This experiment was conducted to determine the effects of corn silage (CS) hybrids and quality of alfalfa hay (AH) in high-forage dairy diets on N utilization, ruminal fermentation, and lactational performance by early-lactating dairy cows. Eight multiparous Holstein cows were used in a duplicated 4 × 4 Latin square experiment with a 2 × 2 factorial arrangement of dietary treatments. The 8 cows (average days in milk = 23 ± 11.2) were surgically fitted with ruminal cannula, and the 2 squares were conducted simultaneously. Within square, cows were randomly assigned to a sequence of 4 diets: conventional CS (CCS) or brown midrib CS (BMR) was combined with fair-quality AH [FAH: 46.7% neutral detergent fiber (NDF) and 18.4% crude protein (CP)] or high-quality AH (HAH: 39.2% NDF and 20.7% CP) to form 4 treatments: CCS with FAH, CCS with HAH, BMR with FAH, and BMR with HAH. Diets were isonitrogenous across treatments, averaging 15.9% CP. Each period lasted a total of 21 d, with 14 d for treatment adaptation and 7 d for data collection and sampling. Intake of DM and milk yield did not differ in response to CS hybrids or AH quality. Although feeding BMR-based diets decreased urinary N output by 24%, it did not affect fecal N output. Feeding HAH decreased urinary N output by 15% but increased fecal N output by 20%. Nitrogen efficiency [milk N (g/d)/intake N (g/d)] tended to increase for BMR treatments. Ruminal ammonia-N concentration was lower for cows fed BMR-based diets than for those fed CCS-based diets but was not affected by quality of AH. Feeding BMR-based diets or HAH decreased milk urea N concentration by 23 or 15%, respectively, compared with CCS-based diets or FAH. Total volatile fatty acid concentration increased with HAH but was not influenced by CS hybrids. Feeding BMR-based diets decreased urinary N-to-fecal N ratio (UN:FN), and it was further reduced by feeding HAH. Although cows fed the BMR-based diets tended to increase milk N-to-manure N ratio, the quality of AH did not affect the ratio. The lower ratio of UN:FN with a higher ratio of milk N-to-manure N ratio for the BMR-based diets indicates that feeding BMR may reduce manure ammonia-N by reducing excretion of urinary N and increasing secretion of milk N per unit of manure N excreted.  相似文献

Daily pasture allowance (PA) is defined as the product of pregrazing pasture mass and offered area, and is the major grazing management factor determining pasture utilization per unit area and daily performance of grazing dairy cows. The objective of the present study was to perform a meta-analysis reviewing the effect of PA on pasture intake, milk production, milk composition, and grazing behavior of dairy cows. Experiments studying the effect of PA on pasture intake or milk production, which eventually included milk composition or grazing behavior data, or both, were selected to create a database. Papers were selected only if at least 2 PA were compared under the same experimental conditions, particularly the same pasture mass (i.e., where PA levels were only obtained through changes in daily offered area). The final database included 97 PA comparisons reported in 56 papers. For analytical purposes, the database was subdivided into 3 subsets that varied according to the estimation height (EH) at which PA was determined; that is, PA above ground level (PA0 subset), PA above 2.5 to 3.5 cm (PA3 subset), and PA above 4 to 5 cm (PA5 subset). Statistical analyses were conducted independently on the PA0, PA3, and PA5 subsets and on the whole database (global analysis) by using linear and nonlinear mixed-model procedures. The curves, either exponential, quadratic, or linear, describing the effects of PA on pasture intake, milk production, or grazing behavior of dairy cows are conceptually similar, whatever the EH. The equations describing these curves are, however, specific for each EH. Accordingly, from typical low to high PA, the increase in pasture intake (0.13 vs. 0.21 vs. 0.28 kg/kg of PA), milk production (0.11 vs. 0.17 vs. 0.24 kg/kg of PA), and milk solids production (0.008 vs. 0.010 vs. 0.013 kg/kg of PA) per kilogram of increase in PA was lower for PA0 than for PA3, and for PA3 than for PA5. Grazing time increased from low to medium PA and did not vary from medium to high PA. Pasture intake rate seemed to increase from low to medium PA because of greater bite mass, whereas it increased from medium to high PA because of greater biting rate. The present meta-analysis demonstrated that the general relationship between PA and any dependent variable is quite strong and independent of EH. This suggests no specific relationship for some parts of the world or methodology approach, with a high portability of the global equations calculated here. These results are useful for improving grazing management and modeling on pasture-based dairy systems.  相似文献

