The present study evaluated biogenic amine (BA) content during the fermentation period in functional sheep milk yogurts. Four treatments were prepared and assessed: natural (NSY), prebiotic (PreSY), probiotic (ProSY), and synbiotic (SynSY). Biogenic amines (putrescine, cadaverine, spermidine, spermine, and tyramine), proteolysis activity, and pH were measured during each hour of fermentation. Grumixama pulp was added to all formulations as a technological strategy and potential substrate for bacteria during fermentation. The yogurt and probiotic bacteria were viable (≥7 log cfu·mL−1) on d 0. The pH levels of the functional sheep milk yogurts had a more pronounced decrease than did the control of NSY. However, all yogurt samples underwent gradual decreases in pH until final fermentation. Proteolytic activity remained constant in all treatments during fermentation. The NSY, PreSY, ProSY, and SynSY presented the same behavior for all BA, with differences in concentration. Putrescine, cadaverine, and spermidine contents decreased, whereas spermine remained constant and tyramine increased. We conclude that fermentation of functional sheep milk yogurts can produce tyramine.  相似文献

A series of food safety incidents related to domestically produced dairy in China harmed consumer confidence and caused a dramatic decrease in dairy consumption from 2008 to 2010. Compared with previous studies that mainly assessed consumer confidence in food or dairy in general, this study focused on 2 specific dairy products, milk and infant milk formula (IMF). Based on data from 934 Chinese consumers, confidence in the safety of milk and IMF was measured by using a multi-item scale with 2 dimensions—optimism and pessimism. We found that Chinese consumers were optimistic about the safety of milk and IMF but, at the same time, were concerned about the risks related to milk and IMF. Chinese consumers have less confidence in IMF than in milk. Structural equation modeling indicated that consumer trust in manufacturers and third parties in the dairy supply chain and confidence in other foods, such as meat and fish, can foster consumer confidence in the safety of milk and IMF, while the extent of recall of the melamine infant formula incident in 2008 and trait worry exert negative effects. In addition, consumers with more children or less family income are likely to be more confident in domestically produced IMF. These insights into consumer confidence and its determinants can assist dairy supply chains and regulatory institutions in formulating effective risk communication strategies to strengthen consumer confidence.  相似文献

Mother's milk provides newborns with various nutrients (e.g., enzymes, proteins, peptides, hormones, antibodies) that help babies grow and protect them from bacterial and viral infections. The functions of many components of breast milk can be very different from their corresponding functions in body fluids of healthy adults. Catalytic antibodies (abzymes) that hydrolyze peptides, proteins, DNA, RNA, and oligosaccharides were detected not only in human milk, but also in the blood sera of autoimmune patients. However, abzymes with unexpected synthetic activities (lipids, oligosaccharides, and protein kinase activities) were revealed in milk that were not found in the blood of autoimmune patients. The nutrition of infants with fresh milk has a very specific role; newborns are well protected by antibodies of mother's milk (passive immunity). Protease abzymes were found in the blood sera of autoimmune patients, whereas healthy humans usually do not contain such autoantibodies. Here, we present the first evidence that the milk of healthy mothers contains secretory (s)IgA that effectively hydrolyze 5 histones (e.g., H1, H2A, H2B, H3, and H4) and myelin basic protein (MBP). Several rigid criteria were applied to show that protease activity is an intrinsic property of sIgA. Milk abzymes against 5 histones cannot hydrolyze different control proteins except histones and MBP, whereas autoantibodies against MBP split this protein and 5 histones. Antibodies against histones and MBP exhibit complexation polyreactivity as well as specific and unusual catalytic cross-reactivity. With some exceptions, the specific sites of hydrolysis of H1, H2A, and H2B by sIgA against histones do not coincide with the sites of hydrolysis by abzymes against MBP. On the whole, fresh human milk is a very specific source of many of the most unusual antibodies and abzymes.  相似文献

