Microstructural evolution and mechanical properties of cryogenic rolled Fe-36Ni steel were investigated. The annealed Fe-36Ni steel was rolled at cryogenic temperature( 123-173 K) with 20%- 90% rolling reduction in thickness.The deformation process was accompanied by twinning at cryogenic temperature,and the mean thickness of deformation twins was about 200 nm with 20% rolling reduction. When the rolling reduction was above 40%,twinning was suppressed due to the stress concentration in the tested steel. Deformation microstructure of Fe-36Ni steel consisted of both twin boundaries and dislocations by cryogenic rolling( CR),while it only contained dislocations after rolling at room temperature( RT). The tensile strength of Fe-36Ni steel was improved to 930 MPa after 90% reduction at cryogenic temperature,while the tensile strength after 90% reduction at RT was only 760 MPa. Goats are an important livestock and goat meat is essential to local people. The intramuscular fat (IMF) content has a great influence on the quality of goat meat. The intramuscular preadipocytes differentiation is closely related to the IMF deposition; however, its potential regulatory mechanisms remain unclear. CircRNAs were revealed to be involved in multiple biological progressions. In this study, we took primary goat intramuscular preadipocyte (GIMPA) as the study model to verify the function and mechanism of chi-circ_0006511, which was abundant and up-regulated in mature adipocytes (GIMA). The results showed that the expression level of chi-circ_0006511 gradually increased in the early stage of GIMPA differentiation, and chi-circ_0006511 was confirmed to promote GIMPA lipid droplets aggregation and up-regulate the adipogenic differentiation determinants, further promoting GIMPA differentiation. Mechanistically, chi-circ_0006511 exerts its function by sponging novel-miR-87, thereby regulating the expression of CD36. The results from this study provided novel significant information to better understand the molecular regulatory mechanism of intramuscular preadipocytes differentiation, thereby providing a new reference for the intramuscular fat adipogenesis in goats. 相似文献

Tumor and stroma coevolve to facilitate tumor growth. Hence, effective tumor therapeutics would not only induce growth suppression of tumor cells but also revert pro-tumor stroma into anti-tumoral type. Previously, we showed that coculturing triple-negative or luminal A breast cancer cells with CD36+ fibroblasts (FBs) in a three-dimensional extracellular matrix induced their growth suppression or phenotypic reversion, respectively. Then, we identified SLIT3, FBLN-1, and PENK as active protein ligands secreted from CD36+ FBs that induced growth suppression of MDA-MB-231 breast cancer cells and determined their minimum effective concentrations. Here, we have expanded our analyses to include additional triple-negative cancer cell lines, BT549 and Hs578T, as well as HCC1937 carrying a BRCA1 mutation. We show that the ectopic addition of each of the three ligands to cancer-associated fibroblasts (CAFs) elevates the expression of CD36, as well as the adipogenic marker FABP4. Lastly, we show that an agonist antibody for one of the PENK receptors induces growth suppression of all cancer cell lines tested but not for non-transformed MCF10A cells. These results clearly suggest that proteins secreted from CD36+ FBs induce not only growth suppression of tumor cells through binding the cognate receptors but also increasing adipogenic markers of CAFs to reprogram tumor stroma. 相似文献

Hardfacing is an effective method for restoring the damaged part. Herein, cobalt-based superalloy Stellite-6 is deposited on A36 substrate using gas metal arc welding process. The mechanical properties, microstructure, and corrosion behavior of Stellite-6 overlays on A36 substrate are examined. Defect-free hard overlays are obtained, and a dilution ratio of 19.9% is achieved with weaving technique. X-ray diffraction analysis reveals the presence of M7C3, M23C6, M6C, and martensite in the Stellite-6 hard overlays. The microstructure of the deposit contains martensite, carbides, and interdendritic precipitates while the mean hardness is 471 HV0.5. Tensile testing with specimens extracted on the longitudinal (LD) and transverse (TD) orientation of the stellite-6 deposit shows a strength above 950 MPa along with the brittle nature of fracture. Grain growth is epitaxial and shows dendritic morphologies. The higher fraction of high-angle grain boundaries improves the tensile strength of the deposit and interface region. The localized corrosion behavior of stellite-6 hard overlays is found to be excellent, and the corrosion analysis exhibits a corrosion rate of 0.10 mils per year (mpy). The outcomes of the study highlight the feasibility of Stellite-6 hard overlays for extending the service life of A36 steel in chloride environments. 相似文献
