Connexins are a 21-member membrane protein family constituting channels evolved in direct communication between adjacent cells by passaging cytoplasmic molecules and ions. Hexametrical assembly of connexin proteins in plasma membrane forms a wide aqueous pore known as connexin hemichannel. These hemichannels mediate cytoplasm and extracellular milieu communication both in many external tissues and in the central nervous system. In this study, a series of molecular dynamics simulations has been performed to investigate the effect of applied static and alternating electric fields on the stability and conformation of human connexin26 hemichannel. The root mean square deviations of C-alpha atoms, the dipole moment distribution, the number of inter-protein hydrogen bonds and the number of water-protein hydrogen bonds were used to assess connexin26 hemichannel stability. In the static field case, our results show that although the lowest field used in this study (0.1 V/nm) does not lead to the hemichannel deconstruction, stronger fields (>0.1 V/nm), however, disrupt the protein structure during the simulations time period. Furthermore, in the alternating case, compared to static field case, field effects on the connexin26 hemichannel conformation are reduced and consequently the protein maintains its native structure for longer times. Specifically, for the highest frequency used in this study (50 GHz), the hemichannel keeps its structure even under the effect of the strongest field (0.4 V/nm). According to our results, the protein secondary structure is preserved in the characteristic times determined for the protein deconstruction. Consequently, we suggest that the protein deconstruction is due to the tertiary and quaternary structure loss.  相似文献

The authors introduce a manual-based treatment, labeled dynamic deconstructive psychotherapy, developed for those patients with borderline personality disorder who are most difficult to engage in therapy, such as those having co-occurring substance use disorders. This treatment model is based on the hypothesis that borderline pathology and related behaviors reflect impairment in specific neurocognitive functions, including association, attribution, and alterity that form the basis for a coherent and differentiated self. Dynamic deconstructive psychotherapy aims to activate and remediate neurocognitive self-capacities by facilitating elaboration of affect-laden interpersonal experiences and integration of attributions, as well as providing novel experiences in the patient-therapist relationship that promote self-other differentiation. Abstract

The markings associated with the physical wear and tear of denim garments are an integral aspect of material culture, pursued as creative concepts through deconstructed fashion approaches. For some designers a textile's heritage and the effects of time are as important as reworking silhouettes from history. While fashion is inherently forward thinking, it is the patina of age that comes from “wearing” a garment that can be integral to the designer's artistic vision. Denim garments possess unique possibilities for articulating the human form in ways that transcend the vagaries of the latest cut, through evidencing (or simulating) practices of wearing and concepts of longevity. This paper explores denim's capacity to act as a canvas for the body through analysis of a series of conceptual and commercial approaches drawn from contemporary fashion and textile designers, who have reinforced denim's origins or reinterpreted it through creative experimentation. The study is underpinned by the author's empirical research, which seeks to reveal both the recognizable and more abstract visual qualities embodied within denim as a fashion construct. Denim is evaluated as a surface for conveying complex visual signifiers relating to temporality, demonstrated through the cloth's unique facility to embody narrative and illusion.  相似文献

The paper discusses Sharrock's and Button's criticism of our attempt to use Habermas' communicative action theory to analyze group work platforms. We demonstrate that they misconstrue our goals of the paper, misinterpret our analysis of Habermas' action types, and misunderstand the concept of critical science.
