Expansion of the amino-acid repertoire with synthetic derivatives introduces novel structures and functionalities into proteins. In this study, we improved the antigen binding of antibodies by incorporating halogenated tyrosines at multiple selective sites. Tyrosines in the Fab fragment of an anti-EGF-receptor antibody 059–152 were systematically replaced with 3-bromo- and 3-chlorotyrosines, and simultaneous replacements at four specific sites were found to cause a tenfold increase in the affinity toward the antigen. Structure modeling suggested that this effect was due to enhanced shape complementarity between the antigen and antibody molecules. On the other hand, we showed that chlorination in the constant domain, far from the binding interface, of Rituximab Fab also increased the affinity significantly (up to 17-fold). The gap between two mating surfaces has a direct influence on sealing performance. However, traditional surface metrology rarely characterizes the gap between two mating surfaces. To solve this problem, a novel concept of surfaces complementarity is proposed in this paper. Surfaces complementarity measures how well two rough surfaces fit into each other. To make this concept applicable in engineering practices, a virtual assembly algorithm is developed. The automatic virtual assembly algorithm aligns the mating surfaces by maximizing the overlap ratio of the surface masks. Then, a sum surface which is complementary to the surface gap is constructed to represent the mating states. The top surface of a cylinder block and corresponding cylinder head surface measured by high definition metrology is mated by the virtual assembly algorithm. The differences of functional parameters between the mating surfaces and the sum surface are discussed thoroughly. Due to surfaces complementarity, parameters of the sum surface has a certain deviation from expected combined parameters from two individual surfaces. A case of square surface shows the practical application potential of the virtual assembly algorithm to optimize the sealing performance of the mating surfaces. 相似文献

Reports an error in "Client-therapist complementarity: An analysis of the Gloria films" by Donald J. Kiesler and Chesley S. Goldston (Journal of Counseling Psychology, 1988[Apr], Vol 35[2], 127-133). In this investigation of Shostrom's (1966) psychotherapy demonstration films, we used a checklist inventory measure derived from the 1982 Interpersonal Circle to examine the interpersonal behavior of Gloria, her three therapists, and their respective transaction patterns. Groups of undergraduate raters observed the three films, after which they rated the interpersonal behavior of either Gloria or the therapist. Analyses of Interpersonal Circle axes and segment scores revealed significant differences in the interpersonal behaviors of the three therapists with Gloria and of Gloria with the three therapists. Analyses of transactional patterns showed departures of Gloria's behaviors from the perfect complementary fit predicted from each of the therapist's interpersonal profiles, especially for behaviors categorized on the hostile half of the Interpersonal Circle. Discussion emphasized the complex of dyadic mix and other contextual factors that need to be addressed in future interpersonal studies of psychotherapy. This study proposes a prototype quantitative method for dynamic revenue management of a private toll road, taking into account
the long-term dynamics of transportation demand. This is first formulated as a stochastic singular control problem, in which
the manager can choose the toll level from two discrete values. Each toll change requires nonnegative adjustment costs. Our
analysis then reveals that the optimality condition reduces to standard linear complementarity problems, by using certain
function transformation techniques. This enables us to develop an efficient algorithm for solving the problem, exploiting

To make CGE models realistic, inequality constraints (e.g., import quotas) or non-differentiable functions (e.g., income tax schedules) are sometimes needed. Both situations may be described using complementarity conditions, which state that either an equation is true or itscomplementary variable is at a boundary value. The paper describes a practical way to solve CGE modelsthat contain such conditions. The technique, which is different from complementarity algorithms commonly used elsewhere (e.g., GAMS), has been implemented in the current version of the GEMPACK system, and has been used in a number of applications.This paper explains why the solution methods used in previous versions of GEMPACK do not handle complementarity conditions well, and describes how a two-pass procedure can overcome these difficulties. The key insight is that if we knew in advance which constraints would be binding in the accurate solution, the complementarity conditions could be reformulated in terms of smooth functions only, via a closure change which allows us to ignore the troublesome equations. 相似文献

We study uncertain linear complementarity problems (LCPs), that is, problems in which the LCP vector q or the LCP matrix M may contain uncertain parameters. To this end, we use the concept of Γ-robust optimization applied to the gap function formulation of the LCP. Thus, this work builds upon Krebs and Schmidt (2020). There, we studied Γ-robustified LCPs for ?1- and box-uncertainty sets, whereas we now focus on ellipsoidal uncertainty sets. For uncertainty in q or M, we derive conditions for the tractability of the robust counterparts. For these counterparts, we also give conditions for the existence and uniqueness of their solutions. Finally, a case study for the uncertain traffic equilibrium problem is considered, which illustrates the effects of the values of Γ on the feasibility and quality of the respective robustified solutions. 相似文献