Effects of glucose infusion on feed intake, milk production, and metabolic responses of early postpartum cows fed diets varying in starch and protein concentration were evaluated by utilizing a randomized complete block design with a 2 × 2 factorial arrangement of treatments. Twenty-four multiparous Holstein cows were blocked by body condition score and 305-d mature-equivalent milk yield and randomly assigned at calving to 1 of 4 treatments. Treatments were continuous intrajugular infusion of glucose (GI) or isotonic saline (SI), and diets containing high starch, low crude protein (HSLP) or high crude protein, low starch (HPLS) concentrations. Treatments were initiated at the first scheduled feeding following parturition and lasted 12 d. The GI reduced cumulative dry matter intake and tended to reduce daily dry matter intake and meal size for HPLS but not HSLP compared with SI. The GI increased cumulative milk yield by 39 kg/12 d compared with SI by increasing it for HSLP but not HPLS. The HPLS treatment tended to increase loss of body condition from 0.65 to 0.82 body condition score units/12 d compared with HSLP. Consistent with this, HPLS increased plasma concentrations of nonesterified fatty acids, β-hydroxybutyrate, liver triglyceride, milk fat concentration and yield, 3.5% fat-corrected milk, and milk urea nitrogen compared with HSLP. Overall, the GI-HPLS treatment reduced feed intake by reducing meal size. The GI-HPLS may have reduced meal size by the independent or additive effects of (1) decreasing hepatic gluconeogenesis and promoting oxidation of acetyl coenzyme A (CoA), (2) elevated plasma nonesterified fatty acids from HPLS increasing the pool of acetyl CoA available to be oxidized, and (3) the HPLS diet increasing urea synthesis, which also provides the tricarboxylic acid cycle intermediate fumarate to promote oxidation of acetyl CoA.  相似文献

Objectives were to determine the effects of GnRH at the initiation of the 5-d timed artificial insemination (AI) program combined with 2 injections of PGF on ovarian responses and pregnancy per AI (P/AI) in dairy heifers, and the role of progesterone concentrations on LH release and ovulation in response to GnRH. In study 1, heifers received a controlled internal drug release (CIDR) insert containing 1.38 g of progesterone on d 0, an injection of 25 mg of PGF and CIDR removal on d 5, and an injection of 100 μg GnRH concurrently with AI on d 8. Heifers were assigned to receive no additional treatment (control; n = 559) or an injection of GnRH on d 0 and a second injection of PGF on d 6 (G2P; n = 547). In study 2, all heifers were treated as described for the control in study 1, and were allocated to receive no additional treatment (control; n = 723), an injection of PGF on d 6 (NG2P; n = 703), or an injection of GnRH on d 0 and an injection of PGF on d 6 (G2P; n = 718). In study 3, heifers received a CIDR on d 7 after ovulation and were assigned randomly to a low-progesterone (LP; n = 6) treatment in which 2 injections of 25 mg of PGF each were administered 12 h apart, on d 7 and 7.5 after ovulation, or to a high-progesterone (HP; n = 12) treatment in which no PGF was administered. On d 8, heifers received 100 μg of GnRH and blood was sampled at every 15 min from −30 to 180 min relative to the GnRH for assessment of LH concentrations. Additionally, 94 heifers were assigned to LP or HP and ovulation in response to GnRH was evaluated. In study 1, P/AI was greater for G2P than for the control on d 32 (59.4 vs. 53.5%) and 60 after AI (56.6 vs. 51.3%). In study 2, administration of GnRH on d 0 increased the proportion of heifers with a new corpus luteum on d 5 (control = 21.9 vs. NG2P = 20.1 vs. G2P = 34.4%). Administration of a second PGF increased the proportion of heifers with progesterone <0.5 ng/mL at AI (control = 83.1 vs. NG2P = 93.0 and G2P = 87.2%). Pregnancy per AI was greater for G2P than for control and NG2P on d 32 (control = 52.9 vs. NG2P = 55.0 vs. G2P = 61.7%) and 60 (control = 49.0 vs. NG2P = 51.6 vs. G2P = 59.1%). In study 3, HP attenuated LH release and reduced ovulation (19.0 vs. 48.4%) in response to GnRH compared with LP. Combining GnRH and 2 doses of PGF in the 5-d timed AI protocol improved follicle turnover, luteolysis, and P/AI in heifers. Elevated concentrations of progesterone suppressed LH release and are linked with the low ovulatory response to the initial GnRH treatment of the protocol.  相似文献

