采用气相色谱-串联质谱技术测定油菜蜜中6 种低聚糖成分,高效液相色谱-串联质谱技术测定油菜蜜中18 种酚酸物质,并结合偏最小二乘判别分析(partial least square-discrimination analysis,PLS-DA)对来自湖北钟祥市、江苏盐城市、青海刚察县3 个具有显著地理、气候、环境差异的51 个油菜蜜样本进行产地鉴别。方差分析结果显示:3 个产地油菜蜜中松二糖含量具有显著性差异,且青海刚察油菜蜜低聚糖含量相对偏高;油菜蜜的18 种多酚类物质中大多数具有显著的地理差异性且湖北钟祥的油菜蜜中多酚含量相对偏高。多元统计分析结果显示多酚具有显著的地理特征性。油菜蜜中低聚糖和多酚的含量结合PLS-DA产地鉴别的预测精度可达到97%。  相似文献

Phosphoryl oligosaccharides of calcium (POs-Ca) is a calcium salt of phosphoryl maltooligosaccharides made from potato starch. POs-Ca is highly water-soluble and can supply both the calcium ion and acidic oligosaccharides in an aqueous solution. In this study, we investigated the effects of POs-Ca on the mycelial growth and fruiting body yield of Pleurotus ostreatus , which is one of the most widely cultivated edible mushrooms in the world. We cultivated the mushroom using both potato dextrose agar (PDA) medium and sawdust-based medium, with added calcium salts. The addition of POs-Ca into the PDA medium with a calcium concentration of 10 mg increased mycelial growth significantly ( p < 0.05, vs . control). POs-Ca addition to the sawdust-based medium at concentrations of 1.0 to 3.0 g/100 g medium increased the amount of calcium in the fruiting bodies but did not affect the length of the cultivation period or the weight of the fruiting body. The calcium content in the fruiting body increased 12-fold when compared to the control. On the other hand, neither the CaHPO 4 ・2H 2 O group nor the CaHPO 4 ・2H 2 O with oligosaccharides group showed changes in the calcium content of the fruiting bodies. Our results indicate that the use of POs-Ca in mushroom cultivation allows for the possibility of developing new functional foods like calcium-enriched edible mushrooms. This is the first report describing the effects of POs-Ca on mushroom cultivation.  相似文献

采用木聚糖酶水解麦麸制备阿魏酸低聚糖,以阿魏酸低聚糖的量为指标,研究了酶解温度、pH值、酶浓度、底物浓度、作用时间五个因素对酶解效果的影响.并在此基础上,通过中心组合试验确定最佳酶解条件.最终的优化条件是木聚糖酶浓度3%,底物浓度80 g/L,作用时间12 h,此条件下阿魏酸低聚糖提取量为0.831×10-5 moL/g麦麸.  相似文献

1 Scope

Mannan oligosaccharides (MOS) have proven effective at improving growth performance, while also reducing hyperlipidemia and inflammation. As atherosclerosis is accelerated both by hyperlipidemia and inflammation, we aim to determine the effect of dietary MOS on atherosclerosis development in hyperlipidemic ApoE*3‐Leiden.CETP (E3L.CETP) mice, a well‐established model for human‐like lipoprotein metabolism.

2 Methods and results

Female E3L.CETP mice were fed a high‐cholesterol diet, with or without 1% MOS for 14 weeks. MOS substantially decreased atherosclerotic lesions up to 54%, as assessed in the valve area of the aortic root. In blood, IL‐1RA, monocyte subtypes, lipids, and bile acids (BAs) were not affected by MOS. Gut microbiota composition was determined using 16S rRNA gene sequencing and MOS increased the abundance of cecal Bacteroides ovatus. MOS did not affect fecal excretion of cholesterol, but increased fecal BAs as well as butyrate in cecum as determined by gas chromatography mass spectrometry.

3 Conclusion

MOS decreased the onset of atherosclerosis development via lowering of plasma cholesterol levels. These effects were accompanied by increased cecal butyrate and fecal excretion of BAs, presumably mediated via interactions of MOS with the gut microbiota.  相似文献

Dietary fiber is a general terra. It covers a wide variety of substances that belong to the family of carbohydrates that resist hydrolysis by human alimentary enzymes but are fermented by colonie microflora.

The main physiological effects of dietary fiber are primarily on gastric emptying and small intestinal transit time, resulting in an improved glucose tolerance and a decreased digestion of starch; second, on colonie transit time and large bowel functions due to fermentation by ceco‐colonic microbial flora or bulking action.

The so‐called soluble dietary fibers are fermented to a large extent by a wide variety of anaerobic bacteria that result in an increase in bacterial biomass, an increase in fecal mass, a change in intracolonic pH, and production of short chain fatty acids and various gases as metabolic end products. The insoluble fibers are only marginally fermented; they serve almost exclusively as bulking agents that result in shorter transit time and increased fecal mass.

The short chain fatty acids resulting from the colonie fermentation of dietary fiber are largely absorbed via the portal blood and reach both the liver and the peripheral tissues. They induce changes in glucose and fat metabolism leading to post‐prandial hypoglycemia and long‐term hypolipidemia.

