The objective of this study was to compare 3-breed rotational crossbred (CB) cows of the Montbéliarde, Viking Red, and Holstein (HO) breeds with HO cows fed 2 alternative diets for dry matter intake (DMI), fat plus protein production (CFP), body weight (BW), body condition score (BCS), feed efficiency, and residual feed intake (RFI) from 46 to 150 days in milk (DIM) during first lactation. The CB cows (n = 17) and HO cows (n = 19) calved from September 2019 to March 2020. Cows were fed either a traditional total mixed ration diet (TRAD) or a higher fiber, lower starch total mixed ration diet (HFLS). The HFLS had 21% more corn silage, 47% more alfalfa hay, 44% less corn grain, and 43% less corn gluten feed than the TRAD. The 2 diets were analyzed for dry matter content, crude protein, forage digestibility, starch, and net energy for lactation. The BW and BCS were recorded once weekly. Daily milk, fat, and protein production were estimated from twice monthly milk recording with random regression. Measures of efficiency were CFP per kilogram of DMI and DMI per kilogram of BW. The RFI from 46 to 150 DIM was the residual error from regression of DMI on milk energy, metabolic BW, and the energy required for change in BW. Statistical analysis of all variables included the fixed effects of diet, breed group, and the interaction of diet and breed group. The CB cows fed HFLS had less DMI (?12%) and lower DMI/BW (?14%) compared with the HO cows fed TRAD. For CFP, CB and HO cows were not different when fed TRAD or HFLS. Furthermore, the CB cows fed HFLS had higher BW (+50 kg) compared with HO cows fed HFLS. The CB cows fed TRAD had higher BCS than HO cows fed TRAD and HO cows fed HFLS (+0.46 and +0.62, respectively). The HO cows fed TRAD had more DMI (+14%) and lower CFP per kilogram of DMI (?12%) compared with the HO cows fed HFLS. In addition, mean RFI from 46 to 150 DIM was lower and more desirable for CB cows fed HFLS (?120.0 kg) compared with HO cows fed TRAD (85.3 kg). Dairy producers may feed either TRAD or HFLS to CB cows without loss of CFP.  相似文献

Residual feed intake (RFI) is a measurement of the difference between actual and predicted feed intake when adjusted for energy sinks; more efficient cows eat less than predicted (low RFI) and inefficient cows eat more than predicted (high RFI). Data evaluating the relationship between RFI and feeding behaviors (FB) are limited in dairy cattle; therefore, the objective of this study was to determine daily and temporal FB in mid-lactation Holstein cows across a range of RFI values. Mid-lactation Holstein cows (n = 592 multiparous; 304 primiparous) were enrolled in 17 cohorts at 97 ± 26 d in milk (± standard deviation), and all cows within a cohort were fed a common diet using automated feeding bins. Cow RFI was calculated as the difference between predicted and observed dry matter intake (DMI) after accounting for parity, days in milk, milk energy, metabolic body weight and change, and experiment. The associations between RFI and FB at the level of meals and daily totals were evaluated using mixed models with the fixed effect of RFI and the random effects of cow and cohort. Daily temporal FB analyses were conducted using 2-h blocks and analyzed using mixed models with the fixed effects of RFI, time, RFI × time, and cohort, and the random effect of cow (cohort). There was a positive linear association between RFI and DMI in multiparous cows and a positive quadratic relationship in primiparous cows, where the rate of increase in DMI was less at higher RFI. Eating rate, DMI per meal, and size of the largest daily meal were positively associated with RFI. Daily temporal analysis of FB revealed an interaction between RFI and time for eating rate in multiparous and primiparous cows. The eating rate increased with greater RFI at 11 of 12 time points throughout the day, and eating rate differed across RFI between multiple time points. There tended to be an interaction between RFI and time for eating time and bin visits in multiparous cows but not primiparous cows. Overall, there was a time effect for all FB variables, where DMI, eating time and rate, and bin visits were greatest after the initial daily feeding at 1200 h, increased slightly after each milking, and reached a nadir at 0600 h (6 h before feeding). Considering the relationship between RFI and eating rate, additional efforts to determine cost-effective methods of quantifying eating rate in group-housed dairy cows is warranted. Further investigation is also warranted to determine if management strategies to alter FB, especially eating rate, can be effective in increasing feed efficiency in lactating dairy cattle.  相似文献

