An appropriate insulin secretion by pancreatic beta-cells is necessary to maintain glucose homeostasis. A rise in plasma glucose leads to increased metabolism and an elevated cytoplasmic ATP/ADP ratio that finally triggers insulin granule exocytosis. In addition to this triggering pathway, one or more amplifying pathways—activated by amino acids or fatty acid—enhance secretion by promoting insulin granule recruitment to, and priming at, the plasma membrane. The aim of this study was to clarify the impact of the mitochondrial respiratory activity on fatty acid-induced insulin secretion that was assessed by an extracellular flux analyzer. Treatment of isolated mouse islets with glucose (20 mM) increased insulin secretion 18-fold and correlated with ATP-synthesizing respiration. Furthermore, oxygen consumption rate (OCR) significantly increased by 62% in response to glucose, whereas the addition of palmitate resulted only in a minor increase of OCR at both 2.8 mM (11%) and 20 mM glucose (21%). The addition of palmitate showed a pronounced increase of coupling efficiency (CE) at 2.8 mM glucose but no further insulin secretion. However, treatment with palmitate at 20 mM glucose increased insulin secretion about 32-fold accompanied by a small increase in CE. Thus, fatty acid induced respiration has a minor impact on insulin secretion. Our data clearly demonstrate that fatty acids in contrast to glucose play a minor role for respiration-mediated insulin secretion. In the presence of high glucose, fatty acids contribute partially to amplifying pathways of insulin secretion by further increasing mitochondrial activity in the islets of Langerhans.  相似文献

In this work we report the evolution of available lysine, lactose and lipid hydroperoxide contents as indicators of food quality and stability of two types of infant formula powder stored at 25° and 37 °C for 12 months. The first type was supplemented with microencapsulated fish oil (MFO), made of little powder particles in a food starch‐coated matrix of caseinate and saccharose to prevent the oxidation of polyunsaturated fatty acid (PUFA). The second type was not supplemented and was used for reference. MFO might not be stable during storage and affect negatively the quality of the formula in two ways: by oxidation of PUFA, which may also easily react with lysine upon oxidation, blocking lysine, or by hydrolysis of saccharose, producing reducing sugars and increasing the Maillard reaction. Slight changes were observed in lysine and lactose evolution. By comparing the two formulae, this study shows that the MFO material did not negatively affect the studied parameters, indicating therefore acceptable stability in the conditions tested.  相似文献

A combination of two‐dimensional (2D) and three‐dimensional (3D) analyses of tissue volume ultrastructure acquired by serial block face scanning electron microscopy can greatly shorten the time required to obtain quantitative information from big data sets that contain many billions of voxels. Thus, to analyse the number of organelles of a specific type, or the total volume enclosed by a population of organelles within a cell, it is possible to estimate the number density or volume fraction of that organelle using a stereological approach to analyse randomly selected 2D block face views through the cells, and to combine such estimates with precise measurement of 3D cell volumes by delineating the plasma membrane in successive block face images. The validity of such an approach can be easily tested since the entire 3D tissue volume is available in the serial block face scanning electron microscopy data set. We have applied this hybrid 3D/2D technique to determine the number of secretory granules in the endocrine α and β cells of mouse pancreatic islets of Langerhans, and have been able to estimate the total insulin content of a β cell.  相似文献

Microencapsulated phase-change materials (MPCM) can be used to develop a structural–functional integrated cement paste having high heat storage efficiency and suitable mechanical strength. However, the incorporation of MPCM has been found to degrade the mechanical properties of cement based composites. Therefore, in this research, the effect of carbon nanotubes (CNTs) on the properties of MPCM cement paste was evaluated. Test results showed that the incorporation of CNTs in MPCM cement paste accelerated the cement hydration reaction. SEM micrograph showed that CNTs were tightly attached to the cement hydration products. At the age of 28 days, the percentage increase in flexural and compressive strength with different dosage of CNTs was found to be up to 41% and 5% respectively. The optimum dosage of CNTs incorporated in MPCM cement paste was found to be 0.5 wt %. From the thermal performance test, it was found that the cement paste panels incorporated with different percentages of MPCM reduced the temperature measured at the center of the room by up to 4.6 °C. Inverse relationship was found between maximum temperature measured at the center of the room and the dosage of MPCM.  相似文献

