The present study discusses optimization of cellulase production from isolated cellulolytic bacterium. A simulated annealing (SA) algorithm is proposed for optimization of these processes to achieve the desired production goal. The approach was compared to the use of evolutionary algorithms, i.e., genetic algorithms (GAs) and response surface methodology (RSM). Ochrobactrum haematophilum was identified as the isolated bacteria. Carboxymethyl cellulose (CMC) concentration, yeast extract, pH, and incubation temperature were the significant factors screened by Plackett–Burman design and further optimized using a central composite design. The optimum values obtained were CMC concentration = 4.76% (w/v), yeast extract = 2.03% (w/v), pH = 6.3, and temperature = 44.2°C. Carboxy methyl cellulase (CMCase) activity at these values was experimentally determined to be 3.55 ± 0.16 U/ml, which was 2.8 times than the unoptimized system (1.23 U/ml). The growth-associated and non-growth-associated Leudeking–Piret constants, α and β, were respectively determined to be 0.3943 and 0.0105. The Michaelis–Menten constants, Vmax and Km, were determined to be 0.67 µmol/min and 2.42 mg CMC/ml, respectively. The variable-sized SA seems to be the best alternative, outperforming the GAs, showing a fast convergence and low variability among the several runs for optimized production cellulose recovery. The SA models are found to be capable of better predictions of cellulase production. The results of the SA-based RSM model indicate that it is much more robust and accurate in estimating the values of dependent variables when compared with the GA-based RSM models and only RSM models. 相似文献

Supercritical fluid extraction (SFE) was studied as an alternative technology in the pharmaceutical industry for the separation of α-tocopherol from gel and skin of Aloe vera and almond leaves. The influence of operating conditions was investigated on the recovery of supercritical carbon dioxide (SC-CO2) extraction of α-tocopherol from three-year old Aloe vera (Aloe barbadensis Miller) leaf gel. The obtained results were compared with the conventional Soxhlet extraction. Response surface methodology (RSM) was applied to optimize effective variables on the extracted recovery of α-tocopherol. The maximum α-tocopherol recovery of 53.41% from Aloe vera gel was obtained with employing RSM predicted optimal operating conditions of 32 MPa, 45.91 °C, 0.84 ml SC-CO2/min and 140 min for extraction. The α-tocopherol extraction yield for gel and skin of Aloe vera and almond leaves at these optimal operating conditions were obtained 1.53, 16.29 and 2.61 mg/100 g dry sample, respectively. 相似文献

The blade element momentum (BEM) equations, though conceptually simple, can be challenging to solve reliably and efficiently with high precision. These requirements are particularly important for efficient rotor blade optimization that utilizes gradient‐based algorithms. Many solution approaches exist for numerically converging the axial and tangential induction factors. These methods all generally suffer from a lack of robustness in some regions of the rotor blade design space, or require significantly increased complexity to promote convergence. The approach described here allows for the BEM equations to be parameterized by one variable—the local inflow angle. This reduction is mathematically equivalent, but greatly simplifies the solution approach. Namely, it allows for the use of one‐dimensional root‐finding algorithms for which very robust and efficient algorithms exist. This paper also discusses an appropriate arrangement of the equation and corresponding bounds for the one‐dimensional search—intervals that bracket the solution and over which the function is well behaved. Optimization of pre-treatment conditions has been achieved for total sugar release from banana peel powder waste (BPPW) feedstock modelled through a three-level Box-Behnken design (BBD) of the response surface methodology (RSM). A series of various runs were executed at varied acid (H2SO4) concentrations (0.05%–0.15% v/v), incubation periods (1 h–3 h) in water bath at 95 °C and alkali (NaOH) concentrations (0.05%–0.15% v/v) according to the Box-Behnken design (BBD). From RSM the significant values of incubation period, acid concentration and alkali concentration were obtained as 3 h, 0.095% v/v, and 0.05% v/v respectively. The maximum total sugar release was reported as 5243.62 μg/ml which was highly close to the predicted value (5010.07 μg/ml). The model P- value (0.001), R-sq (98.26%), (adj) R-sq (95.14%) and (pred) R-sq (79.56%) obtained through ANOVA justified the results. The mutual impact of alkali and incubation period had the highest effect on total sugar release from dried banana peel powder, followed by mutual impact of acid and incubation period based on ANOVA (Analysis of Variance) results.Under optimized conditions of pre-treatment six different substrate concentrations (1%, 3%, 5%, 7% and 9% w/v) of BPPW was hydrolyzed and used to obtain volumetric bio-hydrogen evolution. The highest cumulative volumetric bio hydrogen gas 43 ml H2/30 ml media was achieved at 5% w/v of pretreated BPPW. The substrate concentration above 5% w/v resulted in lowered fermentation process owing to product and substrate inhibition. 相似文献

