Coumarins belong to a group of secondary metabolites well known for their high biological activities including antibacterial and antifungal properties. Recently, an important role of coumarins in plant resistance to pathogens and their release into the rhizosphere upon pathogen infection was discovered. It is also well documented that coumarins play a crucial role in the Arabidopsis thaliana growth under Fe-limited conditions. However, the mechanisms underlying interplay between plant resistance, accumulation of coumarins and Fe status, remain largely unknown. In this work, we investigated the effect of both mentioned factors on the disease severity using the model system of Arabidopsis/Dickeya spp. molecular interactions. We evaluated the disease symptoms in Arabidopsis plants, wild-type Col-0 and its mutants defective in coumarin accumulation, grown in hydroponic cultures with contrasting Fe regimes and in soil mixes. Under all tested conditions, Arabidopsis plants inoculated with Dickeya solani IFB0099 strain developed more severe disease symptoms compared to lines inoculated with Dickeya dadantii 3937. We also showed that the expression of genes encoding plant stress markers were strongly affected by D. solani IFB0099 infection. Interestingly, the response of plants to D. dadantii 3937 infection was genotype-dependent in Fe-deficient hydroponic solution.  相似文献

为计及强散射偶极子间的相互作用,引入参量εh和μh,利用广义多重散射理论及MATLAB计算机软件对在2 GHz~18 GHz频率下的体积占空比为8%的铁纤维随机混合媒质等效磁导率的实验值进行曲线的拟合,导出了耦合系数εh和μh,并在此基础上,对体积占空比分别为30%和20%的铁纤维随机混合媒质等效磁导率进行了计算,计算结果与实验吻合良好.  相似文献

The combined erosion-oxidation of nickel, cobalt, and the oxides of these metals have been studied at 780°C in air to examine two regimes of interactionnamely, a regime of erosion-enhanced oxidation during which an oxide scale of constant thickness covers metal specimens, and a regime of oxidation-affected erosion that is characterized by a composite surface layer of metal, oxide, and erodent. In the case of cobalt, these two regimes have been documented and the transition from one regime to another described. For the range of conditions examined, only the oxidation-affected erosion regime was observed for nickel due to its lower oxidation rate compared to cobalt.  相似文献

[摘 要]目的 克隆并表达大肠杆菌肉碱脱水酶基因。方法 用聚合酶链反应(PCR)从大肠杆菌K12s中扩增大肠杆菌肉碱脱水酶基因caiB,测定其DNA序列,将caiB基因重组到T7启动子控制下的表达载体pET-28a(+)中,构建表达质粒pETCaiB,转化大肠杆菌BL21(DE3),用1mmol/L异两基硫代-β-D半乳糖苷(IPTG)诱导表达。结果 克隆得到的肉碱脱水酶编码基因 caiB长度为 1221bp,与文献报道值相比,DNA序列有26个不同,氨基酸序列只有一个不同,即302位的丙氨酸变为苏氨酸。重组菌经IPTG诱导,可高效表达,SDS-PAGE分析表达蛋白相对分子质量约43000,表达产物含量占菌体总蛋白45%以上。结论 得到了高效表达肉碱脱水酶的重组菌,为制备L-肉碱创造了条件。  相似文献

The foliage and fruit of the tomato plantLycopersicon esculentum contains polyphenol oxidases (PPO) and peroxidases (POD) that are compartmentally separated from orthodihydroxyphenolic substrates in situ. However, when leaf tissue is damaged by insect feeding, the enzyme and phenolic substrates come in contact, resulting in the rapid oxidation of phenolics to orthoquinones. When the tomato fruitwormHeliothis zea or the beet army-wormSpodoptera exigua feed on tomato foliage, a substantial amount of the ingested chlorogenic acid is oxidized to chlorogenoquinone by PPO in the insect gut. Additionally, the digestive enzymes of the fruitworm have the potential to further activate foliar oxidase activity in the gut. Chlorogenoquinone is a highly reactive electrophilic molecule that readily binds cova-lently to nucleophilic groups of amino acids and proteins. In particular, the —SH and —NH2 groups of amino acids are susceptible to binding or alkylation. In experiments with tomato foliage, the relative growth rate of the fruitworm was negatively correlated with PPO activity. As the tomato plant matures, foliar PPO activity may increase nearly 10-fold while the growth rate of the fruitworm is severely depressed. In tomato fruit, the levels of PPO are highest in small immature fruit but are essentially negligible in mature fruit. The growth rate of larvae on fruit was also negatively correlated with PPO activity, with the fastest larval growth rate occurring when larvae fed on mature fruit. The reduction in larval growth is proposed to result from the alkylation of amino acids/protein byo-quinones, and the subsequent reduction in the nutritive quality of foliage. This alkylation reduces the digestibility of dietary protein and the bioavailability of amino acids. We believe that this mechanism of digestibility reduction may be extrapolatable to other plant-insect systems because of the ubiquitous cooccurrence of PPO and phenolic substrates among vascular plant species.  相似文献