The objective of this study was to compare a GnRH-based to an estrogen/progesterone (E2/P4)-based protocol for estrous cycle synchronization and fixed timed artificial insemination (TAI), both designed for synchronization of ovulation and to reduce the period from follicular emergence until ovulation in cows with a synchronized follicular wave. A total of 1,190 lactating Holstein cows (primiparous: n = 685 and multiparous: n = 505) yielding 26.5 ± 0.30 kg of milk/d at 177 ± 5.02 d in milk were randomly assigned to receive one of the following programs: 5-d Cosynch protocol [d −8: controlled internal drug release (CIDR) + GnRH; d −3: CIDR removal + PGF; d −2: PGF; d 0: TAI + GnRH] or E2/P4 protocol (d −10: CIDR + estradiol benzoate; d −3: PGF; d −2: CIDR removal + estradiol cypionate; d 0: TAI). Rectal temperature and circulating progesterone (P4) were measured on d −3, −2, 0 (TAI), and 7. The estrous cycle was considered to be synchronized when P4 was ≥1.0 ng/mL on d 7 in cows that had luteolysis (P4 ≤0.4 ng/mL on d 0). To evaluate the effects of heat stress, cows were classified by number of heat stress events: 0, 1, and 2-or-more measurements of elevated body temperature (≥39.1°C). Pregnancy success (pregnancy per artificial insemination, P/AI) was determined at d 32 and 60 after TAI. The cows in the 5-d Cosynch protocol had increased circulating P4 at the time of PGF injection (2.66 ± 0.13 vs. 1.66 ± 0.13 ng/mL). The cows in the E2/P4 protocol were more likely to be detected in estrus (62.8 vs. 43.4%) compared with the cows in the 5-d Cosynch protocol, and expression of estrus improved P/AI in both treatments. The cows in the 5-d Cosynch protocol had greater percentage of synchronized estrous cycle (78.2%), compared with cows in the E2/P4 protocol (70.7%). On d 60, the E2/P4 protocol tended to improve P/AI (20.7 vs. 16.7%) and reduced pregnancy loss from 32 to 60 d (11.0 vs. 19.6%), compared with the 5-d Cosynch protocol. In cows withtheir estrous cycle synchronized, the E2/P4 protocol had greater P/AI (25.6 vs. 17.7%) on d 60 and lower pregnancy loss from 32 to 60 d (6.7 vs. 21.7%) compared with cows in the 5-d Cosynch protocol. Follicle diameter affected pregnancy loss from 32 to 60 d only in the cows in the 5-d Cosynch protocol, with smaller follicles resulting in greater pregnancy loss. Pregnancy per AI at d 60 was different between protocols in the cows with 2 or more measurements of heat stress (5-d Cosynch = 12.2% vs. E2/P4 = 22.8%), but not in the cows without or with 1 heat stress measurement. In conclusion, the 5-d Cosynch protocol apparently produced better estrous cycle synchronization than the E2/P4 protocol but did not improve P/AI. The potential explanation for these results is that increased E2 concentrations during the periovulatory period can improve pregnancy success and pregnancy maintenance, and this effect appears to be greatest in heat-stressed cows when circulating E2 may be reduced.  相似文献

The objectives of this study were to describe, using the goat SNP50 BeadChip (Illumina Inc., San Diego, CA), molecular data for the French dairy goat population and compare the effect of using genomic information on breeding value accuracy in different reference populations. Several multi-breed (Alpine and Saanen) reference population sizes, including or excluding female genotypes (from 67 males to 677 males, and 1,985 females), were used. Genomic evaluations were performed using genomic best linear unbiased predictor for milk production traits, somatic cell score, and some udder type traits. At a marker distance of 50 kb, the average r2 (squared correlation coefficient) value of linkage disequilibrium was 0.14, and persistence of linkage disequilibrium as correlation of r-values among Saanen and Alpine breeds was 0.56. Genomic evaluation accuracies obtained from cross validation ranged from 36 to 53%. Biases of these estimations assessed by regression coefficients (from 0.73 to 0.98) of phenotypes on genomic breeding values were higher for traits such as protein yield than for udder type traits. Using the reference population that included all males and females, accuracies of genomic breeding values derived from prediction error variances (model accuracy) obtained for young buck candidates without phenotypes ranged from 52 to 56%. This was lower than the average pedigree-derived breeding value accuracies obtained at birth for these males from the official genetic evaluation (62%). Adding females to the reference population of 677 males improved accuracy by 5 to 9% depending on the trait considered. Gains in model accuracies of genomic breeding values ranged from 1 to 7%, lower than reported in other studies. The gains in breeding value accuracy obtained using genomic information were not as good as expected because of the limited size (at most 677 males and 1,985 females) and the structure of the reference population.  相似文献

