The GNAQ T96S Mutation Affects Cell Signaling and Enhances the Oncogenic Properties of Hepatocellular Carcinoma

Hepatocellular carcinoma (HCC), the most common malignant tumor in the liver, grows and metastasizes rapidly. Despite advances in treatment modalities, the five-year survival rate of HCC remains less than 30%. We sought genetic mutations that may affect the oncogenic properties of HCC, using The Cancer Genome Atlas (TCGA) data analysis. We found that the GNAQ T96S mutation (threonine 96 to serine alteration of the Gαq protein) was present in 12 out of 373 HCC patients (3.2%). To examine the effect of the GNAQ T96S mutation on HCC, we transfected the SK-Hep-1 cell line with the wild-type or the mutant GNAQ T96S expression vector. Transfection with the wild-type GNAQ expression vector enhanced anchorage-independent growth, migration, and the MAPK pathways in the SK-Hep-1 cells compared to control vector transfection. Moreover, cell proliferation, anchorage-independent growth, migration, and the MAPK pathways were further enhanced in the SK-Hep-1 cells transfected with the GNAQ T96S expression vector compared to the wild-type GNAQ-transfected cells. In silico structural analysis shows that the substitution of the GNAQ amino acid threonine 96 with a serine may destabilize the interaction between the regulator of G protein signaling (RGS) protein and GNAQ. This may reduce the inhibitory effect of RGS on GNAQ signaling, enhancing the GNAQ signaling pathway. Single nucleotide polymorphism (SNP) genotyping analysis for Korean HCC patients shows that the GNAQ T96S mutation was found in only one of the 456 patients (0.22%). Our data suggest that the GNAQ T96S hotspot mutation may play an oncogenic role in HCC by potentiating the GNAQ signal transduction pathway.

Adenosine Signaling in Mast Cells and Allergic Diseases

Adenosine is a nucleoside involved in the pathogenesis of allergic diseases. Its effects are mediated through its binding to G protein-coupled receptors: A1, A2a, A2b and A3. The receptors differ in the type of G protein they recruit, in the effect on adenylyl cyclase (AC) activity and the downstream signaling pathway triggered. Adenosine can produce both an enhancement and an inhibition of mast cell degranulation, indicating that adenosine effects on these receptors is controversial and remains to be clarified. Depending on the study model, A1, A2b, and A3 receptors have shown anti- or pro-inflammatory activity. However, most studies reported an anti-inflammatory activity of A2a receptor. The precise knowledge of the adenosine mechanism of action may allow to develop more efficient therapies for allergic diseases by using selective agonist and antagonist against specific receptor subtypes.     

Cu（II）掺杂纳米TiO2的制备及其应用研究

本文利用水热法制备了Cu(II)掺杂的纳米TiO2，并用X射线衍射分析仪(XRD)和扫描电镜(SEM)对其进行了表征。将该材料固定于球形分子筛表面作为催化剂，在流动式圆筒反应器中对罗丹明6G(R-6G)和印染污水进行处理，表现出优良的光催化性能。     

Implicit Bayesian Inference Using Option Prices

Abstract. A Bayesian approach to option pricing is presented in which posterior inference about the underlying returns process is conducted implicitly via observed option prices. A range of models allowing for conditional leptokurtosis, skewness and time‐varying volatility in returns are considered, with posterior parameter distributions and model probabilities backed out from the option prices. Models are ranked according to several criteria, including out‐of‐sample predictive and hedging performance. The methodology accommodates heteroscedasticity and autocorrelation in the option pricing errors, as well as regime shifts across contract groups. The method is applied to intraday option price data on the S&P500 stock index for 1995. While the results provide support for models that accommodate leptokurtosis and skewness, no one model dominates when all criteria are considered.     

基于射频技术的智能清洁管理系统研究

针对当前城市清洁工出现无效劳动且管理不便的问题,提出并开发了一套基于无线射频识别技术和超声波传感器技术的垃圾桶智能化管理系统。该系统通过超声波传感器自动检测垃圾桶,实现对每个垃圾桶的监控与管理。并通过RFID技术以及3G通信技术,对存储清洁工信息的电子标签进行识别,及时通知清洁工处理。由于本系统实现了城市环境智能检测、及时处理垃圾、方便工作监督的效果,因而具有较高的应用价值。     

基于乘坐舒适、节能的电梯变频调速系统设计及实现

电梯作为高层建筑中大量使用的机电一体化设备,为了改善乘坐的舒适度、节能,电梯的控制系统中广泛使用交流变压变频调速,作为变压变频控制装置,变频器以其成熟的技术和可靠性被应用于电梯控制系统中,与其他类型交流调速系统相比具有运行效率高、节能效果显著,电梯运行乘坐舒适等优点。本文以日本富士电梯专用变频器5000G11UD为控制器,组成电梯变频调速系统,从理论上分析了实现电梯乘坐舒适、节能的控制关键点,并给出了实际操作方法,具体应用取得了较好的效果。     

4D MRI Flow Coupled to Physics‐Based Fluid Simulation for Blood‐Flow Visualization

Modern MRI measurements deliver volumetric and time‐varying blood‐flow data of unprecedented quality. Visual analysis of these data potentially leads to a better diagnosis and risk assessment of various cardiovascular diseases. Recent advances have improved the speed and quality of the imaging data considerably. Nevertheless, the data remains compromised by noise and a lack of spatiotemporal resolution. Besides imaging data, also numerical simulations are employed. These are based on mathematical models of specific features of physical reality. However, these models require realistic parameters and boundary conditions based on measurements. We propose to use data assimilation to bring measured data and physically‐based simulation together, and to harness the mutual benefits. The accuracy and noise robustness of the coupled approach is validated using an analytic flow field. Furthermore, we present a comparative visualization that conveys the differences between using conventional interpolation and our coupled approach.
