Lameness has been extensively studied in lactating cows, whereas few studies have reported on lameness during the dry period. We conducted a prospective longitudinal study to describe the epidemiology of lameness during the dry period and to identify risk factors associated with onset, cure, and chronic cases of lameness. A total of 455 cows from 6 freestall commercial dairy farms were enrolled at 9 wk before calving and gait scored weekly until calving using a 5-point scale. A subset of cows was also followed fortnightly after calving to measure the association between lameness during the dry period and lameness during early lactation. Body condition score (BCS) was assessed in a 5-point scale using increments of 0.5. Hoof-trimming records, parity, and previous lactation milk production were retrieved from farm's database. Cows were considered sound when 2 consecutive scores were ≤2 and lame when 2 consecutive scores = 3, or any assessment with score >3; when in a sequence of scores only one score = 3 (or ≤2), the cow was considered sound (or alternatively lame). Following this lameness definition, we derived weekly lameness status for each cow and calculated the number of new cases of lameness, the number of cure cases and the number of chronic cases. The incidence rate of lameness cases during the dry period was 8.2 lameness cases/100 cow per wk, whereas cure rate was 7.1 cure cases/100 cow per wk; at the end of the dry period 50% of cows had developed lameness and 36% were cured. Multilevel logistic regression models using farm as random effect were fitted to assess (1) the association between being lame in wk 2 or 8 postcalving with being lame in the last week precalving, (2) risk factors for lameness onset, (3) risk factors for lameness cure, and (4) risk factors for chronic lameness. Cows that were lame in the week immediately before calving were more likely to be lame in wk 2 and 8 after calving. We found that the interaction between parity and hoof-trimming before dry-off was associated with lameness onset; primiparous cows that were trimmed before dry-off had lower odds of developing lameness, whereas the opposite was found for multiparous cows. The same interaction was also associated with the odds of chronic lameness. Cows that were diagnosed with noninfectious hoof lesions compared with cows that were not diagnosed with hoof lesions before dry-off, and cows that had BCS <3 compared with cows with BCS 3.0 to 3.5 at dry-off had higher odds of chronic lameness. Conversely, primiparous cows and cows with BCS 3.0 to 3.5 had higher odds of curing lameness during the dry period. Our results suggest that the dry period may be a period of high risk for lameness development and that hoof-trimming before dry-off may not be effective for all cows.  相似文献

The aim of this study was to investigate the feasibility of using mid-infrared (MIR) spectroscopy analysis of milk samples to increase the power and precision of genome-wide association studies (GWAS) for milk composition and to better distinguish linked quantitative trait loci (QTL). To achieve this goal, we analyzed phenotypic data of milk composition traits, related MIR spectra, and genotypic data comprising 626,777 SNP on 5,202 Holstein, Jersey, and crossbred cows. We performed a conventional GWAS on protein, lactose, fat, and fatty acid concentrations in milk, a GWAS on individual MIR wavenumbers, and a partial least squares regression (PLS), which is equivalent to a multi-trait GWAS, exploiting MIR data simultaneously to predict SNP genotypes. The PLS detected most of the QTL identified using single-trait GWAS, usually with a higher significance value, as well as previously undetected QTL for milk composition. Each QTL tends to have a different pattern of effects across the MIR spectrum and this explains the increased power. Because SNP tracking different QTL tend to have different patterns of effect, it was possible to distinguish closely linked QTL. Overall, the results of this study suggest that using MIR data through either GWAS or PLS analysis applied to genomic data can provide a powerful tool to distinguish milk composition QTL.  相似文献

The aim of the present study was to assess genetic variation and heritability of a novel indicator of udder health, milk differential somatic cell count (DSCC), which represents the percentage of neutrophils plus lymphocytes in the total somatic cell count (SCC). Furthermore, we estimated genetic and phenotypic correlations of DSCC with other milk traits routinely measured in Italian Holstein cows. Besides DSCC, test-day data included milk yield, composition traits (i.e., fat, protein, casein, and lactose percentages), pH, milk urea nitrogen, and SCC. After editing, the final data set included 10,709 test-day records of 5,142 cows in 299 herds. Mean of DSCC was 62.07%, which means that macrophages were approximately 38% of total SCC. Comparing our results with the literature offered compelling evidence of the importance of acquiring information about the proportion of the different cell types in milk to better define the udder health status. In addition, our analysis revealed, for the first time, that DSCC is a heritable trait, and heritability (0.08 ± 0.02) was higher than that of traditional somatic cell score (0.04 ± 0.02). Nevertheless, heritability of DSCC is still low compared with milk yield and quality traits. Single-trait analysis within parity showed that DSCC was less heritable in primiparous than in multiparous cows, whereas bivariate analysis confirmed that DSCC and somatic cell score were 2 different traits, as their genetic and phenotypic correlations differed from unity. From a genetic point of view, the DSCC was positively weakly associated with milk yield, lactose percentage, and milk urea nitrogen, and negatively associated with pH. Our findings contributed to the understanding of the genetic background of DSCC and are a precursor to the potential use of DSCC in breeding programs to enhance cow resistance to mastitis. However, further research is needed to determine the weight this novel trait should receive in a selection program aimed to reduce udder health problems.  相似文献