ABSTRACT

Dried feed materials, such as sugar beet pellets, may be a source of high levels of PCDD/Fs. The studies followed up dioxin congener elimination from contaminated milk (5.02 ± 1.39 pg WHO PCDD/F-PCBs-TEQ g?1 fat) up to 338 days after withdrawal of polluted feed ingredient (4.37 ± 0.25 ng PCDD/F/DL-PCBs-TEQ kg?1 feed). The main objectives of the study were to estimate the length of the time needed to achieve an acceptable PCDD/F milk level as set by European Union law, to track the fate of the PCDD/F congeners, and to assess the potential risk for consumers associated with long-term consumption of contaminated milk. After approximately 200 days, the PCDD/F levels reached ML (2.5 pg WHO-TEQ g?1 fat), then decreased to the action level after more than 300 days (1.75 pg WHO-TEQ g?1 fat) for most of cows. The potential risk of PCDD/F/DL-PCBs intake was characterised by comparing the theoretically calculated weekly and monthly intakes with the toxicological reference values (TRVs). The dioxin intake for the average adult and high-milk consumers did not exceed half of any of the TRVs applied (TWI, PTMI). The consumption of milk by children in the amount recommended by nutritionists (500 ml day–1) resulted in an exposure equal to 317% TWI and 275% PTMI.  相似文献

This study was designed to evaluate the response of hematological and biochemical parameters submitted to pegbovigrastim administration and postchallenge with lipopolysaccharide (LPS). In experiment 1, 20 newborn Holstein calves were divided into 2 groups: the Imrestor (Elanco Saúde Animal, São Paulo, Brazil) group (IMR, n = 10), which received a 25 μg/kg of body weight (BW) subcutaneous administration of pegbovigrastim, and the control group (CTR, n = 10), which received a subcutaneous administration of 0.9% saline solution. Blood samples were collected on d 0, 10, 12, and 14 relative to birth to analyze the biochemical and hematological parameters. Moreover, growth measurements were taken on d 0, 7, 14, 21, and 60 relative to birth. The number of total leukocytes in the IMR group increased on d 12 and 14 in comparison to the CTR group, as well as the counts of segmented neutrophils, band cells, and monocytes. No differences were observed in the other hematological, biochemical, and growth parameters. In experiment 2, 20 Holstein calves from 30 to 60 d old were divided into 4 groups: group 1 (LPS, n = 5) received a 0.25 μg/kg of BW single intravenous dose of Escherichia coli LPS at d 0; group 2 (IMR, n = 5) received a 25 μg/kg of BW subcutaneous dose of pegbovigrastim at d 1; group 3 (IMR + LPS, n = 5) received a 0.25 μg/kg of BW intravenous LPS dose at d 0 and a 25 μg/kg of BW subcutaneous dose of pegbovigrastim at d 1; and group 4 (CTR, n = 5) received an intravenous dose of 0.9% sodium chloride at d 0 and a subcutaneous dose of 0.9% sodium chloride at d 1. For the analysis of biochemical and hematological parameters, blood samples were collected on d ?1, 0, 1, 2, 3, 4, 8, 14, and 21 relative to LPS administration. An increase in the number of total leukocytes was observed in the IMR, IMR + LPS, and LPS groups, and the IMR group remained as the highest from d 2 to 21. The levels of paraoxonase 1 were higher in the IMR group compared with all the others. The administration of pegbovigrastim in the dairy calves increased the number of circulating leukocytes, especially neutrophils, with an increase in paraoxonase 1, without altering the metabolites for the hepatic function.  相似文献

