Biodegradation is cost-effective, environmentally friendly treatment for oily contaminated sites by the use of microorganisms. In this study, laboratory experiments were conducted to establish the performance of bacterial isolates in degradation of organic compounds contained in oily sludge from the Jordanian Oil Refinery plant. As a result of the laboratory screening, three natural bacterial consortia capable of degrading total organic carbons (TOC) were prepared from isolates enriched from the oil sludge. Experiments were conducted in Erlenmeyer flasks under aerobic conditions, with TOC removal percentage varied from 0.3 to 28% depending on consortia type and concentration. Consortia 7B and 13B exhibited the highest TOC removal percentage of 28 and 22%, respectively, before nutrient addition. TOC removal rate was enhanced after addition of nutrients to incubated flasks. The highest TOC reduction (43%) was estimated after addition of combination of nitrogen, phosphorus and sulphur to consortia 7B.
A significant variation (P < 0.005) was obser0ved between the effect of consortia type and concentration on TOC% reduction. No significant variation was observed between incubation at 10 and 18 days in TOC% reduction. This is the first report concerning biological treatment of TOC by bacteria isolated from the oil refinery plants, where it lays the ground for full integrated studies recommended for the degradation of organic compounds that assist in solving sludge problems. 相似文献

Total RNA concentration as an index of microbial activity and oxygen supply in an oxidation ditch 总被引:2,自引:0,他引:2
Total RNA and chromosomal DNA concentrations at a municipal wastewater treatment plant with an oxidation ditch (OD) were monitored for 1.5 years using commercial extraction kits for DNA and RNA. No parameters correlated with the chromosomal DNA concentration. The total RNA concentration exhibited better correlation than the solids retention time and the mixed liquor suspended solids with the removal rate of total organic carbon, and can be regarded as an index of microbial activity. The total RNA concentration varied with a cycle of one year and increased at lower water temperatures in this OD. When diffusion theory was taken into account, it was found that the oxygen dissolution rate increased at lower temperature, and a small change in the oxygen dissolution rate caused a large variation in microbial activity and also affected nitrification and denitrification. The information was insufficient to clarify the various reaction relationships, but total RNA concentration will likely be useful as an index of microbial activity in actual wastewater treatment reactors. 相似文献

Thermophilic activated sludge treatment is often hampered by a turbid effluent. Reasons for this phenomenon are so far unknown. Here, the hypothesis of the temperature dependency of the hydrophobic interaction as a possible cause for diminished thermophilic activated sludge bioflocculation was tested. Adsorption of wastewater colloidal particles was monitored on different flat surfaces as a function of temperature. Adsorption on a hydrophobic surface varied with temperature between 20 and 60 degrees C and no upward or downward trend could be observed. This makes the hydrophobic interaction hypothesis unlikely in explaining the differences in mesophilic and thermophilic activated sludge bioflocculation. Both mesophilic and thermophilic biomass did not flocculate with wastewater colloidal particles under anaerobic conditions. Only in the presence of oxygen, with biologically active bacteria, the differences in bioflocculation behavior became evident. Bioflocculation was shown only to occur with the combination of wastewater and viable mesophilic biomass at 30 degrees C, in the presence of oxygen. Bioflocculation did not occur in case the biomass was inactivated or when oxygen was absent. Thermophilic activated sludge hardly showed any bioflocculation, also under mesophilic conditions. Despite the differences in bioflocculation behavior, sludge hydrophobicity and sludge zetapotentials were almost similar. Theoretical calculations using the DLVO (Derjaguin, Landau, Verweij and Overbeek) theory showed that flocculation is unlikely in all cases due to long-range electrostatic forces. These calculations, combined with the fact that bioflocculation actually did occur at 30 degrees C and the unlikelyness of the hydrophobic interaction, point in the direction of bacterial exo-polymers governing bridging flocculation. Quantification of bacteria using Fluorescence In Situ Hybridization (FISH), confocal laser scanning microscopy (CLSM) and image analysis is very time consuming and requires the availability of an expensive microscope. Therefore, a rapid method to quantify nitrifying bacteria in activated sludge using FISH and epifluorescence microscopy was developed. The quantification of the biovolume is based on manual counting of the aggregates formed by nitrifying bacteria and determination of their size. The overall uncertainty of the method was evaluated as a function of the number of analyzed microscopic fields. It was found that 10-15 microscopic fields for ammonia-oxidizing bacteria and 6-8 microscopic fields for nitrite-oxidizing bacteria per sample were optimal regarding effort and accuracy. Accordingly, the time needed for one sample was only 5-15 min, compared to about 1h for the quantification with CLSM and image analysis. As a consequence, this method also allows for the measurement of extended time series with a reasonable effort. The comparison of the determined biovolume and the measured activity showed an explicit correlation. 相似文献

