产气肠杆菌和阴沟肠杆菌是发酵香肠中产尸胺和腐胺的主要肠杆菌,常同时被检测分离出来。本研究将产气肠杆菌和阴沟肠杆菌按7:1、5:3、4:4、3:5、1:7的比例混合接种培养,及分别纯菌培养,发现在48h培养过程中,产气肠杆菌有较强的产尸胺能力,而阴沟肠杆菌的产腐胺能力则相对较强;两者混合接种培养时,在产尸胺方面存在明显的协同作用,几乎所用混合体系的尸胺产量都高于两个纯菌体系(P<0.01),在产腐胺方面亦存在协同交互作用,但相对较弱,混合体系的腐胺产量一直介于两个纯菌体系之间。  相似文献

从冷藏兔肉中分离出10 株嗜冷菌,用16S rDNA对菌株进行鉴定。B25为沙雷氏菌(Serratia),其他9 株为假单胞菌(Pseudomonas)。采用薄层层析法检测10 株菌的产生生物胺能力,检测到B25可产生多种生物胺,其他9 株没产生生物胺。将B25菌接到加氨基酸前体的假单胞菌肉汤培养基中培养24 h,经高效液相色谱法分析该菌株产生生物胺的种类和含量。最终测得该菌株可产生腐胺、苯乙胺和尸胺,产生这3 种生物胺的能力均比较强,测得腐胺的含量为0.41 mg/mL,尸胺的含量为0.23 mg/mL,苯乙胺的含量为0.05 mg/mL。因此,该菌对冷藏兔肉产品的质量安全威胁较大。  相似文献

The role of post-harvest putrescine and calcium (at 1 mM ), exogenously applied during storage of lemon at 15 °C, in relation to fruit firmness (fruit deformation force) and peel resistance (puncture force) was investigated. Also, the levels of endogenous polyamines (putrescine, spermidine and spermine) and abscisic acid (ABA) and the colour development in mechanically damaged fruits (50 N) were studied. Fruits under putrescine and calcium treatments maintained higher firmness values and more resistance to peel rupture than control ones during storage. Treated lemons showed also less deformation when the compression force (50 N) to induce mechanical damage was applied. Treated and damaged fruits showed a decline in polyamine content while ABA and colour changed in parallel with maturation during storage. In damaged control fruits, an increase of spermine and ABA levels as a consequence of mechanical damage was observed. From the results reported, spermine and ABA could be considered physiological markers of mechanical stress. © 1999 Society of Chemical Industry  相似文献

BACKGROUND: Plum has a very short storage life. The role of pre‐ or post‐harvest applications of putrescine (PUT) and low temperature storage on fruit ripening and quality was investigated in plum fruit (Prunus salicina Lindl. cv. Angelino). RESULTS: Pre‐ or post‐harvest PUT treatments [(0.1, 1.0 or 2.0 mmol L?1) + 0.01% Tween‐20 as a surfactant] delayed and suppressed the climacteric ethylene production and respiration rate irrespective of the method used to apply PUT. PUT‐treated fruit following low temperature storage (0 ± 1 °C; 90 ± 5% RH), at the ripe stage exhibited higher fruit firmness and titratable acidity (TA), while soluble solids content (SSC), levels of ascorbic acid, total carotenoids and total antioxidants were lower than in untreated fruit. Fruit both sprayed with PUT and stored in low temperature for 6 weeks, at the ripe stage showed reduced respiration rate, delayed changes in the SSC:TA ratio and levels of total carotenoids compared to post‐harvest PUT application. CONCLUSION: Pre‐harvest application of 2.0 mmol L?1 PUT 1 week before the anticipated commercial harvest was more effective in delaying plum fruit ripening and can be used to extend the storage (0 ± 1 °C) life of plums for up to 6 weeks with minimum losses in fruit quality. 黄酒发酵液中产生物胺乳酸菌的分离鉴定与评价   总被引:1,自引:1,他引:1  
从黄酒发酵液中分离得到6株乳酸菌,16S rDNA序列分析和生理生化鉴定结果表明:菌株R1、R4、R5、R8、R20为Lactobacillus rossiae,菌株R2为Lactobacillus casei。采用平板检测法对6株乳酸菌产生物胺能力进行了评价,结果显示只有乳酸菌R1具有产生物胺的能力。利用HPLC检测对平板检测结果进行了验证,结果表明乳酸菌R1能产598.61 mg/L的腐胺,其他菌株不具有产生物胺的能力。平板检测与HPLC检测结果相一致,表明平板检测可作为一种有效、操作简单、费用低的定性评价乳酸菌产生物胺能力的手段。  相似文献