Inulin and oligofructose are fructans with a degree of polymerization of 2 to 60 and 2 to 20. respectively. Due to the structural conformation of their osidic bridge ((3 2–1), they both resist the hydrolysis by human alimentary enzymes. Moreover, when reaching the colon, both inulin and oligofructose are almost quantitatively fermented almost exclusively by colonie bifidobacteria and bacteroides. Such an extensive fermentation causes an increase in fecal bacterial biomass, a decrease in ceco‐colonic pH. and produces a large amount of fermentation products among which the short chain fatty acids that exert systemic effects on lipid metabolism.

Thus, both inulin and oligofructose have most of the characteristics of a dietary fiber and the proposal is made to classify them as such. Moreover, they are bifidogenic factors, because, due to still unknown reasons, they are primarily fermented by bifidobacteria.

It is concluded from this review that “nondigestible fructooligosaccharides.” even though they arc not included in the carbohydrate fraction that is quantified as dietary fiber by classic analytical methods, have most of the physiological effects of a dietary fiber. Because it has become necessary for the consumer to be informed clearly and specifically on the nature and the beneficial effects of all fermentable carbohydrates that will “feed” their colon, it is suggested that food labeling should be extended to include such products in the dietary liber fraction.  相似文献

以豌豆纤维粉为原料,以低聚糖得率、清除1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基能力和清除羟自由基能力为考察指标,采用4因素5水平二次旋转组合试验设计,研究酶解工艺参数(料液比、木聚糖酶添加量、纤维素酶添加量、反应温度)对以上考察指标的影响。结果表明最佳酶解工艺参数范围为料液比1∶20.7~1∶24.5(g/mL)、木聚糖酶添加量170.9~176.7?U/g、纤维素酶添加量316.6~320.4?U/g、反应温度52.1~57.6?℃。在优化的条件下,酶解豌豆纤维粉的最优低聚糖得率为13.98%~15.02%,DPPH自由基清除率为30.56%~33.21%,羟自由基清除率为39.36%~42.44%。通过液相色谱分析可得该低聚糖主要组分为阿拉伯糖、葡萄糖、半乳糖、纤维二糖、纤维三糖和纤维四糖。  相似文献

A number of life forms, including seeds, certain nematodes, bacterial and fungal spores, and cysts of certain crustaceans, show an ability to survive desiccation. The present article reviews the literature available on this subject and critically evaluates the evidence for various mechanisms that may be responsible for these phenomena. Specific mechanisms considered include vitrification (glass formation) by sugars and other polyhydroxy compounds that are accumulated by the desiccated structures, specific effects of polyhydroxy compounds on membranes, effect of “compatible solutes” on conformation of key proteins, as well as other biochemical mechanisms.

The article presents potential applications relevant to food technology and to biotechnology and reviews the research required to materialize more effective use of desiccation in food and biopreservation.  相似文献

The content of low molecular weight carbohydrates (LMWC, glucose, fructose, sucrose, raffinose, stachyose and verbascose) was investigated in 29 cultivars of vining peas after blanching and freezing. The peas were harvested at about the same degree of ripeness. There was a positive correlation between the amount of raffinose, stachyose and sorbitol, while a high content of verbascose only could be seen when there was a low content of these sugars. High concentrations of sucrose were generally associated with high concentrations of glucose and fructose. Samples from 1999 had a significantly (p < 0.001) higher content of raffinose, stachyose and sorbitol and lower of verbascose, probably because of the drier and warmer summer that year compared with 1998. Furthermore, medium and late maturing cultivars contained larger amounts of raffinose, stachyose and sorbitol than early cultivars, possibly due to the fact that these cultivars, to a greater extent, were exposed to a higher temperature and more intense sunlight for a longer time. Considerable differences in the content of LMWC could be seen both between the different cultivars and the various trials. Glucose and sucrose were generally similarly affected by cultivar and cultivation conditions, while cultivation conditions had a greater influence on the content of sorbitol and the raffinose family oligosaccharides (RFO; raffinose, stachyose, verbascose) in late‐maturing groups. It is concluded that many factors affect the content of LMWC in peas. However, as the content of RFO is comparatively small in vining peas, any nutritional significance of these sugars in the human colon is probably rather small. ABSTRACT: Microorganisms possess endogenous enzymes, however the stability of these enzymes during storage in soymilk has not been studied. β-glucosidase is an important enzyme that could be used in the bioconversion of the predominant soy isoflavone glucosides to their bioactive aglycone forms. Fifteen probiotic microorganisms including bifidobacterium, Lactobacillus acidophilus , and Lactobacillus casei were screened for β-glucosidase activity using p-nitrophenyl-β-d-glucopyranoside as a substrate. Six strains were selected on the basis of β-glucosidase activity produced during fermentation of soymilk. The stability of the enzyme activity was assessed during incubation for up to 48 h and storage for 8 wk at frozen (-80°C), refrigerated (4°C), room (24.8°C), and incubation (37°C) temperatures. L. casei strains showed the highest β-glucosidase activity after 24 h of incubation followed by L. acidophilus strains, whereas bifidobacterium strains showedleast activity. However, p-glucosidase from Bifidobacterium animalis BB12 showed the best stability during the 48 h fermentation. Lower storage temperatures (-80°C and 4°C) showed significantly higher ( P < 0.05) β-glucosidase activity and better stability than that at higher temperatures (24.8°C and 37°C). The stability of β-glucosidase from these microorganisms should be considered for enzymic biotransformation during storage of isoflavone β-glucosides to bioactive isoflavone aglycone forms with potential health benefits.  相似文献
