Resistin is a novel hormone that is secreted by human adipocytes and mononuclear cells and is associated with obesity, insulin resistance and inflammation. Recently, resistin has been postulated to play a role in angiogenesis. Here, we investigated the hypothesis that resistin regulates ovary carcinoma production of vascular endothelial growth factor (VEGF) and the angiogenic processes. We found that in human ovarian epithelial carcinoma cells (HO-8910), resistin (10–150 ng/mL) enhanced both VEGF protein and mRNA expression in a time- and concentration-dependent manner, as well as promoter activity. Furthermore, resistin enhanced DNA-binding activity of Sp1 with VEGF promoter in a PI3K/Akt-dependent manner. PI3K/Akt activated by resistin led to increasing interaction with Sp1, triggering a progressive phosphorylation of Sp1 on Thr453 and Thr739, resulting in the upregulation of VEGF expression. In an in vitro angiogenesis system for endothelial cells (EA.hy926) co-cultured with HO-8910 cells, we observed that the addition of resistin stimulated endothelial cell tube formation, which could be abolished by VEGF neutralizing antibody. Our findings suggest that the PI3K/Akt-Sp1 pathway is involved in resistin-induced VEGF expression in HO-8910 cells and indicates that antiangiogenesis therapy may be beneficial treatment against ovarian epithelial carcinoma, especially in obese patients. 相似文献

Engineering managers involved in product development and manufacturing are responsible for proposing and developing ideas and concepts that will benefit new or existing products and processes. Improved methods are required to identify, prioritize, and integrate the opinions, ideas, and concepts proposed by specialized professionals (SP) or subject matter experts into their existing product development and process improvement. Identifying the optimal SPs and utilizing these for concept generation efforts can lead engineering management working in the new product/technology development space toward an improved idea/concept generation, increased innovation, and ultimately, superior products and processes. Methods for locating, assessing, and ranking SPs vary with every organization, but in general, systematic methods and processes are not considered, which results in the reliance on internal or limited external experts. This study aims to directly impact the engineering managers by bridging the gap between the traditional business management and core engineering management for providing guidance and improving the way in which the practicing engineering managers deal with new product development and management of technology. This experimental study is derived using a real-world engineering problem, which is faced in the field of engineering management, and the results support the implementation of a conceptual SP-focused framework that provides an increased quality of the concept generation outputs, improved preparation for future product development stages, and reduced costs. 相似文献

The soil microorganism Myxobacter Sp. AL-1 regulates in a differential manner the production of five extracellular cellulases during its life cycle. The nucleotide sequence of a cel9-cel48 cluster from the genome of this microorganism was recently obtained. Cel48 was expressed in Escherichia coli to generate a His6-Cel48 protein and the biochemical properties of the pure protein were determined. Cel48 was more efficient in degrading acid-swollen avicel (ASC) than carboxymethylcellulose (CMC). On the other hand, cel9 was expressed in Bacillus subtilis from an IPTG-inducible promoter. Zymogram analysis showed that after IPTG-induction, Cel9 existed in both the cell fraction and the culture medium of B. subtilis and the secreted protein was purified to homogeneity by FPLC-ionic exchange chromatography. The exocellobiohydrolase Cel48 showed a synergism of 1.68 times with the endocellulase Cel9 during ASC degradation using an 8.1-fold excess of Cel48 over Cel9. Western blot analysis revealed that both proteins were synthesized and secreted to the culture medium of Myxobacter Sp. AL-1. These results show that the cel9-cel48 cluster encodes functional endo- and exo-acting cellulases that allows Myobacter Sp. AL-1 to hydrolyse cellulose. 相似文献

小鼠骨髓瘤细胞系(Sp2/0—Ag14)是不分泌型的浆细胞样细胞。我们用Sp2/0—Ag14细胞与免疫小鼠的脾细胞融合,建立了抗本周氏蛋白单克隆抗体的杂交瘤细胞系并采用立体计量方法对亲本及杂种细胞进行了形态定量测量。以便探讨细胞形态结构的变化与分泌抗体的关系。从半薄切片上细胞的结构数据,计算出平均细胞的各种三维空间结构参数。在超薄切片上得到了胞浆内结构成分的三维超微结构参数。结果表明:杂种与亲本细胞相比,杂种细胞的体积明显增大;粗面内质网扩张显著;高尔基体小泡的数量有增多的趋势;线粒体平均体积增大。揭示:杂种细胞的分泌能力较强。 相似文献