The genetic correlations (ra) of milk lactose percentage (LP), lactose yield (LY), and ratios of LP to other milk solids with udder, metabolic, and fertility disorders have not been assessed in dairy cattle so far. To evaluate the potential role of milk lactose as indicator of cow health, 142,285 lactation records of 84,289 Austrian Fleckvieh cows were analyzed with univariate and bivariate animal models. Milk traits were on a 150-d basis and health traits were coded as binary (0/1). Other than LP and LY, 3 new phenotypes were defined and included in the present study, namely the lactose-to-fat, lactose-to-protein, and lactose-to-solids ratios. The most heritable trait was LP (0.566 ± 0.008) and heritability of LY was much lower (0.145 ± 0.005). Heritability estimates close to 0.50 were assessed for the ratios. The frequency of health disorders was higher in multiparous cows yielding milk with low LP (≤4.553%) compared with cows yielding milk with high LP (≥5.045%). Heritabilities of health traits were in the expected ranges, with the highest estimate for ovarian cysts (CYS; 0.037 ± 0.004) and the lowest for retained placenta (0.005 ± 0.001). Mastitis (MAS) genetically correlated with LY (0.518 ± 0.057); considering that the amount of synthesized lactose is the key regulator of milk volume, this result confirmed that high-producing cows are more genetically susceptible to MAS than low-producing animals. Similar to MAS, ketosis (KET) was also positively genetically associated with LY (0.420 ± 0.077) and a weak and unfavorable ra between KET and lactose-to-protein ratio was estimated (0.159 ± 0.077). The ra of LY with milk fever (MFV) and CYS were approximately 0.20. The ra of LP with MAS, KET, and MFV were negative (?0.142 on average), supporting the idea that LP is a potential health indicator. Genetic correlations between health traits ranged from zero (retained placenta with MAS and CYS) to 0.463 ± 0.090 (MAS and MFV). Results of the present study suggest that LP has potentiality to be used as indicator trait to improve udder health in Austrian Fleckvieh population.  相似文献

Machine vision technology has been used in automated body condition score (BCS) classification of dairy cows. The current vision-based classifications use information acquired from a limited number of body regions of the cow. Our study aimed to improve automated BCS classification by including multiple body condition–related features extracted from 3 viewpoints in 8 body regions. The data set of this study included 44 lactating cows with their BCS evenly distributed over the scale of BCS from 1.5 to 4.5 units. The body images of these cows were recorded over 2 consecutive days using 3-dimensional cameras positioned to view the cow from the top, right side, and rear. Each image was automatically processed to identify anatomical landmarks on the body surface. Around these anatomical landmarks, the bony prominences and body surface depressions were quantified to describe 8 body condition–related features. A manual BCS of each cow was independently assigned by 2 trained assessors using the same predefined protocol. With the extracted features as inputs and manual BCS as the reference, we built a nearest-neighbor classification model to classify BCS and obtained an overall classification sensitivity of 0.72 using a 10-fold cross-validation. We conclude that the sensitivity of automated BCS classification has been improved by expanding the selection of body condition–related features extracted from multiple body regions.  相似文献