实验研究了相变微胶囊颗粒(囊芯材料为正十六烷,壳材为尿素-甲醛树脂)和去离子水混合制成的潜热型功能热流体流过等热流细小圆管的流动与传热特性,同时以去离子水作为传热工质在相同条件下进行了对比实验。得到了压降随质量流量的变化规律,实验段出、入口温度以及量纲1出口温度随Reynolds数变化规律,量纲1壁面温度沿轴向的分布规律,平均Nusselt数随Reynolds数的变化关系。结果表明,相变微胶囊颗粒的加入会导致流动压降增大,但随着流量增加,流动压降逐渐与单相液体的接近;出口温度及壁面温度要比相同条件下单相液体的低;含有较小相变微胶囊颗粒浓度的潜热型功能热流体的平均Nusselt数是相同条件下单相液体的2.0~4.0倍。  相似文献

The stability of microencapsulated fish oil was studied during storage at 4 °C for up to 20 wk. Different coating mixtures consisting of gelatin or caseinate in blends with carbohydrates (sucrose, lactose, maltodextrin) were investigated. Oxidative stability of the microencapsulated fish oil was monitored by analysis of volatile compounds using gas chromatography olfactometry (GC‐O) or GC flame ionization (GC‐FID) (SPME‐HS‐GC/O or GC/ FID and HS‐GC/MS), Oxipres test, thiobarbituric acid‐reactive substances (TBARS), and sensory analysis. Coating mixture of caseinate and lactose showed slightly better stability than the sucrose and maltodextrin caseinate mixtures. Combination of fish gelatin and maltodextrin did not show as good oxidative stability as the coating blend of caseinate, lactose, and lecithin. Hexanal, 2‐nonenal and 2,4‐decadienals were selected as quality indicators to monitor the lipid oxidation during storage of the samples. SPME‐GC‐O analysis of these indicators showed that they were representative for the oxidation occurring in the microencapsulated fish oil. SPME‐GC‐FID analysis was sensitive enough to detect oxidative changes during storage. Oxidative stability test, TBARS results, and sensory analysis were in agreement with the SPME, indicating that SPME (polydimethylsiloxane/divinylbenzene [PDMS/ DVB] fiber) can be a useful tool for rapid analysis of lipid oxidation in microencapsulated fish oil.  相似文献

Carbon capture is widely recognised as an essential strategy to meet global goals for climate protection. Although various CO2 capture technologies including absorption, adsorption and membrane exist, they are not yet mature for post-combustion power plants mainly due to high energy penalty. Hence researchers are concentrating on developing non-aqueous solvents like ionic liquids, CO2-binding organic liquids, nanoparticle hybrid materials and microencapsulated sorbents to minimize the energy consumption for carbon capture. This research aims to develop a novel and efficient approach by encapsulating sorbents to capture CO2 in a cold environment. The conventional emulsion technique was selected for the microcapsule formulation by using 2-amino-2-methyl-1-propanol (AMP) as the core sorbent and silicon dioxide as the shell. This paper reports the findings on the formulated microcapsules including key formulation parameters, microstructure, size distribution and thermal cycling stability. Furthermore, the effects of microcapsule quality and absorption temperature on the CO2 loading capacity of the microcapsules were investigated using a self-developed pressure decay method. The preliminary results have shown that the AMP microcapsules are promising to replace conventional sorbents.  相似文献

使用微胶囊化和直接吸附法两种方式将相变材料与水泥结合分别制成了含相变微胶囊水泥基复合建材及含相变材料水泥基复合建材,搭建实验测试系统,对复合建材的调温储能特性进行了测试。微胶囊化相变材料和纯相变材料的加入对水泥基建材的微观结构形貌具有较为明显的影响。热重曲线显示纯水泥、相变微胶囊-水泥、相变材料-水泥试样的失重率依次增加,相变微胶囊-水泥式样的稳定性最好;复合成分所占质量分数相同时,相变材料-水泥试块强度略低于相变微胶囊-水泥试块;调温测试表明,在水泥中添加相变微胶囊颗粒或纯相变材料时均具有一定的调温储能功能,能明显减缓水泥基建材的温度在相变温度附近的波动;尽管直接吸附法制备的相变材料-水泥复合建材具有较好的调温性能,但其可循环性较差,伴有刺激性气味逸出,实用性较差。因此相变微胶囊-水泥复合较适宜作为新型建材推广使用。  相似文献