We determined the effect of prepartum plane of energy intake on liver function and metabolism pre- and postpartum by combining in vivo and in vitro data with mRNA expression data. A subset of multiparous prepartal Holsteins (n = 18) from a previously conducted experiment consumed 1 of 3 amounts of dietary energy intake, relative to their requirements. A diet formulated to allow consumption of ≥150% of net energy requirements during the far-off dry period and the close-up dry period was fed for ad libitum intake (150E) or in restricted amounts so that cows consumed 80% of requirements for energy (80E). A second diet was formulated to include wheat straw (26.1% of dry matter) to limit energy intake to 100% of NRC (2001) requirements for energy when fed ad libitum during the far-off period (100E). In the close-up period, 100E was fed the 150E diet for ad libitum intake. Expression of mRNA for genes related to fatty acid oxidation (PPARA, CPT1A, ACOX1) was greater for 100E cows than 150E cows on d 14 postpartum. These expression patterns were related to in vitro data for conversion of palmitate to CO2, acid-soluble products, and esterified products by liver slices. Abundance of mRNA for PC displayed a sharp peak for all groups on d 1 postpartum, but serum glucose did not reflect this peak. The mRNA expression of SREBF1 was greater for 150E and 100E cows prepartum compared with 80E, and was positively related to rate of palmitate esterification postpartum. Expression of NR1H3 (LXRA) mRNA was greater for 100E cows on d 14 postpartum compared with 150E cows, which corresponded to expression of PPARA. An inflammatory response occurred in the liver around the time of parturition for 150E cows, as expression of IL1B was elevated both pre- and postpartum compared with 100E cows. The spike in IL1B expression for 150E cows on d 14 postpartum corresponded to the peak concentration of total lipids in liver tissue for all groups in this experiment. Overconsumption of energy prepartum was detrimental to the expression of important genes related to PPAR and liver function, especially postpartum. Furthermore, results provide evidence for inflammation related to accumulation of lipids in liver and overnutrition prepartum.  相似文献

Monitoring and detecting individual cows' liveweight (LW) and liveweight change (LWC) are important for estimation of nutritional requirements and health management, and could be useful to measure short-term feed intake, water consumption, defecation, and urination. Walk-over weighing (WOW) systems can facilitate measurements of LW for these purposes, providing automated LW recorded at different times of the day. We conducted a field study to (1) quantify the contribution of feed and water intake, as well as urine and feces excretions, to short-term LWC and (2) determine the feasibility of stationary and WOW scales to detect subtle changes in LW as a result of feed and water intake, urination, and defecation. In this experiment, 10 cows walked through a WOW system and then stood individually on a stationary scale collecting weights at 10 and 3.3 Hz, respectively. Cows were offered 4 kg of feed and 10 kg of water on the stationary scale. For each animal, LW before and after eating and drinking was then calculated using different approaches. Liveweight change was calculated as the difference between the initial and final LW before and after eating and drinking for each statistical measure. The weights of feed intake, water consumption, urination, and defecation were measured and used as predictors of LWC. Urine and feces were collected from individual cows while the cow was on the scale, using a container, and weighed separately. The agreement between LWC measured using either stationary or WOW scales was assessed to determine the sensitivity of the scales to detect subtle changes in LW using the coefficient of determination (R2), Lin's concordance correlation coefficient (CCC), and mean bias. The prediction model showed that most of the regression coefficients were not significantly different from +1.0 for feed and water, or ?1.0 for urine and feces. The R2 and CCC values demonstrated a satisfactory agreement between calculated and stationary LWC and values ranged from 0.60 to 0.92 and 0.71 to 0.94, respectively. A moderate agreement was achieved between calculated and automated LWC with R2 and Lin's CCC values of 0.45 to 0.63 and 0.60 to 0.74, respectively. Therefore, results demonstrated that new algorithms and data processing methods need to be continuously explored and improved to obtain accurate measurements of LW to measure changes in LW, especially from WOW scales.  相似文献