Hot-iron disbudding, the process of cauterizing the horn buds of calves or goat kids at an early age to prevent horn growth, is routinely practiced in dairy production. The wounds take weeks to heal and are painful to touch throughout this time. Possible strategies to hasten the healing of disbudding wounds are not well understood, but the type of iron used may be an important factor to consider. When evaluating strategies to hasten healing, a within-subjects design may be preferable, but laterality effects might act as a potential source of variation and confounding in this type of experiment. Our objectives were to compare healing after disbudding with 2 commercially available irons, and to determine whether wounds healed differently on the left versus the right side of the head. Ten Holstein calves 4 to 10 d of age were disbudded using the Rhinehart X50A electric disbudder (Rhinehart Development Corp., Spencerville, IN) on one horn bud and the Portasol gas disbudder (Portasol USA, Elmira, OR) on the other; side (left vs. right) was balanced between treatments. We scored wounds daily for the presence of 6 tissue types: attached necrotic tissue, detached necrotic tissue, exudate, granulation, crust, and epithelium. Surface temperature and size of the wound were measured twice-weekly using thermal and digital photographs, respectively. The type of iron used did not affect latency to re-epithelialize, which took on average (mean ± standard deviation) 53 ± 3 d and 55 ± 3 d for Portasol and Rhinehart wounds, respectively (range: 40–70 d). However, compared with Portasol wounds, those from the Rhinehart had fewer days of granulation tissue and tended to have more days of detached necrotic tissue. The Portasol tip had a smaller total surface area than the Rhinehart, which may have resulted in a less severe burn, causing the necrotic tissue to fall off sooner. The left side tended to re-epithelialize faster than the right side (mean ± standard error: left 51 ± 3 d; right 57 ± 3 d) and have fewer days of crust. Left-sided wounds were also cooler and tended to be smaller than those on the right. To assess the external validity of these laterality effects in our primary experiment (experiment A), we analyzed wound healing data from 2 other disbudding studies, one on calves (experiment B) and one on goat kids (experiment C). We observed laterality effects in the opposite direction in Experiment B, but negligible effects in experiment C, indicating that the differences in laterality had low external validity; the biological meaning of this asymmetry is unclear. Nonetheless, if using a within-subjects design, asymmetries in wound healing should be considered to avoid confounding effects. In conclusion, wounds from both irons took 7 to 8 wk to heal, on average; other strategies to accelerate healing should be explored.  相似文献

Mycobacterium avium ssp. paratuberculosis (MAP) is the causative agent of Johne's disease, an enteric infection of ruminants that causes significant economic burden for dairy and beef producers. Efforts to control MAP in endemic herds typically focus on herd management practices such as limiting exposure or early culling of infected animals and, occasionally, vaccination. The ionophore monensin sodium may have protective effects against MAP both in vivo and in vitro; however, this has not been thoroughly evaluated experimentally. Using a direct intestinal MAP challenge model, we have observed similarities regarding persistence of MAP in tissues and apparent resilience to infection compared with experimental oral infection or natural disease. Here we sought to investigate the effects of oral monensin supplementation in experimentally MAP-infected calves. We examined the persistence of MAP in the intestinal tissues, MAP-induced intestinal inflammation, fecal MAP shedding, and seroconversion using a commercial serologic assay. Monensin-supplemented MAP-infected calves demonstrated evidence for resilience to MAP infection earlier in this study compared with monensin-free MAP-infected calves. However, statistical modeling did not identify a significant effect of monensin on outcomes of infection, and more work is required to understand how monensin affects early tissue colonization of MAP in calves.  相似文献

Staphylococcus aureus is a major pathogen in humans and animals. In cattle, it is one of the most important agents of mastitis, causing serious costs in the dairy industry. Early diagnosis and adequate therapy are therefore 2 key factors to deal with the problems caused by this bacterium, and benzylpenicillin (penicillin) is usually the first choice to treat these infections. Unfortunately, penicillin resistance testing in bovine S. aureus strains shows discrepant results depending on the test used; consequently, the best method for assessing penicillin resistance is still unknown. The aim of this study was therefore to find a method that assesses penicillin resistance in S. aureus and to elucidate the mechanisms leading to the observed discrepancies. A total of 146 methicillin-sensitive S. aureus strains isolated from bovine mastitis were tested for penicillin resistance using a broth microdilution [minimum inhibitory concentration (MIC)] and 2 different disk diffusion protocols. Furthermore, the strains were analyzed for the presence of the bla operon genes (blaI, blaR1, blaZ) by PCR, and a subset of 45 strains was also subjected to whole genome sequencing (WGS). Discrepant results were obtained when penicillin resistance of bovine S. aureus was evaluated by disk diffusion, MIC, and PCR methods. The discrepancies, however, could be fully explained by WGS analysis. In fact, it turned out that penicillin resistance is highly dependent on the completeness of the bla operon promotor: when the bla operon was complete based on WGS analysis, all strains showed MIC ≥1 µg/mL, whereas when the bla operon was mutated (31-nucleotide deletion), they were penicillin sensitive except in those strains where an additional, bla operon-independent resistance mechanism was observed. Further, WGS analyses showed that penicillin resistance is truly assessed by the MIC assay. In contrast, caution is required when interpreting disk diffusion and PCR results.  相似文献

