A real-time PCR-based method for the detection of macadamia nuts (fruits of Macadamia integrifolia or M. tetraphylla or their hybrids) in food products is described. The method consists of DNA isolation by chaotropic solid phase extraction
and subsequent PCR with macadamia-specific primers and a TaqMan fluorescent probe. The primers and the probe were targeted
to the gene encoding for vicilin precursor. The method was positive for M. integrifolia and M. tetraphylla and negative for 16 other plant species used in food industry, including peanuts, walnuts, hazelnuts, almonds, pistachio
nuts, cashew nuts, Brazil nuts, and chestnuts. The DNA-based detection limit of the method was 1.45 pg. Using a series of
model samples with defined macadamia nut contents, a practical detection limit of 0.02% (w/w) macadamia nuts was determined.
Practical applicability of the PCR method was tested by the analysis of 14 confectionery samples. For all of the samples,
results conforming to the labeling were obtained. The presented PCR method is useful for relatively fast, highly selective,
and moderately sensitive detection of macadamia nuts in food samples. 相似文献

BACKGROUND: Medium‐ and long‐chain triacylglyerols (MLCT) enzymatically esterified using Lipozyme RM IM lipase has very low oxidative stability as it does not contain any antioxidants. The aim of this work was to study the ability of various antioxidants to increase the oxidative stability of palm‐ and soybean‐based MLCT blends which assist to bring up the oxidative stability of both MLCT blends. In this study, the effectiveness of rosemary extracts, sage extracts, tert‐butylhydroquinone (TBHQ) and mixtures of tert‐butyl‐4‐hydroxyanisole (BHA) and tert‐butyl‐p‐hydroxytoluene (BHT) in protecting against oxidation of various MLCT blends was investigated. RESULTS: Blending of MLCT oil with either palm olein or soybean oil improved its smoke point values and oxidative stability. TBHQ addition to both palm‐ and soybean‐based MLCT blends increased oxidative stability. Combination of BHA and BHT showed no significant improvement (P > 0.05) in ability to protect blends from oxidation compared to natural antioxidants such as sage or rosemary extracts. CONCLUSION: Blended oils with 500 g kg?1 MLCT and 500 g kg?1 palm olein (MP5) were the most suitable for use at high temperature based on the fatty acid composition of the MLCT blends, which subsequently had an effect on thermal oxidative stability. In general, addition of either natural or synthetic antioxidant assisted in improving the antioxidative strength of both MLCT blends. In a previous study we modified a double lattice model by introducing a new interaction parameter, which improved the mathematical approximation defect, and gave a new expression for the Helmholtz energy of mixing. In the model the universal constants Cβ and Cγ in the primary lattice were determined by comparing them with literature Monte Carlo simulation data, which is the only case for r1 = 1 and r2 = 100 (case I). In this study we introduce new universal constants, Cβ and Cγ, as a function of the chain length of a polymer in a solvent (case II) by comparing them with other literature simulation data for various polymer chain lengths. The proposed model is compared with polymer–solvent systems. In an upper critical solution temperature phase behavior the theoretical results of case II were improved over those of case I.