The probiotic strains Lactobacillus brevis CCMA1284 and Lactobacillus plantarum CCMA0359 were microencapsulated by spray drying using different matrices – whey powder (W), whey powder with inulin (WI) and whey powder with maltodextrin (WM). Viability of the microencapsulated strains in acid and bile juices and during 90 days of storage (seven and 25 °C) was evaluated. The two strains exhibited high encapsulation efficiency (> 86%) by spray drying. The different matrices maintained L. plantarum viability above six log CFU g−1 at 7 °C for 90 days, whereas similar results for L. brevis were observed only for W. The use of inulin as matrix of encapsulation did not enhance bacterial viability in the evaluated conditions. In general, the use of W and WM as matrices was effective for L. plantarum viability. However, only W was effective for L. brevis in the evaluated conditions. The spray drying technique was successfully adopted for the encapsulation of L. plantarum CCMA0359 and L. brevis CCMA1284 strains.  相似文献

The aim of this study was to improve textural and antioxidant capacity of dromedary yogurt using ultrafiltration process and date powder. Ultrafiltration increased total solids content of dromedary milk within the range considered optimal to develop yogurt. Texture profile of Greek yogurt fortified with date powder (GYD) improved considerably compared to control. Sample of GYD are more appealing to consumer than control. LC-ESI-MS analysis of GYD extracts allowed the identification of fifteen phenolic compounds, among which quinic acid and cirsiliol were found to be the major phenolic acid and flavonol, respectively. GYD exhibited the highest DPPH•-radical scavenging activity, Fe2+ chelating capacity and Fe3+ reducing power. The follow-up of physical and microbiological stability of GYD and control during cold storage showed that date powder significantly increased bacterial counts and decreased syneresis. Therefore, ultrafiltration and date powder could be valued as effective tool to solve the poor consistency of dromedary milk products.  相似文献

采用液相色谱-串联质谱技术分析我国部分地区市售牛肉、牛奶中可的松和氢化可的松的含量水平。均质的牛肉或牛奶样品在乙酸-乙酸钠缓冲液中酶解后用甲醇提取,提取液用石墨化碳黑固相萃取小柱和氨基固相萃取小柱净化,洗脱液氮吹至干并用甲醇溶液定容,液相色谱-串联质谱测定。采用SPSS?21.0软件进行数据分析。对采自我国9?省市的346?份牛肉和牛奶样品进行检测,分析牛肉和牛奶中可的松和氢化可的松的存在水平。牛肉和牛奶中均检出可的松和氢化可的松,检出率在86.1%~100.0%之间,检出含量范围为0.02~74.88?μg/kg。牛肉中可的松和氢化可的松的含量平均值(中位数)分别为1.69(1.14)?μg/kg和12.16(8.14)?μg/kg,牛奶中可的松和氢化可的松的含量平均值(中位数)分别为0.23(0.23)?μg/kg和0.72(0.72)?μg/kg。牛肉和牛奶中氢化可的松与可的松的含量均呈显著正相关。  相似文献

Fourier-transform infrared (FTIR) spectroscopy is a powerful high-throughput phenotyping tool for predicting traits that are expensive and difficult to measure in dairy cattle. Calibration equations are often developed using standard methods, such as partial least squares (PLS) regression. Methods that employ penalization, rank-reduction, and variable selection, as well as being able to model the nonlinear relations between phenotype and FTIR, might offer improvements in predictive ability and model robustness. This study aimed to compare the predictive ability of 2 machine learning methods, namely random forest (RF) and gradient boosting machine (GBM), and penalized regression against PLS regression for predicting 3 phenotypes differing in terms of biological meaning and relationships with milk composition (i.e., phenotypes measurable directly and not directly in milk, reflecting different biological processes which can be captured using milk spectra) in Holstein-Friesian cattle under 2 cross-validation scenarios. The data set comprised phenotypic information from 471 Holstein-Friesian cows, and 3 target phenotypes were evaluated: (1) body condition score (BCS), (2) blood β-hydroxybutyrate (BHB, mmol/L), and (3) κ-casein expressed as a percentage of nitrogen (κ-CN, % N). The data set was split considering 2 cross-validation scenarios: samples-out random in which the population was randomly split into 10-folds (8-folds for training and 1-fold for validation and testing); and herd/date-out in which the population was randomly assigned to training (70% herd), validation (10%), and testing (20% herd) based on the herd and date in which the samples were collected. The random grid search was performed using the training subset for the hyperparameter optimization and the validation set was used for the generalization of prediction error. The trained model was then used to assess the final prediction in the testing subset. The grid search for penalized regression evidenced that the elastic net (EN) was the best regularization with increase in predictive ability of 5%. The performance of PLS (standard model) was compared against 2 machine learning techniques and penalized regression using 2 cross-validation scenarios. Machine learning methods showed a greater predictive ability for BCS (0.63 for GBM and 0.61 for RF), BHB (0.80 for GBM and 0.79 for RF), and κ-CN (0.81 for GBM and 0.80 for RF) in samples-out cross-validation. Considering a herd/date-out cross-validation these values were 0.58 (GBM and RF) for BCS, 0.73 (GBM and RF) for BHB, and 0.77 (GBM and RF) for κ-CN. The GBM model tended to outperform other methods in predictive ability around 4%, 1%, and 7% for EN, RF, and PLS, respectively. The prediction accuracies of the GBM and RF models were similar, and differed statistically from the PLS model in samples-out random cross-validation. Although, machine learning techniques outperformed PLS in herd/date-out cross-validation, no significant differences were observed in terms of predictive ability due to the large standard deviation observed for predictions. Overall, GBM achieved the highest accuracy of FTIR-based prediction of the different phenotypic traits across the cross-validation scenarios. These results indicate that GBM is a promising method for obtaining more accurate FTIR-based predictions for different phenotypes in dairy cattle.  相似文献