Among Italian dairy cattle, the Holstein is the most reared breed for the production of Parmigiano Reggiano protected designation of origin cheese, which represents one of the most renowned products in the entire Italian dairy industry. In this work, we used a medium-density genome-wide data set consisting of 79,464 imputed SNPs to study the genetic structure of Italian Holstein breed, including the population reared in the area of Parmigiano Reggiano cheese production, and assessing its distinctiveness from the North American population. Multidimensional scaling and ADMIXTURE approaches were used to explore the genetic structure among populations. We also investigated putative genomic regions under selection among these 3 populations by combining 4 different statistical methods based either on allele frequencies (single marker and window-based) or extended haplotype homozygosity (EHH; standardized log-ratio of integrated EHH and cross-population EHH). The genetic structure results allowed us to clearly distinguish the 3 Holstein populations; however, the most remarkable difference was observed between Italian and North American stock. Selection signature analyses identified several significant SNPs falling within or closer to genes with known roles in several traits such as milk quality, resistance to disease, and fertility. In particular, a total of 22 genes related to milk production have been identified using the 2 allele frequency approaches. Among these, a convergent signal has been found in the VPS8 gene which resulted to be involved in milk traits, whereas other genes (CYP7B1, KSR2, C4A, LIPE, DCDC1, GPR20, and ST3GAL1) resulted to be associated with quantitative trait loci related to milk yield and composition in terms of fat and protein percentage. In contrast, a total of 7 genomic regions were identified combining the results of standardized log-ratio of integrated EHH and cross-population EHH. In these regions candidate genes for milk traits were also identified. Moreover, this was also confirmed by the enrichment analyses in which we found that the majority of the significantly enriched quantitative trait loci were linked to milk traits, whereas the gene ontology and pathway enrichment analysis pointed to molecular functions and biological processes involved in AA transmembrane transport and methane metabolism pathway. This study provides information on the genetic structure of the examined populations, showing that they are distinguishable from each other. Furthermore, the selection signature analyses can be considered as a starting point for future studies in the identification of causal mutations and consequent implementation of more practical application.  相似文献

Monitoring and detecting individual cows' liveweight (LW) and liveweight change (LWC) are important for estimation of nutritional requirements and health management, and could be useful to measure short-term feed intake, water consumption, defecation, and urination. Walk-over weighing (WOW) systems can facilitate measurements of LW for these purposes, providing automated LW recorded at different times of the day. We conducted a field study to (1) quantify the contribution of feed and water intake, as well as urine and feces excretions, to short-term LWC and (2) determine the feasibility of stationary and WOW scales to detect subtle changes in LW as a result of feed and water intake, urination, and defecation. In this experiment, 10 cows walked through a WOW system and then stood individually on a stationary scale collecting weights at 10 and 3.3 Hz, respectively. Cows were offered 4 kg of feed and 10 kg of water on the stationary scale. For each animal, LW before and after eating and drinking was then calculated using different approaches. Liveweight change was calculated as the difference between the initial and final LW before and after eating and drinking for each statistical measure. The weights of feed intake, water consumption, urination, and defecation were measured and used as predictors of LWC. Urine and feces were collected from individual cows while the cow was on the scale, using a container, and weighed separately. The agreement between LWC measured using either stationary or WOW scales was assessed to determine the sensitivity of the scales to detect subtle changes in LW using the coefficient of determination (R2), Lin's concordance correlation coefficient (CCC), and mean bias. The prediction model showed that most of the regression coefficients were not significantly different from +1.0 for feed and water, or ?1.0 for urine and feces. The R2 and CCC values demonstrated a satisfactory agreement between calculated and stationary LWC and values ranged from 0.60 to 0.92 and 0.71 to 0.94, respectively. A moderate agreement was achieved between calculated and automated LWC with R2 and Lin's CCC values of 0.45 to 0.63 and 0.60 to 0.74, respectively. Therefore, results demonstrated that new algorithms and data processing methods need to be continuously explored and improved to obtain accurate measurements of LW to measure changes in LW, especially from WOW scales.  相似文献