The importance of drinking water for production and animal welfare is widely recognized, but surveys and animal welfare assessment schemes suggest that many dairy calves and dairy cows do not have sufficient access. Limit milk-fed calves drink more water than calves fed milk ad libitum, but ad libitum milk-fed calves also require access to drinking water, as milk does not meet the animal's requirement for water. At hot ambient temperatures and when calves are sick, access to water is especially important and should be provided at all times. Many young calves do not have access to water throughout 24 h, and whether healthy young calves require free access to water at all times, or from which age, is not clear and requires further study. Dairy cow free water intake (FWI) is largely determined by milk yield, and high-yielding dairy cows may drink up 100 L of water per day. Dry matter, crude protein, and salt content of feed, as well as ambient temperature, have considerable effects on dairy cow water intake. Deprivation of water affects meal patterning for the cow, as well as increased subsequent rate of drinking and compensatory water intake. Although dairy cow ad libitum water intake may exceed the water provision necessary to maintain production, offering water for ad libitum intake may be necessary to safe guard animal welfare. Cattle are suction drinkers that prefer to drink from large open water surfaces, and Holstein dairy cows can drink at a rate of up to 24 L/min. Research on the effect of design and placement of water troughs for indoor-housed dairy cows on their drinking behavior and water intake is limited. Access to a water source at pasture increases the time cows spend there, and access to shade reduces water requirements during periods of warm weather. In both indoor and pastured cattle, there is a lack of knowledge about the effect of stocking of water troughs on competition, drinking behavior, and intake in dairy cows. Studies on the effect of available water trough length and placement, and of the number of cows being able to drink from the same trough of a given dimension, are needed to evaluate current recommendations.  相似文献

We aimed to identify relations between satiation and subsequent satiety for six plant-based foods (apple, avocado, banana, carrot, chick pea and macadamia) using a panel of ten healthy participants and a complete crossover randomised design. Food was served as a mid-morning snack ad libitum over 20 min until participants were comfortably full, and subsequent satiety was monitored for up to 180 min. Carrot and macadamia had significantly higher perceived fullness during eating, but also significantly lower perceived satiety per gram than other foods. Food energy factors had no strong relationship with perceived fullness, but were significantly positively correlated with satiety. Mastication number had significant effects on both perceived fullness (negative) and satiety (positive). This pilot study showed that plant food factors affecting perceived fullness during eating and subsequent satiety are different, and suggests how portion control through satiation for different plant-based snacks may influence both energy intake and subsequent satiety.  相似文献

The microplastic particles (MPs) and effects of storage time and direct sunlight on the MPs in bottled mineral waters were investigated by three experiments conditions. The mean MPs concentration was 63.9 ± 38.9 MPs/L. Pellet forms, white/yellow colour and sizes < 100 μm were predominant MPs, accounted for 35.3%, 51% and 60.2% of the total MPs, respectively. Storage of bottled water under darkness and sunlight caused an increase of 1.5% and 2.5% of MPs pollution, respectively. Also, the estimated daily intake (EDI) of MPs was 2.1, 6.4 and 9.6 MPs/kg BW.day for adults, children and infants, respectively. It is concluded that high storage time and direct sunlight may lead to a greater MPs pollution in mineral water and also high human intake through drinking bottled water. Therefore, it is suggested to reduce the expiry date of the bottled mineral water and avoid sunlight contact before consumption.  相似文献

Triple-negative breast cancer (TNBC), one of the most aggressive types of breast cancer, currently lacks a targeted therapy and has a high clinical recurrence rate. The present study reports an engineered magnetic nanodrug based on Fe3O4 vortex nanorods coated with a macrophage membrane loaded with doxorubicin (DOX) and Enhancer of zeste 2 polycomb repressive complex 2 subunit (EZH2) siRNA. This novel nanodrug displays excellent tissue penetration and preferential tumor accumulation. More importantly, it significantly increases tumor suppression compared to chemotherapy, suggesting the synergistic activity of the combination of doxorubicin and EZH2-inhibition. Importantly, owing to tumor-targeted delivery, nanomedicine shows an excellent safety profile after systemic delivery, unlike conventional chemotherapy. In summary, chemotherapy and gene therapy are combined into a novel magnetic nanodrug carrying doxorubicin and EZH2 siRNA, which shows promising clinical application potential in TNBC therapy.  相似文献
