酶法拆分N-乙酰-D,L-蛋氨酸转化条件优化 总被引:1,自引:0,他引:1
重组大肠杆菌BL21/pET22b-argE表达的N-乙酰鸟氨酸脱酰基酶(NAOase)可用于脂肪族氨基酸的手性拆分.水解N-乙酰-D,L-氨基酸中的L-型底物,N-乙酰-D,L-蛋氨酸为其最合适的底物.为了确定NAOase拆分N-乙酰-D,L-蛋氨酸合适的转化条件,考察了反应温度、pH值、Co2+浓度、转化时间、底物浓度和加酶量对产物的影响.结果表明,合适的反应条件为37℃,pH值7.0,Co2+ 1 mmol/L,转化时间20 min,底物浓度150 mmol/L,菌泥5 g/L.在上述反应条件下,N-乙酰-D,L-蛋氨酸的转化率可达81.4%. 相似文献

Neutrase 0.8L and N120P proteases were used for oligopeptide production from apricot almonds meal, and response surface design was carried out to optimize the effect of hydrolysis conditions on hydrolysis degree (DH) and oligopeptide yield rate. Four independent variables were used to optimize the hydrolysis process: hydrolysis temperature (X(1)), enzyme-to substrate ratio (E/S) (X(2)), substrate concentration (X(3)) and reaction time (X(4)). Statistical analysis indicated that the four variables, quadratic terms of X(1), X(3), and X(4), and the interaction terms with X(1) had a significant (p < 0.05) effect on DH. The yield rate was also significantly affected by the four variables and quadratic terms of X(1), X(2) and X(4). Two mathematical models with high determination coefficient were obtained and could be employed to optimize protein hydrolysis. The optimal hydrolysis conditions were determined as follows: hydrolysis temperature 52.5 °C; enzyme-to-substrate ratio (E/S) 7200 U/g; substrate concentration 2%; reaction time 173 min. The initial pH 6.5 and Neutrase-to-N120P dosage ratio 2:1 were fixed in this study according to the preliminary research. ABSTRACT: 1-Octen-3-ol and 10-oxo- trans -8-decenoic acid are metabolites of the breakdown of linoleic acid (LA) by mushroom enzymes. These compounds can be produced in a bioreactor using a crude mushroom homogenate and the exogenous addition of LA and oxygen. The factors' duration of blending, mushroom–buffer ratio, effect of a surfactant, whole against partially clarified reaction broths, purity of LA, and utilization of stumps instead of whole mushrooms were studied for their effect on reaction yield using a 1-L bioreactor. The results showed the feasibility of using the more inexpensive 60%-pure LA instead of the 99%-pure LA even when a yield loss was involved. Fat and water contents of meat tissues strongly influence their thermo-physical properties. Seven groups of ground chicken breast meat containing varying amounts of fat and water were prepared for this investigation. Each group was heated to end-point temperatures (EPTs) of 69.0 to 71.0°C at 0.5°C intervals. Severity of heat treatments was evaluated by the total process lethality calculated using the General Method. The fat and water contents affected the heating time to reach specific EPTs and the residual catalatic activity at EPTs of 69.0, 69.5 and 70.0°C, but did not affect total process lethality. The oxidative stability of structured lipids (SLs) synthesised by specific sn‐1,3‐lipase catalysed interesterification of soybean oil (SBO) with caprylic acid (CA) in a stirred batch reactor was studied. SLs contained considerable amounts of tocopherol (TOH) isomers, although they lost almost 25% of endogenous TOHs during production. The effects of the addition of different TOH homologues (α, β, γ, δ), ascorbyl palmitate (AP, 200 ppm), lecithin (Le, 1000 ppm), butylated hydroxytoluene (BHT, 100 ppm) and butylated hydroxyanisole (BHA, 100 ppm) on the oxidative stability of SLs were investigated. Induction time (IT) of SBO, determined by the Rancimat method, decreased from 8.4 to 5.8 h at 110 °C after the modification. On the other hand, purified SLs and purified SBO had the same IT due to the tocopherol reduction during silica purification. No significant difference was observed between IT of SLs and SLs plus different α‐tocopherol concentrations (50, 100, 150, 200, 300, 500 and 1000 ppm) (P > 0.05). However, the addition of Le and/or AP significantly improved oxidative stability of purified SLs and SBO. The ternary blend containing δ‐TOH, AP and Le had higher IT than ternary blends of α‐TOH, β‐TOH or γ‐TOH. Furthermore, ternary blend containing BHA, AP and Le had higher IT than ternary blends of BHT, AP and Le. In addition, there was an increase in peroxide value (PV), conjugated diene (CD) content and p‐anisidine value (AV) during oxidation of oils at 60 °C. 为了综合利用芝麻粕资源,以芝麻粕为原料,利用硬脂酸、吐温80、十二烷基硫酸钠三种表面活性剂对其进行脱脂处理,再采用水剂法去除表面活性剂,最后利用中性蛋白酶、碱性蛋白酶和木瓜蛋白酶对芝麻粕蛋白进行水解,以芝麻粕蛋白溶出率为考察指标优选出最佳水解酶。在此基础上,通过单因素试验和正交试验对酶解工艺条件进行优化,并利用2,4-二硝基氟苯柱前衍生高效液相色谱法分析水解物成分。结果表明,吐温80脱脂效果最好,芝麻粕脱脂率达93.49%;碱性蛋白酶为最佳水解酶,其最佳酶解条件为料水比1∶10(g∶mL)、酶解温度50 ℃、加酶量0.04%、酶解时间3.0 h。在此最佳酶解条件下,芝麻粕蛋白溶出率为88.94%。水解物中含有20种氨基酸,其中8种人体必需氨基酸含量高于FAO/WHO的推荐值。 相似文献