Background: The present study is to investigate the neuroprotective effect of ibuprofen by intranasal administration of mucoadhesive microemulsion (MMEI) against inflammation-mediated by dopaminergic neurodegeneration in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) model of Parkinson’s disease (PD).Methods: Ibuprofen-loaded polycarbophil-based MMEI was developed by using response surface methodology (RSM). Ibuprofen with dose of 2.86 mg/kg/day was administered intranasally to male C57BL/6 mice for two consecutive weeks which were pre-treated with four intraperitoneal injections of MPTP (20?mg/kg of body weight) at 2?h intervals. Immunohistochemistry was performed.Results: Optimal MMEI was stable and non-ciliotoxic with 66.29?±?4.15?nm as average globule size and??20.9?±?3.98?mV as zeta potential. PDI value and transmission electron microscopy result showed the narrow globule size distribution of MMEI. The result showed that all three independent variables had a significant effect (p?<?0.05) on the responses. Rota-rod and open-field test findings revealed the significant improvement in motor performance and gross behavioral activity of the mice. The results from in vivo study and immunohistochemistry showed that nasal administration of Ibuprofen significantly reduced the MPTP-mediated dopamine depletion. Furthermore TH neurons count in the substantia nigra and the density of striatal dopaminergic nerve terminals were found to be significant higher for ibuprofen treated groups.Conclusion: Findings of the investigation revealed that Ibuprofen through developed MMEI was shown to protect neurons against MPTP-induced injury in the Substantia nigra pars compacta (SNpc) and striatum and hence, could be a promising approach for brain targeting of Ibuprofen through intranasal route to treat PD. 相似文献

This paper addresses a number of issues for work environment intervention (WEI) researchers in light of the mixed results reported in the literature. If researchers emphasise study quality over intervention quality, reviews that exclude case studies with high quality and multifactorial interventions may be vulnerable to ‘quality criteria selection bias’. Learning from ‘failed’ interventions is inhibited by both publication bias and reporting lengths that limit information on relevant contextual and implementation factors. The authors argue for the need to develop evaluation approaches consistent with the complexity of multifactorial WEIs that: a) are owned by and aimed at the whole organisation; and b) include intervention in early design stages where potential impact is highest. Context variety, complexity and instability in and around organisations suggest that attention might usefully shift from generalisable ‘proof of effectiveness’ to a more nuanced identification of intervention elements and the situations in which they are more likely to work as intended. Statement of Relevance: This paper considers ergonomics interventions from perspectives of what constitutes quality and ‘proof”. It points to limitations of traditional experimental intervention designs and argues that the complexity of organisational change, and the need for multifactorial interventions that reach deep into work processes for greater impact, should be recognised. 相似文献

Alkali‐solution and acid‐isolation method (ASAI) and aqueous two‐phase system separation method (ATSS) have been reported to extract natural colourant carthamin from Carthamus tinctorius. In this study, these two methods were compared based on the optimisation results of extraction conditions by response surface methodology. In ASAI, the maximum extraction yield reached 0.779% at pH value of potassium carbonate solution of 11.16, ratio of potassium carbonate solution to raw material of fifteen and extraction time of 18 min, while 2.652% was achieved with concentration of acetone of 58%, ratio of acetone solution to raw material of twenty‐three and extraction time of 41 min in ATSS. From the point of view of extraction yield, ATSS had more superiority than ASAI. However, more pure carthamin was provided in ASAI according to HPLC assay. The obtained results in our experiments could be utilised for further researches of carthamin. 相似文献
