobilized MV. However, TCE or cis‐DCE dechlorination rates with MV‐modified electrodes were remarkably lower than with soluble MV. Both cultures produced significant amounts of H2 in the presence of electrically reduced, soluble MV, whereas no H2 was produced when the mediator was immobilized at the electrode surface, regardless of the potential applied to the electrode, in the range ?425 to ?500 mV versus SHE. CONCLUSIONS: The process, operated with modified electrodes, supports the microbial dechlorination of TCE to ethene. Immobilization not only allows retention of the mediator within the system, but also increases process efficiency by preventing bioelectrochemical H2 formation. The starch content of four edible aroid (Araceae) cultivars was determined using acid and enzyme methods of hydrolysis. Hydrolysates were analysed using ferricyanide reduction, glucose oxidase and high performance liquid chromatography methods. Differences in starch values were obtained between the method of sugar analysis used when acid hydrolysis of samples was employed. The results obtained using enzyme hydrolysis, however, showed no significant differences between the methods of sugar analysis used. The results of these experiments indicate that, to obtain accurate starch values for edible aroids using the least expensive options, it is recommended that acid hydrolysis and the glucose oxidase-peroxidase glucose-specific assay methods or enzyme hydrolysis and ferricyanide sugar analysis be used. Starch degradation of ungelatinised samples by enzymic hydrolysis was very limited and showed considerable cultivar variation.  相似文献

The purpose of this study was a detailed characterization of Shewanella sp. O23S, a strain involved in arsenic transformation in ancient gold mine waters contaminated with arsenic and other heavy metals. Physiological analysis of Shewanella sp. O23S showed that it is a facultative anaerobe, capable of growth using arsenate, thiosulfate, nitrate, iron or manganite as a terminal electron acceptor, and lactate or citrate as an electron donor. The strain can grow under anaerobic conditions and utilize arsenate in the respiratory process in a broad range of temperatures (10–37 °C), pH (4–8), salinity (0%–2%), and the presence of heavy metals (Cd, Co, Cr, Cu, Mn, Mo, Se, V and Zn). Under reductive conditions this strain can simultaneously use arsenate and thiosulfate as electron acceptors and produce yellow arsenic (III) sulfide (As2S3) precipitate. Simulation of As-removal from water containing arsenate (2.5 mM) and thiosulfate (5 mM) showed 82.5% efficiency after 21 days of incubation at room temperature. Based on the obtained results, we have proposed a model of a microbially mediated system for self-cleaning of mine waters contaminated with arsenic, in which Shewanella sp.