Polyacrylamides are widely used as flocculants to enhance clarification of drinking waters and domestic wastewaters, for stabilization of agricultural soils, and to aid in managing mine tailings. The flocs produced with polyacrylamide may be deposited into retention areas that become anaerobic. Although it is unlikely that the carbon backbone of these polymers would be cleaved by microbial activity, the amide group could serve as a nitrogen source for microorganisms. Previous studies have shown that aerobic bacteria utilize the nitrogen from polyacrylamide. This study assessed whether methanogenesis was stimulated when an anionic polyacrylamide (Magnafloc LT27AG) was the sole fixed nitrogen source in serum-bottle microcosms. Microorganisms from two oil sands tailings sources, and a domestic anaerobic sewage sludge were used, with benzoate or acetate provided as carbon and energy sources. In each inoculum-substrate combination, the presence of polyacrylamide-enhanced methane production, indicating that polyacrylamide may stimulate microbial activities in anaerobic environments that are rich in fermentable carbon, but lack nitrogen sources. 相似文献

Alum and ferric chloride sludges from two water treatment plants (WTPs) were analyzed regarding their physicochemical characteristics and toxicity to Daphnia similis. Experiments were carried out in the dry and rainy seasons. Acute and chronic toxicity was measured using survival and reproduction as measurement endpoints. No acute toxicity of the sludge was observed in 48 h exposure. Ferric chloride sludge caused chronic toxicity, demonstrated by low fecundity and some mortality, while alum sludge caused chronic toxicity characterized by low fecundity. Some sludge characteristics varied between samplings, including turbidity, solids contents, N, P and metal (Al and Fe) concentrations. These variables and the increase of chemical oxygen demand (COD) were identified as the main cause of degradation of the receiving waters. However, no relationship was observed between these variables and degree of toxicity. It is apparent from these results that water treatment sludges may be toxic and therefore may impair receiving waters. Alum sludge was less toxic than ferric chloride sludge. 相似文献

The influence of membrane separation and mass transfer effects on the kinetics of nitrifiers was evaluated by running a membrane bioreactor (MBR) and a conventional activated sludge (CAS) plant in parallel. Both pilot plants were operated at the same sludge age and treated the same domestic wastewater. The half-saturation constants for the substrate were low in both MBR and CAS and did not differ significantly between the two processes (K(NH(4))) and 0.14+/-0.10 g(N)m(-3) and (K(NO(2))) and 0.28+/-0.20 g(N)m(-3) for the MBR and CAS, respectively). However, the half-saturation constants for oxygen exhibited a major difference between the two processes for both the ammonia-oxidizing (AOB) and nitrite-oxidizing (NOB) bacteria. The experiments yielded K(O,AOB)=0.18+/-0.04 and 0.79+/-0.08 g(O2) as well as K(O,NOB)=0.13+/-0.06 and 0.47+/-0.04 g(O2) m(-3) (substrate only NO(2)) for the MBR and CAS, respectively. The higher K(0) values of the CAS were attributed to mass transfer effects within the large flocs prevailing in the conventional system. In contrast, the sludge from the MBR consisted of very small flocs for which the diffusion resistance can be neglected. On the basis of these results, the implementation of mass transfer effects in activated sludge models is discussed and consequences for the operation of MBRs are highlighted. 相似文献

通过控制曝气量的方式研究了溶解氧对污泥减量系统除磷脱氮过程的影响。发现在低剂量2,4,5三氯苯酚(TCP)作用下,活性污泥的内源SOUR值增加,SBR系统的低DO状态持续时间增长,周期平均DO降低,形成了有利于同时硝化反硝化SND脱氮的低DO环境。综合考虑TCP浓度对污泥减量、除磷脱氮和污泥性能的影响,TCP浓度建议为2 mg/L,SBR周期平均DO值控制为2 mg/L。与对照系统相比,2 mg/LTCP污泥减量系统的曝气量增加了23%,剩余污泥排放量减少34.6%,出水水质与对照系统相当,实现了达标排放。表明低DO控制状态下、辅以排富磷污水除磷方式,TCP系统可以同时获得优异的除磷脱氮和污泥减量效果。 相似文献