目的构建真核表达质粒pcDNA3.1(+)-Vasostatin并检测其在真核细胞内的表达水平。方法将带有信号肽的Vasostatin基因片段克隆至pcDNA3.1(+)真核表达载体上,经酶切鉴定及测序分析证明构建成功后,以脂质体介导法转染293T细胞,通过Westernblot法,检测其在293T细胞内的表达水平。结果所构建的真核表达质粒pcDNA3.1(+)-Va-sostatin转染293T细胞后,在其裂解的上清液中,检测到目的基因的表达。结论已成功构建了pcDNA3.1(+)-Vasostatin表达载体,并在真核细胞中表达了目的蛋白。  相似文献

On the Predominant Electron-Donicity of Polar Solid Surfaces   总被引:2,自引:0,他引:2  
The reasons for the predominant electron-donicity of almost all solid polar surfaces and its implications are discussed in this paper. By contact angle or interfacial tension measurements, the electron-accepting as well as the electron-donating properties of polar liquids can be ascertained, through the interplay between their energies of adhesion and cohesion. For the solid-liquid interface, direct interfacial tension measurements are not possible, but indirectly, solid/liquid interfacial tensions of polar systems can be obtained by contact angle measurement. However, as the energy of cohesion of a solid does not influence the contact angle formed by a liquid drop placed upon its surface, one can only measure the solid surface'ks residual polar property, manifested by the energy of adhesion between solid and liquid. This residual polar property is of necessity the dominant component; in most cases this turns out to be its electron donicity. When, by means of contact angle measurements with polar liquids, both electron-accepting and electron-donating potentials are found on a polar solid, it is most likely still partly covered with a polar liquid: usually water. The amount of residual water of hydration of a polar solid follows from its polar (Lewis acid-base) surface tension component (γAB). The degree of orientation of the residual water of hydration on a polar solid can be expressed by the ratio of the electron-donating to electron-accepting potentials (γ), measured on the hydrated surface.  相似文献

目的分析三个代次CTN-1-V病毒株糖蛋白(GP)基因部分序列,并与代表性街毒株进行比较。方法利用RTPCR反应,从感染了CTN-1-V病毒的小鼠脑内获得GPcDNA的部分片段,并进行序列测定。结果三个代次的CTN-1-V病毒株GPcDNA序列的690个核苷酸与代表性狂犬病毒GP核苷酸相应序列同源性为83.2%~96.8%,氨基酸序列同源性为90.0%~97.4%。CTN1V株三代之间的核苷酸序列几乎相同。结论CTN1V株在传代过程中GP基因结构基本稳定,与中国流行的代表性街毒株的同源性高于aG株和PV株。  相似文献

Shear-induced interactions between high molecular mass polyethylene (HMMPE) melt and a thermotropic liquid crystalline copolyester, HBA/HQ/SA (TLCP) were investigated using large amplitude oscillatory shear and capillary shear. Polarized optical microscopy (POM) observations show that the mono-domain nematic TLCP droplets embedded inside a HMMPE melt may be readily elongated using large amplitude oscillatory shear. The HMMPE melt adjacent to the elongated TLCP filament was observed to crystallize faster than that in the matrix away from the interface. TEM analysis on the 1 wt% TLCP/HMMPE blend quenched after capillary shear shows that there are strong interfacial interactions between the elongated TLCP filament and the HMMPE matrix. Long range PE lamellae orientational order up to 2 μm away from the TLCP filament surface were observed, with all the lamellae surface normal parallel to the TLCP fiber. Additionally, a strong interfacial compatibility between the TLCP filament and the HMMPE matrix with an interfacial thickness of ∼30 nm has also been observed. The enhanced interfacial compatibility is attributed to the -CH2- group interactions due to chain alignment in both components at their interface. These results provide a fundamental insight to other TLCP containing thermoplastics where compatibilities may be present due to segmental interactions.  相似文献

The effect of shear on dewatering behaviour and particle interactions of Na-exchanged smectite and kaolinite clay dispersions has been investigated at pH 7.5, using hydrolysable Ca(II) and Mn(II) ions as coagulants and high molecular weight anionic and non-ionic polyacrylamide (PAM A and PAM N, respectively) and polyethylene oxide (PEO) flocculants. Metal ion addition enhanced the flocculation performance by dramatically reducing the magnitude of the particle zeta potential and, in the case of smectite pulp, suppressing osmotic swelling. Under optimum orthokinetic flocculation conditions of controlled agitation rate and duration, PAM A and PEO-based flocs settled faster than those of PAM N whilst kaolinite pulps produced higher sedimentation rates than smectite pulps. The settling rates are nearly an order of magnitude greater than those observed under standard flocculant-pulp mixing/flocculation methods of inversion and plunging. The difference in the flocculant behaviour is attributed to the more expanded conformation of PAM A and PEO polymer chains in contrast to PAM N, whilst the lower yield stresses, reflecting inter-particle bridging and floc network structure strength that are conducive to faster clarification, were displayed by kaolinite pulps. Following shear, similar consolidation enhancement of ≈ 5-7 wt.% solid for both pulps was achieved at an optimum agitation range of 100-200 rpm. This was accompanied by decreased yield stress in the case of PAM A-based pulps, indicating non-reversible disruption of polymer mediated particle and floc network structure. In contrast, the yield stresses of PAM N and PEO flocculated dispersions indicated similar and stronger particle interactions, respectively, upon consolidation following shear. The findings show clear links between effect of shear, interfacial chemistry and polymer structure on pulp particle interactions and dewaterability.  相似文献