Keeping replacement heifers that were the progeny of primiparous cows mated by artificial insemination enhances rates of genetic gain. Previous research has shown that heifers that were the progeny of primiparous cows were lighter at birth and grew at a slower rate to first calving compared with heifers born to multiparous dams. Furthermore, heifers that were heavier before first calving produced more milk than did lighter heifers. This study aimed to determine whether there were body weight, milk production, or reproductive disadvantages for heifers born from primiparous compared with multiparous dams. Data comprised body weight records from 189,936 New Zealand dairy heifers. Dams were allocated to 4 groups according to their age: 2 yr old (n = 13,717), 3 yr old (n = 39,258), 4 to 8 yr old (n = 120,859), and 9 yr or older (n = 16,102). Heifers that were the progeny of 2-yr-old dams were lighter from 3 to 21 mo of age than heifers that were the progeny of 3-yr-old and 4- to 8-yr-old dams. The progeny of 2- and 3-yr-old dams produced similar milk solids yields (± standard error of the mean) during their first lactation (304.9 ± 1.6 and 304.1 ± 1.5 kg, respectively), but more than that of 4- to 8-yr-old dams (302.4 ± 1.5). Furthermore, the progeny of 2-yr-old dams had similar stayabilities to first, second, and third calving to that of the progeny of 4- to 8-yr-old and ≥9-yr-old dams. Reproductive performance, as measured by calving and recalving rates was similar in first-calving heifers of all age-of-dam classes. Additionally, second and third calving rates were similar for the progeny of 2- and 3-yr-old dams. Interestingly, the progeny of dams ≥9 yr old had the lowest milk solids production in first (297.8 ± 1.6 kg), second (341.6 ± 1.8 kg), and third lactations (393.2 ± 2.4 kg). Based on the results of this study, keeping replacements from dams aged 9 yr and over could not be recommended. Furthermore, heifers born to 2-yr-old dams were lighter but produced more milk than heifers from older dams, in addition to having superior genetic merit.  相似文献

The objective of this study was to investigate the protective effect of five different emulsifiers on iron‐mediated lipid oxidation in 70% fish oil‐in‐water emulsions. The emulsifiers were either based on protein (whey protein isolate and sodium caseinate) or based on phospholipid (soy lecithin and two milk phospholipids with different phospholipid contents, MPL20 and MPL75). Lipid oxidation was studied at pH 4.5 and 7.0, and results were compared to lipid oxidation in neat fish oil. Results showed that all emulsions oxidised more than neat oil. Furthermore, emulsions prepared with proteins oxidised more at low pH than at high pH, and casein emulsions oxidised the least (Peroxide value (PV) at day 7 was 0.5–0.7 meq kg?1). Among emulsions prepared with phospholipids, emulsions with MPL75 were the most oxidised followed by emulsions prepared with lecithin and MPL20. Thus, PV in MPL75 emulsions was 5.0–5.5 meq kg?1 at day 7 compared with 0.9–1.9 meq kg?1 in MPL20 emulsions.  相似文献
