Proton-detected 100 kHz magic-angle-spinning (MAS) solid-state NMR is an emerging analysis method for proteins with only hundreds of microgram quantities, and thus allows structural investigation of eukaryotic membrane proteins. This is the case for the cell-free synthesized hepatitis C virus (HCV) nonstructural membrane protein 4B (NS4B). We demonstrate NS4B sample optimization using fast reconstitution schemes that enable lipid-environment screening directly by NMR. 2D spectra and relaxation properties guide the choice of the best sample preparation to record 2D 1H-detected 1H,15N and 3D 1H,13C,15N correlation experiments with linewidths and sensitivity suitable to initiate sequential assignments. Amino-acid-selectively labeled NS4B can be readily obtained using cell-free synthesis, opening the door to combinatorial labeling approaches which should enable structural studies. 相似文献

This work examines the correlation between the 1H‐NMR T2 relaxation constant and the mechanical properties of aged crosslinked polyolefin cable insulation. T2 experiments on unswollen samples could not differentiate between unaged and highly aged materials; all exhibited 1H T2 constants of approximately 0.5 ms. To accentuate the effects of aging, samples were swollen in various solvents. Unaged samples had T2 values of approximately 15 ms in good solvents. With thermal aging, T2 values decreased as the ultimate tensile elongation decreased. However, the correlation between T2 and elongation differed for samples irradiated with high‐energy radiation and for materials aged above versus below the crystalline melting temperature. © 2003 Wiley Periodicals, Inc. The GC/MS and NMR analysis of the reaction products of naphthalene with 1-bromohexane were reported. The products obtained were suggested to have six isomers of linear hexylnaphthalene and the structure of each isomer was determined by GC/MS and NMR spectra. For the first time, the unique chemical shift data were obtained. The content of each isomer calculated by 1H NMR and 13C NMR spectra was consistent with that by GC/MS spectra, which confirmed the accuracy of NMR data. 相似文献

刘堂宴 ,王绍民 ,傅容珊 ,周明顺 ,李艳华 ,罗曼 .核磁共振谱的岩石孔喉结构分析 .石油地球物理勘探 ,2 0 0 3,38(3) :32 8~ 333岩心核磁共振 T2 谱和压汞分析数据均在一定程度上反映了岩石的孔喉结构 ,理论分析表明 ,这两组数据具有相关性。应用岩心核磁共振 T2 谱研究岩石孔喉结构 ,关键是确定 T2 与 Pc的转换系数。但以前的方法在 T2与 Pc 的转换过程中 ,需要涉及某些岩石特性参数 ,实用中有一定困难。本文直接利用岩心核磁共振 T2 谱和压汞分析数据之间的相关性 ,客观地确定 T2 与 Pc之间的转换系数 ,避免了确定岩石特性参数的困难。应用本文方法 ,对 6块岩心的多种核磁共振分析数据进行了对比分析 ,做出了 NMR (Nuclear Magnetic Resonance,简称NMR) T2 毛管力曲线和孔喉半径分布 ,并将这些结果与压汞分析的结果进行了对比。结果表明 ,岩心 NMR T2谱在实用性和评价精度上均略显优势 ;至少是在饱含油的条件下 ,岩心的 NMR T2 谱可以用于研究孔喉结构分布 ,油气的弛豫特性作为影响背景值存在 ,对于评价结果没有明显的影响 ;在全部 6块岩心中 ,T2 与 Pc 的转换系数位于 2 5 0 0~ 4 0 0 0μs· MPa之间。 相似文献

An efficient, convenient and quantitative method for characterising polyester end-groups is described. We have found that trichloroacetyl isocyanate (TAI) reacts rapidly and quantitatively with both carboxyl [C(O)OH] and hydroxyl (OH) chain ends to form derivatives that can be readily determined by 1H NMR spectroscopy. The TAI capped end-groups give rise to characteristic imidic NH resonances in a normally clear region of the 1H NMR spectrum [δ∼10-11.5 for C(O)-O-C(O)-NH-C(O)CCl3 from C(O)OH, δ∼8-9 for O-C(O)-NH-C(O)CCl3 from OH]. The method has been successfully applied to quantitative determination of the end-groups of a wide variety of oligomeric polyesters. It has also been applied to higher molecular weight polyesters including commercial, bottle grade, poly(ethylene terephthalate) (PET) and PET based copolyesters (e.g. PETG). 相似文献

In the present work, we have studied the feasibility of a method based on polyallyldiglycol-carbonate (PADC) films to investigate the effects of alpha particles on HeLa cervix cancer cells. Thin PADC films with thickness of about 20 μm were prepared from commercially available CR-39 films by chemical etching to fabricate custom-made petri dishes for cell culture, which could accurately record alpha particle hit positions. A special method involving “base tracks” for aligning the images of cell nuclei and alpha particle hits has been proposed, so that alpha particle transversals of cell nuclei can be visually counted. Radiobiological experiments were carried out to induce DNA damages, with the TdT-mediated dUTP Nick-End Labeling (TUNEL) fluorescence method employed to detect DNA strand breaks. The staining results were investigated by flow cytometer. The preliminary results showed that more strand breaks occurred in cells hit by alpha particles with lower energies. Moreover, large TUNEL positive signals were obtained even with small percentages of cells irradiated and TUNEL signals were also obtained from non-targeted cells.