Subclinical ketosis is a common metabolic disorder affecting dairy cattle that results in a greater risk for the development of subsequent metabolic and infectious disease. Canwest Dairy Herd Improvement (DHI; Guelph, ON, Canada) has begun to use an infrared test (MilkoScan FT600, Foss Analytical A/S, Hillerød, Denmark) applied to metered composite milk samples to detect β-hydroxybutyrate (BHB) levels as a herd surveillance test for hyperketonemia. However, the test has not been compared with the gold standard, serum BHB as determined in a reference diagnostic laboratory. The objective of this cross-sectional diagnostic accuracy study was to validate the DHI milk BHB test to identify cows with hyperketonemia as determined by quantification of BHB in serum. A total of 316 cows from 17 dairy herds in southwestern Ontario had a milk and blood sample taken. Milk was collected at a routine DHI test, and blood from the same cow was sampled within 24 h of the milk test. The BHB concentration in milk was determined using the DHI milk BHB test, and serum was sent to the Animal Health Laboratory at the University of Guelph (Guelph, ON, Canada). A nonparametric receiver operating characteristic curve was generated to compare DHI milk BHB concentrations with serum BHB concentrations. Overall, a total of 34 cows (11%) had a level of serum BHB ≥1.2 mmol/L. The concentration of DHI milk BHB was moderately correlated with the concentration of serum BHB, yielding a coefficient of determination value of 0.61. The optimal cut point for determining hyperketonemia (≥1.2 mmol/L) on the DHI milk BHB test was ≥0.14 mmol/L, yielding a sensitivity of 81% and specificity of 92%. The performance of the DHI milk BHB test varied depending on the days in milk (DIM) of the cows tested, with a higher specificity being found in cows that were ≤25 DIM compared with cows tested >25 DIM. If the herd-level prevalence of hyperketonemia was ≥14%, the DHI milk BHB test had an improved sensitivity when compared with a herd-level prevalence of <14%. This study demonstrates that the DHI milk BHB test is a reliable measure for evaluating hyperketonemia using routine DHI milk samples and could be used as a herd-level monitoring tool for ketosis when evaluating nutritional management or preventative medicine strategies.  相似文献

After parturition, the start of copious milk production in dairy cows requires the closure of tight junctions (TJ) to form the blood–milk barrier and prevent paracellular transfer of blood constituents into milk [e.g., lactate dehydrogenase (LDH) and serum albumin (SA)] and vice versa [e.g., appearance of α-lactalbumin (α-LA) in blood]. Serotonin (5-HT) has been demonstrated to alter tight junction permeability in the mammary gland. The present study investigated individual differences of TJ permeability of mammary epithelium at the beginning of lactation in relation to circulating 5-HT in dairy cows. Blood and milk samples were obtained from 11 multiparous Holstein dairy cows for the first time at 4 h after parturition, at the following 5 milkings, and at the evening milkings on d 5, 8, 10, and 14 of lactation. Retrospectively, cows were split into 2 groups according to their calculated areas under the curve of serum 5-HT during the entire experimental period: a high-serum 5-HT (HSS) group (5 cows) and a low-serum 5-HT (LSS) group (6 cows). The areas under the curve of serum 5-HT concentrations over the 324-h experimental period were 62 ± 2 × 103 ng/mL in HSS and 25 ± 5 × 103 ng/mL in LSS. Plasma α-LA concentration was greater in LSS than in HSS cows at the first milking, but no difference between groups was found from the second to sixth milking. Yield of α-LA in milk was lower in HSS than in LSS during the first 6 milkings postpartum, especially in colostrum. Concentrations of α-LA, IgG1, and IgG2 in milk did not differ between groups during the entire experiment except for higher IgG observed in LSS than in HSS at the second milking and for higher IgG2 found in HSS compared with LSS on d 5. In contrast, SA concentrations and LDH activity in milk were lower in LSS compared with HSS cows during the first 6 milkings postpartum, particularly in colostrum. Milk somatic cell count was higher in HSS than in LSS throughout the study. Higher circulating 5-HT concentrations were associated with an increased transfer of the paracellularly transported SA, LDH, and somatic cell count, especially at the first milking, suggesting that 5-HT affects TJ permeability during closure of the blood–milk barrier at the onset of lactation. Furthermore, higher serum 5-HT concentrations were associated with a lower α-LA yield in milk. A consistent relationship with serum 5-HT concentrations was neither observed for the transfer of IgG2 nor the primarily transcellular transport of IgG1 during the first milkings after parturition.  相似文献
