Inositol 1,4,5-triphosphate receptor-associated 2 (IRAG2) is a type II membrane protein located at the endoplasmic reticulum. It is a homologue of inositol 1,4,5-triphosphate receptor-associated cGMP kinase substrate 1 (IRAG1), a substrate protein of cGMP-dependent protein kinase I (PKGI), and is among others expressed in platelets. Here, we studied if IRAG2 is also located in platelets and might be a substrate protein of PKGI. IRAG2 was detected in platelets of IRAG2-WT animals but not in those of IRAG2-KO animals. Next, we validated by co-immunoprecipitation studies that IRAG2 is associated with IP3R1-3. No direct stable interaction with PKGIβ or with IRAG1 was observed. Phosphorylation of IRAG2 in murine platelets using a Ser/Thr-specific phospho-antibody was found in vitro and ex vivo upon cGMP stimulation. To gain insight into the function of IRAG2, platelet aggregation studies were performed using thrombin and collagen as agonists for treatment of isolated IRAG2-WT or IRAG2-KO platelets. Interestingly, platelet aggregation was reduced in the absence of IRAG2. Pretreatment of wild type or IRAG2-KO platelets with sodium nitroprusside (SNP) or 8-pCPT-cGMP revealed a further reduction in platelet aggregation in the absence of IRAG2. These results show that IRAG2 is a substrate of PKGI in murine platelets. Furthermore, our results indicate that IRAG2 is involved in the induction of thrombin- or collagen-induced platelet aggregation and that this effect is enhanced by cGMP-dependent phosphorylation of IRAG2. As IRAG1 was previously shown to inhibit platelet aggregation in a cGMP-dependent manner, it can be speculated that IRAG2 exerts an opposing function and might be an IRAG1 counterpart in murine platelets.  相似文献

Many purely physical complex systems, in which there are both stochasticity and local interactions between the components, exhibit record dynamics. The temporal statistics of record dynamics is a Poisson process operating on a logarithmic rather than a linear time scale (i.e. a log-Poisson process). Record dynamics often drive substantial changes in complex systems when new high water marks in partially stochastic processes trigger new events. Social insect colonies are exemplary complex biological systems in which many of the local interactions of the components have been moulded by natural selection for the common good. Here, we combine experimental manipulation of ant colony demography with modelling to test the hypothesis that social interactions are the mechanism underlying the record dynamics. We found that compared with the control, log-Poisson statistics were disrupted in colonies in which the pattern of interactions was modified by the removal of the brood, and disappeared completely in ‘callow’ colonies composed entirely of very young workers from the same age cohort. We conclude that a subtle interplay between the demography of the society and the pattern of the interactions between the ants is crucial for the emergence of record dynamics. This could help identify what makes an ant colony a cohesive society.  相似文献

Luminescent materials with thermally activated delayed fluorescence (TADF) can harvest singlet and triplet excitons to afford high electroluminescence (EL) efficiencies for organic light‐emitting diodes (OLEDs). However, TADF emitters generally have to be dispersed into host matrices to suppress emission quenching and/or exciton annihilation, and most doped OLEDs of TADF emitters encounter a thorny problem of swift efficiency roll‐off as luminance increases. To address this issue, in this study, a new tailor‐made luminogen (dibenzothiophene‐benzoyl‐9,9‐dimethyl‐9,10‐dihydroacridine, DBT‐BZ‐DMAC) with an unsymmetrical structure is synthesized and investigated by crystallography, theoretical calculation, spectroscopies, etc. It shows aggregation‐induced emission, prominent TADF, and interesting mechanoluminescence property. Doped OLEDs of DBT‐BZ‐DMAC show high peak current and external quantum efficiencies of up to 51.7 cd A?1 and 17.9%, respectively, but the efficiency roll‐off is large at high luminance. High‐performance nondoped OLED is also achieved with neat film of DBT‐BZ‐DMAC, providing excellent maxima EL efficiencies of 43.3 cd A?1 and 14.2%, negligible current efficiency roll‐off of 0.46%, and external quantum efficiency roll‐off approaching null from peak values to those at 1000 cd m?2. To the best of the authors' knowledge, this is one of the most efficient nondoped TADF OLEDs with small efficiency roll‐off reported so far.  相似文献

传感器网络为减少冗余数据的传输耗能。降低延迟,需要在路由过程中采用数据聚合技术。文中采用定向传输方式,在消息路由机制基础上提出了一种基于蚁群算法的数据聚合路由算法。该算法主要思想在于将节点能耗、传输距离与聚合收益3方面作为启发因子,通过一组称为“蚂蚁”的人工代理寻找到达汇聚节点的最优路径。该算法利用蚁群算法的正反馈效应来达到数据汇集的目的,不需要网络节点维护全局信息,因此是一种实现数据聚合在能量与时延上折中的分布式路由算法。理论分析和仿真结果说明了新算法的有效性。  相似文献

An efficient data process technology is needed for wireless sensor networks composed of many sensors with constrained communication, computational, and memory resources. Data aggregation is presented as an efficient and significant method to reduce transmitted data and prolong lifetime for wireless sensor networks. Meanwhile, many applications require preserving privacy for secure data aggregation. In this paper, we propose a high energy‐efficient and privacy‐preserving scheme for secure data aggregation. Because of the importance of communication overhead and accuracy, our scheme achieves less communication overhead and higher data accuracy besides providing for privacy preservation. For extensive simulations, we evaluate and conclude the performance of our high energy‐efficient and privacy‐preserving scheme. The conclusion shows that the high energy‐efficient and privacy‐preserving scheme provides better privacy preservation and is more efficient than existing schemes.