Endogenous anticonvulsant mechanisms represent a reliable and currently underdeveloped strategy against recurrent seizures and may recall novel original therapeutics. Here, we investigated whether the intensification of the astroglial Glu-GABA exchange mechanism by application of the GABA precursor putrescine (PUT) may be effective against convulsive and non-convulsive seizures. We explored the potential of PUT to inhibit spontaneous spike-and-wave discharges (SWDs) in WAG/Rij rats, a genetic model of absence epilepsy. Significant shortening of SWDs in response to intraperitoneally applied PUT has been observed, which could be antagonized by blocking GAT-2/3-mediated astrocytic GABA release with the specific inhibitor SNAP-5114. Direct application of exogenous GABA also reduced SWD duration, suggesting that PUT-triggered astroglial GABA release through GAT-2/3 may be a critical step in limiting seizure duration. PUT application also dose-dependently shortened seizure-like events (SLEs) in the low-[Mg2+] in vitro model of temporal lobe epilepsy. SNAP-5114 reversed the antiepileptic effect of PUT in the in vitro model as well, further confirming that PUT reduces seizure duration by triggering glial GABA release. In accordance, we observed that PUT specifically reduces the frequency of excitatory synaptic potentials, suggesting that it specifically acts at excitatory synapses. We also identified that PUT specifically eliminated the tonic depolarization-induced desynchronization of SLEs. Since PUT is an important source of glial GABA and we previously showed significant GABA release, it is suggested that the astroglial Glu-GABA exchange mechanism plays a key role in limiting ictal discharges, potentially opening up novel pathways to control seizure propagation and generalization.  相似文献

Polyamines such as putrescine, spermidine and spermine have been implicated in preventing food allergies in early life, but they have also been reported to be able to bind to immunoglobulin E (IgE) antibodies in vitro (ie they are possibly allergenic). The objective of this study was to determine if polyamines bind in vitro to IgE antibodies from a pooled serum of five peanut‐allergic individuals. Levels of polyamines were also determined by ion‐exchange chromatography. Indirect and inhibition enzyme immunosorbent assays (ELISAs) were used to determine the IgE binding or allergenic properties of polyamines. Results showed that, of the three polyamines, spermidine was predominant in peanuts. In both indirect and inhibition ELISAs, IgE antibodies did not bind to the polyamines. It was concluded that polyamines from peanuts, unlike peanut proteins, are not allergenic or an additional threat to patients who are allergic to peanuts. Scombroid fish poisoning is usually associated with consumption of fish containing high levels of histamine. However, reports indicate that some cases have responded to antihistamine therapy while ingested histamine levels in these cases were low. Potentiation of histamine toxicity by some biogenic amines, and release of endogenous histamine by other compounds such as cis‐urocanic acid (UCA) are some hypotheses that have been put forth to explain this anomaly. Very little is known about the effects of storage conditions on the production of both UCA isomers and biogenic amines in tuna. Thus, the production of trans‐ and cis‐UCA, histamine, putrescine, and cadaverine in tuna during 15 d of storage at 0, 3, and 10 °C and 2 d storage at ambient temperature were monitored. The initial trans‐ and cis‐UCA contents in fresh tuna were 2.90 and 1.47 mg/kg, respectively, whereas the levels of putrescine and cadaverine were less than 2 mg/kg, and histamine was not detected. The highest levels of trans‐ and cis‐UCA were obtained during 15 d storage at 3 °C (23.74 and 21.79 mg/kg, respectively) while the highest concentrations of histamine (2796 mg/kg), putrescine (220.32 mg/kg) and cadaverine (1045.20 mg/kg) were obtained during storage at room temperature, 10 and 10 °C, respectively. Histamine content increased considerably during storage at 10 °C whereas trans‐ and cis‐UCA contents changed slightly. The initial trans‐UCA content decreased during storage at ambient temperature. Thus, unlike histamine, concentrations of trans‐ and cis‐UCA did not result in elevated levels during storage of tuna.  相似文献

Cheeses are among those high-protein-containing foodstuffs in which enzymatic and microbial activities cause the formation of biogenic amines from amino acids decarboxylation. Most of the methods for amine determination in this products involve acid extraction followed by a liquid–liquid purification step to selectively separate amines and amino acids. In this work a simple direct derivatization of the acidic extract with dansyl chloride (DCl) was applied. After an extraction step with diethyl ether, the amines were separated on a Kromasil C18 column using a water–acetonitrile elution gradient. Taking into consideration the recovery and repeatability data we can conclude that, when free amino acid determination is not of concern, the direct DCl derivatization procedure can be applied also for cheese samples with high free amino acid contents.  相似文献

The objective of this work was to compare the contents of polyamines (putrescine, spermidine and spermine), and total soluble phenols and flavonoids in parts of plants grown under either organic or conventional cropping, commonly discarded during food preparation. The contents of free polyamines, total phenols and total soluble flavonoids in peels (zucchini squash, banana, potato, eggplant, orange, lime, mango, passion fruit and radish), leaves (zucchini squash, broccoli, carrot, collard, cassava, radish and grape), stalks (broccoli, collard and spinach) and zucchini seeds were analysed. Most analysed vegetables presented higher contents of polyamines and total phenols under organic cropping, contrary to the results obtained for total flavonoids, possibly because of the cultural practices adopted.  相似文献
