Rapid determination of chlorophylls in vegetable oils by laser-based fluorometry

The presence of chlorophylls in vegetable oils is considered undesirable due to their adverse effects on oxidative deterioration, bleachability and hydrogenation. The usual methods for determination of chlorophylls in vegetable oils are absorption spectrophotometry, fluorometry and liquid chromatography using fluorescence detectors. This paper outlines a fluorometric method using a laser as the excitation source. The chlorophylls in crude palm oil, rapeseed oil, soya bean oil and olive oil were determined directly by laser-based fluorometry using (a) a helium-neon laser (632-8 nm, 5 mW) as the excitation source and an optical-multichannel analyser (OMA) with an intensified diode array as the detector, and (b) a helium-neon laser and a photomultiplier tube as the detector. The method is especially suitable for the detection of chlorophylls in crude palm oil since no pre-extraction step is required. The levels of chlorophylls measured in vegetable oils using the laser technique was also compared against values obtained by conventional spectrophotometric methods. The simplicity and rapid quantification of laser based fluorometry suggest that this technique can be readily applied to the detection or a wide variety of fluorescing species in vegetable oils.

原油沥青质沉积引起储集层损害的室内评价

防止沥青质沉积造成的储集层损害是渤海稠油油藏开发生产中遇到的技术难题之一。结合渤海SZ36-1油田实际情况，使用岩心流动实验装置，建立沥青质沉积导致储集层岩心渗透率下降的室内评价方法，并应用SARA法对稠油发生沥青质沉积的可能性进行预测。评价实验结果表明，损害程度与沥青质沉积量和储集层岩心的原始渗透率有关。沉积量越大，原始渗透率越低，则损害越严重。表5参12     

An adaptive non‐conforming finite‐element method for Reissner–Mindlin plates

Adaptive algorithms are important tools for efficient finite‐element mesh design. In this paper, an error controlled adaptive mesh‐refining algorithm is proposed for a non‐conforming low‐order finite‐element method for the Reissner–Mindlin plate model. The algorithm is controlled by a reliable and efficient residual‐based a posteriori error estimate, which is robust with respect to the plate's thickness. Numerical evidence for this and the efficiency of the new algorithm is provided in the sense that non‐optimal convergence rates are optimally improved in our numerical experiments. 紧凑式轧机变形规程优化设计

本文从紧凑式轧机的轧制变形特点出发,分析了其变形规程设计的主要特点和方法,讨论了限制其道次变形量的基本因素,给出了优化设计的目标函数和约束条件。指出,类似这种具有复杂约束的轧制变形问题采用混合罚函数法处理更为有效,并成功地采用该方法进行了变形规程的试设定。

Process heat integration of a heavy crude hydrotreatment plant

The present paper focuses on the study of process alternatives for heat integration of a heavy crude oil hydrotreatment plant. Experimental information obtained from a pilot scale, kinetics and reactor modeling tools, and a commercial process simulator were employed to develop mass and energy balances. Four study cases, which employ a combination of quenching and heat exchangers, were identified and evaluated. The results indicated that the case with two quenches with hydrogen and three heat exchangers is the best process heat integration (PHI) option from an energetic point of view.     

An integrated downstream processing strategy for the recovery and partial purification of penicillin acylase from crude media

An integrated process strategy for the recovery of penicillin acylase was developed, based on precipitation of non‐enzymatic proteins directly from Escherichia coli homogenates or crude extracts using Rolquat (quaternary ammonium salt) and adsorption of the enzyme on Amp‐Seph (3.8 µmole ampicillin cm^?3) under pseudo‐affinity conditions. The effect of pH, concentrations of ammonium sulfate and Rolquat, and also concentrations of protein and cell debris on the precipitation of non‐enzymatic proteins from homogenates and crude extracts of penicillin acylase were analysed. The method of addition of Rolquat to homogenates and crude extracts significantly influenced the size of the precipitated particles. Improved results on the specific activity of penicillin acylase were obtained for 22% and 1% (w/v) of ammonium sulfate and Rolquat, respectively, added sequentially to enzyme solutions and at room temperature. Under these experimental conditions, the specific activity of penicillin acylase in homogenates and crude extracts was enhanced 2.5–3.0‐fold. Finally, the integrated process strategy was implemented first by precipitation of non‐enzymatic proteins and recovery of penicillin acylase directly from the enzyme solution treated with Rolquat using an adsorption/filtration system with an overall yield of 86%. This system allows simultaneously the filtration of cell debris and fine precipitated particles, in situ recovery of penicillin acylase by its adsorption on Amp‐Seph, and selective desorption of the enzyme with a specific activity of 11 IU (mg prot)^?1 and a desorption yield of 95%. © 2002 Society of Chemical Industry
