Triton X-100 pretreatment of LR-white thin sections improves immunofluorescence specificity and intensity

The staining of intracellular antigenic sites in postembedded samples is a challenging problem. Deterioration of antigenicity and limited antibody accessibility to the antigen are commonly encountered on account of processing steps. In this study preservation of the antigen was achieved by fixing the tissues with mild fixatives, performing partial dehydration, and embedding in a low crosslinked hydrophilic acrylic resin, LR-White. Permeabilization of cell membranes with Triton X-100 is well documented but can affect some antigen conformations. We tested the effect of Triton X-100 on the ED1 antigen present in the lysosomal membrane of the macrophage in cell culture. The ED1 antigen in the lysosome was resistant to extraction by Triton X-100. Interestingly pretreating the LR-White sections of macrophage pellets with Triton X-100 improved the staining intensity of ED1. The most intense and clear specific fluorescent staining was observed when sections were pretreated with 0.2% Triton X-100 for 2 min. Longer exposure of sections to 0.2% Triton or 2 min exposure to 2% Triton lead to reduced ED1 labeling. SEM observations indicated that the detergent extracted a component from the cells and not the resin and was determined to be lipid. This novel technique could be applied in many research areas where postembedding fluorescent immunolabeling with higher labeling intensity is desired.

A method of predicting the number of clusters using Rand's statistic

Distributional and asymptotic results on the moment of Rand's C_k statistic were derived by DuBien and Warde [1981. Some distributional results concerning a comparative statistic used in cluster analysis. ASA Proceedings of the Social Statistics Section, 309–313.]. Based on those results, a method to predict the number of clusters is suggested by applying various agglomerative clustering algorithms. In the procedure, the methods using different indexes are examined and compared based on the concept of agreement (or, disagreement) between clusterings generated by different clustering algorithms on the set of data. Our method having practical generality works better than the other methods and assigns statistical meaning to C_k values in determining the number of clusters from the comparison.     

基于IBL算法的CBR系统中索引与检索机制研究

文章介绍了基于示例学习算法IBL的概况,并对其加以改进,提出了一个新的算法IBL－Cluster。它主要由概念描述形成算法和概念描述修改算法构成。在此基础上建立了应用在基于事例的推理系统CBR中的基于IBL算法的索引与检索机制。实验表明新算法IBL－Cluster在存储空间及测试正确率方面均有改善。

低铬X70管线钢组织及其抗CO2

为了明确Cr的质量分数高低对埋地用输油输气管线钢性能的影响,设计了4种Cr的质量分数的X70管线钢,研究了不同Cr的质量分数下管线钢组织及其力学性能,并采用高温高压反应釜进行了实验钢的CO₂腐蚀试验.结果表明：钢中加入0.1%~0.8%的Cr后,其组织均由针状铁素体和准多边形铁素体构成,Cr元素均呈现明显的沿晶界分布状态;随着Cr的质量分数的增加,钢板强度随之升高,晶界中Cr分布密度随之增大,Cr（OH）₃在腐蚀产物膜中的富集量增加,促使钢板的平均腐蚀速率降低;同时由于Cr（OH）₃可以有效阻碍阴离子穿透腐蚀产物膜,因而大大减少了Cl-的催化作用导致的点蚀,使得钢板点蚀速率明显降低.     

Microstructure characteristics and mechanical properties of X80 pipeline steels

The relation between microstructure characteristics and mechanical properties of X80 pipeline steels was investigated using optical microscopy,scanning electron microscopy,etc.It is shown that the structure consists of polygonal ferrite(PF),quasi-polygonal ferrite(QPF),acicular ferrite(AF),and granular bainitic ferrite(GF).With increasing volume fraction of M-A islands(below 3%),the yield strength increases.With increasing content of higher angle grain boundaries(HAGBs),the yield strength,elongation,and DWTT properties at-15 ℃ increase,and the volume fraction of M-A islands reaches its highest point in the steel containing the most volume fraction of GF.
