Purpose: In (hemoglobin, Hb) HbEβ‐thalassemia, HbE (β‐26 Glu→Lys) interacts with β‐thalassemia to produce clinical manifestation of varying severity. This is the first proteomic effort to study changes in protein levels of erythrocytes isolated from HbEβ‐thalassemic patients compared to normal. Experimental design: We have used 2‐DE and MALDI‐MS/MS‐based techniques to investigate the differential proteome profiling of membrane and Hb‐depleted fraction of cytosolic proteins of erythrocytes isolated from the peripheral blood samples of HbEβ‐thalassemia patients and normal volunteers. Results: Our study showed that redox regulators such as peroxiredoxin 2, Cu‐Zn superoxide dismutase and thioredoxin and chaperones such as α‐hemoglobin stabilizing protein and HSP‐70 were upregulated in HbEβ‐thalassemia. We have also observed larger amounts of membrane associated globin chains and indications of disruption of spectrin‐based junctional complex in the membrane skeleton of HbEβ‐thalassemic erythrocytes upon detection of low molecular weight fragments of β‐spectrin and decrease in β‐actin and dematin content. Conclusion and clinical relevance: We have observed interesting changes in the proteomic levels of redox regulators and chaperons in the thalassemic hemolysates and have observed strong correlation or association of the extent of such proteomic changes with HbE levels. This could be important in understanding the role of HbE in disease progression and pathophysiology. 相似文献

Chitin synthase synthesizes chitin, which is critical for the arthropod exoskeleton. In this study, we cloned the cDNA sequences of a chitin synthase 1 gene, PcCHS1, in the citrus red mite, Panonychus citri (McGregor), which is one of the most economically important pests of citrus worldwide. The full-length cDNA of PcCHS1 contains an open reading frame of 4605 bp of nucleotides, which encodes a protein of 1535 amino acid residues with a predicted molecular mass of 175.0 kDa. A phylogenetic analysis showed that PcCHS1 was most closely related to CHS1 from Tetranychus urticae. During P. citri development, PcCHS1 was constantly expressed in all stages but highly expressed in the egg stage (114.8-fold higher than in the adult). When larvae were exposed to diflubenzuron (DFB) for 6 h, the mite had a significantly high mortality rate, and the mRNA expression levels of PcCHS1 were significantly enhanced. These results indicate a promising use of DFB to control P. citri, by possibly acting as an inhibitor in chitin synthesis as indicated by the up-regulation of PcCHS1 after exposure to DFB. 相似文献

In power systems, the automatic voltage regulator (AVR) is designed to make the terminal voltage to accurately track the deviation of the reference voltage. To achieve the desired requirements, this paper proposes the modified discrete integral variable structure model following control (MDIVSMFC) approach. The MDIVSMFC approach comprises a reference model part for specifying the design requirements and a MDIVSC part for minimizing the errors between the plant and model. The non-ideal sliding motion is introduced to take the place of the ideal sliding motion in discrete-time systems. Its existence conditions are used to adapt the proposed approach to the discrete systems. The application of MDIVSMFC to an AVR design is developed. Procedures for determining the control function, the coefficients of the sliding surface and the integral control gain are also proposed in detail. Simulation results are compared with the IEEE type-I voltage regulator to demonstrate that the proposed scheme can achieve fast and robust responses. 相似文献

This paper presents a set of simple Ziegler-Nichols-type functions for implementing generalized predictive controllers (GPCs). By using this set of functions, the implementation of GPCs is simplified considerably and the heavy computational requirements needed are avoided. In this report, the behaviour of the first prototype high voltage generator (HVG) that might be used in the time of flight (TOF) system for the AMS-2 experiment is described. The system receives a positive continuous voltage about 100–120 V as input, and it provides a programmable negative continuous voltage from −1600 to −2400 V as output, versus a total load of 50 MΩ. The most important aspect is the absence of a transformer which usually is used in the step-up DC–DC converters. In the TOF system of alpha magnetic spectrometer (AMS)-2 experiment there is a big magnetic field, higher than 2 kG, that does not allow to use a transformer, therefore this prompted us to use the Cockroft–Walton system. The power consumption is about 300 mW and the peak-to-peak high frequency ripple is lower than 0.3% of the output high voltage. We also estimated the reliability of the HVG and we obtained a failure probability lower than 0.5% after three years of continuous functioning. Besides, in this report, much importance was given to the calculation of a simple model of the system to estimate the stability margins. 相似文献
