To study the function of GCR1, a gene involved in the expression of glycolytic genes in Saccharomyces cerevisiae, a Kluyveromyces lactis gene that complements the growth defect of a S. cerevisiae Deltagcr1 mutant was isolated. Introduction of this gene into the Deltagcr1 mutant also restored the activities of glycolytic enzymes. DNA sequencing of KlGCR1 predicted an open reading frame of a 767 amino acid protein with an overall identity of 33% and similarity of 48% to Gcr1p from S. cerevisiae. We report the amino acid sequence of 13 open reading frames (ORF > 299 bp) located on a 21·7 kb DNA segment from the left arm of chromosome XIV of Saccharomyces cerevisiae. Five open reading frames had been entirely or partially sequenced previously: WHI3, GCR2, SPX19, SPX18 and a heat shock gene similar to SSB1. The products of 8 other ORFs are new putative proteins among which N1394 is probably a membrane protein. N1346 contains a leucine zipper pattern and the corresponding ORF presents an HAP (global regulator of respiratory genes) upstream activating sequence in the promoting region. N1386 shares homologies with the DNA structure-specific recognition protein family SSRPs and the corresponding ORF is preceded by an MCB (MluI cell cycle box) upstream activating factor. The sequence has been deposited in the EMBL data library under Accession Number X78898.  相似文献

The hypothalamus–pituitary–adrenal (HPA) axis was described as the principal component of the stress response 85 years ago, along with the acute-phase reaction, and the defense response at the tissue level. The orchestration of these processes is essential since systemic inflammation is a double-edged sword; whereas inflammation that is timely and of appropriate magnitude is beneficial, exuberant systemic inflammation incites tissue damage with potentially devastating consequences. Apart from its beneficial cardiovascular and metabolic effects, cortisol exerts a significant immunoregulatory role, a major attribute being that it restrains the excessive inflammatory reaction, thereby preventing unwanted tissue damage. In this review, we will discuss the role of the HPA axis in the normal stress response and in critical illness, especially in critically ill patients with coronavirus disease 2019 (COVID-19). Finally, a chapter will be dedicated to the findings from clinical studies in critical illness and COVID-19 on the expression of the mediator of glucocorticoid actions, the glucocorticoid receptor (GCR).  相似文献

A new essential gene of Saccharomyces cerevisiae was found upstream of GCR1. Its cloning and sequencing predict a 280 amino acid protein (32 577 Da). The predicted protein is fairly hydrophobic, and a search of the database did not identify any homologous proteins. A LEU2 disruption at codon 104 was lethal, but disruption at codon 221 showed a temperature-sensitive conditional growth phenotype. Abnormalities were observed in some glycolytic enzyme levels. The principal objective of this study is to analyze the performance of sun‐trackers devices compared with fixed flat plate systems using data from a widely used irradiation prediction software (PVGIS). We analyze typical parameters as daily and monthly sum of global irradiation (Hd and Hm) or average daily and monthly estimated electricity production (Eh and Em) and also their associated costs and land requirements (usually described by the ground cover ratio, GCR). It was observed that the influence of these two last parameters is quite important to the calculation of the payback time of the whole installation. As a final result, it is concluded that although the dual‐axis tracker allows the maximum energetic performance, a 38% higher than a fixed system on average, if we take into account the GCR value and calculate the surface performance ratio, the most efficient configuration is the horizontal‐axis solar tracker. This work presents an automatic tonal/nontonal preclassification-based Indian language identification (LID) system. Languages are firstly classified into tonal and nontonal categories, and then, individual languages are identified from the languages of the respective categories. This work proposes the use of pitch Chroma and formant features for this task, and also investigates how Mel-frequency Cepstral Coefficients (MFCCs) complement these features. It further explores block processing (BP), pitch synchronous analysis (PSA)- and glottal closure regions (GCRs)-based approaches for feature extraction, using syllables as basic units. Cascade convolutional neural network (CNN)-long short-term memory (LSTM) model using syllable-level features has been developed. National Institute of Technology Silchar language database (NITS-LD) and OGI-Multilingual Telephone Speech Corpus (OGI-MLTS) have been used for experimental validation. The proposed system based on the score combination of Cascade CNN-LSTM models of Chroma (extracted from BP method), first two formants and MFCCs (both extracted from GCR method) reports the highest accuracies. In the preclassification stage, the observed accuracies are 91%, 87.3%, and 85.1% for NITS-LD, for 30 s, 10 s, and 3 s test data respectively. For OGI-MLTS database, the respective accuracies are 86.7%, 83.1%, and 80.6%. That amounts to absolute improvements of 11.6%, 12.3%, and 13.9% for NITS-LD, and 12.5%, 11.9%, and 12.6% for OGI-MLTS database with respect to that of the baseline system. The proposed preclassification-based LID system shows improvements of 7.3%, 6.4%, and 7.4% for NITS-LD and 6.1%, 6.7%, and 7.2% for OGI-MLTS database over the baseline system for the three respective test data conditions.  相似文献

The Saccharomyces sensu stricto complex consists of six closely related species and one natural hybrid. Intra- and inter- species variability in repetitive elements can help elucidate the population structure and evolution of these close relatives. The chromosome positions of several telomeric associated sequences (TASs) and LTR-retrotransposons have been determined, using PFGE, in 112 isolates. Most of the repetitive elements studied are found in multiple copies in each strain, although in some subpopulations these elements are present in low copy number or are absent. Hybridization patterns and copy numbers of the repetitive elements correlate with geographic distribution. These patterns may yield interesting clues as to the origins and evolution of some TASs and retrotransposons, e.g. we can infer that Y' originated on the left end of chromosome XIV. There is strong evidence for horizontal transfer of Ty2 between S. cerevisiae and S. mikatae. Ty1 and Ty5 are either lost easily or frequently horizontally transferred. We have also found some gross chromosomal rearrangements in isolates within species and a few new natural hybrids between species, indicating that these processes occur in the wild and are not limited to conditions of human influence.
